ที่มาและความสำคัญ

โรคอัลไซเมอร์เป็นโรคความเสื่อมระบบประสาทลำดับต้นๆ ซึ่งปัจจุบันยังไม่มียารักษาที่มีประสิทธิภาพ ปลิงทะเลดำถูกนำมาใช้เป็นอาหารและสารบำรุงสุขภาพในประเทศแถบเอเชียอย่างยาวนาน การศึกษาวิจัยนี้จึงศึกษาการออกฤทธิ์ของสารสกัดจากปลิงทะเลดำในการลดความเป็นพิษจากการเกาะกลุ่มของโปรตีนอะไมลอยด์เบต้า ซึ่งเป็นพยาธิสภาพหลักของโรค เพื่อเป็นแนวทางในการป้องกันและรักษาโรคอัลไซเมอร์ต่อไป

Abstract

The black sea cucumber Holothuria leucospilota, an Asian traditional food and medicine, has been explored for health benefits, including neuroprotective effects. Although its anti-Parkinson’s effects have been demonstrated, its potential anti-Alzheimer’s effects remain unexplored. Therefore, this study aimed to investigate the effects and underlying mechanisms of H. leucospilota extracts on amyloid-β (Aβ) proteotoxicity in a Caenorhabditis elegans model of Alzheimer’s disease (AD). All fractions of H. leucospilota extracts were proven to be non-toxic to C. elegans. Among them, the ethyl acetate fraction (HLEA) at 500 μg/mL exhibited the greatest efficacy due to its ability to delay Aβ-induced paralysis in muscular Aβ-expressing C. elegans. Moreover, HLEA at 500 μg/mL rescued the chemotaxis defects in neuronal Aβ-expressing worms. The levels of Aβ oligomers and deposits declined after HLEA treatment of AD worms. Correspondingly, HLEA alleviated Aβ toxicity by lowering ROS levels and extending the lifespan of AD worms. Furthermore, differential RNA sequencing analysis revealed that the lysosome, proteasome, and antioxidant pathways were activated in HLEA-treated AD worms. RT-qPCR assays confirmed that HLEA upregulated ubiquitin-proteasome system, autophago-lysosomal, and antioxidant genes. Moreover, HLEA treatment promoted the nuclear localization of key transcription factors, SKN-1 and DAF-16, along with upregulation of their downstream antioxidant genes, indicating activation of SKN-1 and DAF-16 signaling pathways. Lastly, mass spectrometry analysis revealed that HLEA was predominantly composed of triterpenoid saponins. In summary, HLEA at 500 μg/mL diminished Aβ proteotoxicity by activating the protein clearance mechanism and SKN-1 and DAF-16 stress resistance pathways in the C. elegans AD model.

Abstract

Background and aim

Alzheimer’s disease (AD) is the most common aged-related neurodegenerative disorder that is associated with the toxic amyloid-β (Aβ) aggregation in the brain. While the efficacies of available drugs against AD are still limited, natural products have been shown to possess neuroprotective potential for prevention and therapy of AD. This study aimed to investigate the neuroprotective effects of H. scabra extracts against Aβ aggregation and proteotoxicity in C. elegans model of Alzheimer’s diseases.

Experimental procedure

Whole bodies (WB) and body wall (BW) of H. scabra were extracted and fractionated into ethyl acetate (WBEA, BWEA), butanol (WBBU, BWBU), and ethanol (BWET). Then C. elegans AD models were treated with these fractions and investigated for Aβ aggregation and polymerization, biochemical and behavioral changes, and level of oxidative stress, as well as lifespan extension.

Results and conclusion

C. elegans AD model treated with H. scabra extracts, especially triterpene glycoside-rich ethyl acetate and butanol fractions, exhibited significant reduction of Aβ deposition. These H. scabra extracts also attenuated the paralysis behavior and improved the neurological defects in chemotaxis caused by Aβ aggregation. Immunoblot analysis revealed decreased level of Aβ oligomeric forms and the increased level of Aβ monomers after treatments with H. scabra extracts. In addition, H. scabra extracts reduced reactive oxygen species and increased the mean lifespan of the treated AD worms. In conclusion, this study demonstrated strong evidence of anti-Alzheimer effects by H. scabra extracts, implying that these extracts can potentially be applied as natural preventive and therapeutic agents for AD.

โรคอัลไซเมอร์เป็นโรคหนึ่งในความเสื่อมของระบบประสาท การคิดค้นหาตัวยาหรือปัจจัยที่มีผลต่อการลดพยาธิสภาพหรือลดความรุนแรงของโรคจึงมีส่วนสำคัญต่อผู้ป่วยโรคนี้ โปรตีน Brain-derived neurotrophic factor (BDNF) เป็นโปรตีนที่ควบคุมกระบวนการสร้างเซลล์ระบบประสาทใหม่ แต่การทดสอบผลของโปรตีนนี้ต่อเซลล์ต้นกำเนิดเซลล์ระบบประสาทในโรคอัลไซเมอร์ที่มียีนกลายพันธุ์ที่จำเพาะยังขาดการศึกษาและความเข้าใจอยู่ ดังนั้นงานวิจัยนี้จึงทำการศึกษาฤทธิ์ของโปรตีน BDNF ต่อการเพิ่มจำนวนของเซลล์ต้นกำเนิดระบบประสาท การเพิ่มความยาวของแขนงประสาท โดยใช้เซลล์ต้นกำเนิดระบบประสาทที่พัฒนามาจากเซลล์ต้นกำเนิดแบบชักนำ (induced pluripotent stem cells) ของคนสุขภาพปกติและคนที่เป็นโรคอัลไซเมอร์เป็นเซลล์ต้นแบบ นอกจากนี้ผู้วิจัยได้ทำการศึกษาการส่งทอดสัญญาณเข้าสู่เซลล์ที่จำเพาะต่อ BDNF ต่อการเพิ่มจำนวนของเซลล์ต้นกำเนิดระบบประสาท ซึ่งพบว่าโปรตีน BDNF สามารถเพิ่มจำนวนของเซลล์ต้นกำเนิดระบบประสาทและเซลล์ประสาท ได้ทั้งในเซลล์ที่มาจากคนสุขภาพปกติและคนที่เป็นโรคอัลไซเมอร์ โดยมีการยับยั้งการเพิ่มจำนวนเซลล์เหล่านี้บางส่วนอาจผ่านทางกลไกล JAK/STAT pathway จึงมีความเป็นไปได้ในการนำ BDNF ไปใช้เพื่อลดความรุนแรงของโรคอย่างน้อยในแง่ของการเพิ่มจำนวนเซลล์ต้นกำเนิดระบบประสาทและเซลล์ประสาทในผู้ป่วยโรคอัลไซเมอร์

Abstract

Background: Several pieces of evidence from in vitro studies showed that brain-derived neurotrophic factor (BDNF) promotes proliferation and differentiation of neural stem/progenitor cells (NSCs) into neurons. Moreover, the JAK2 pathway was proposed to be associated with mouse NSC proliferation. BDNF could activate the STAT-3 pathway and induce proliferation in mouse NSCs. However, its effects on proliferation are not fully understood and JAK/STAT pathway was proposed to play a role in this activity.

Methods: In the present study, the effects of BDNF on cell proliferation and neurite outgrowth of Alzheimer’s disease (AD) induced pluripotent stem cells (iPSCs)-derived human neural progenitor cells (hNPCs) were examined. Moreover, a specific signal transduction pathway important in cell proliferation was investigated using a JAK2 inhibitor (AG490) to clarify the role of that pathway.

Results: The proliferative effect of BDNF was remarkably observed as an increase in Ki-67 positive cells. The cell number of hNPCs was significantly increased after BDNF treatment represented by cellular metabolic activity of the cells measured by MTT assay. This noticeable effect was statistically shown at 20 ng/ml of BDNF treatment. BDNF, however, did not promote neurite outgrowth but increased neuronal cell number. It was found that AG490 suppressed hNPCs proliferation. However, this inhibitor partially decreased BDNF-induced hNPCs proliferation. These results demonstrated the potential role of BDNF for the amelioration of AD through the increase of AD-derived hNPCs number.

Abstract
The pathological finding of amyloid-β (Aβ) aggregates is thought to be a leading cause of untreated Alzheimer’s disease (AD). In this study, we isolated 2-butoxytetrahydrofuran (2-BTHF), a small cyclic ether, from Holothuria scabra and demonstrated its therapeutic potential against AD through the attenuation of Aβ aggregation in a transgenic Caenorhabditis elegans model. Our results revealed that amongst the five H. scabra isolated compounds, 2-BTHF was shown to be the most effective in suppressing worm paralysis caused by Aβ toxicity and in expressing strong neuroprotection in CL4176 and CL2355 strains, respectively. An immunoblot analysis showed that CL4176 and CL2006 treated with 2-BTHF showed no effect on the level of Aβ monomers but significantly reduced the toxic oligomeric form and the amount of 1,4-bis(3-carboxy-hydroxy-phenylethenyl)-benzene (X-34)-positive fibril deposits. This concurrently occurred with a reduction of reactive oxygen species (ROS) in the treated CL4176 worms. Mechanistically, heat shock factor 1 (HSF-1) (at residues histidine 63 (HIS63) and glutamine 72 (GLN72)) was shown to be 2-BTHF’s potential target that might contribute to an increased expression of autophagy-related genes required for the breakdown of the Aβ aggregate, thus attenuating its toxicity. In conclusion, 2-BTHF from H. scabra could protect C. elegans from Aβ toxicity by suppressing its aggregation via an HSF-1-regulated autophagic pathway and has been implicated as a potential drug for AD.

โรคความจำเสื่อม Alzheimer เป็นโรคความเสื่อมของระบบประสาทซึ่งเกิดจากการเกาะกลุ่มกันของโปรตีน amyloid-beta และส่งผลเสียต่อการทำงานของเซลล์ประสาทในระยะยาว การศึกษาวิจัยนี้แสดงให้เห็นฤทธิ์ของสาร frondoside A จากปลิงทะเล Cucumaria frondosa ในการลดการเกาะกลุ่มของโปรตีน amyloid-beta ในหนอน Caenorhabditis elegans ที่ใช้เป็นสัตว์ทดลองต้นแบบของโรค Alzheimer โดยสาร frondoside A มีผลโดยตรงในการลดระดับ amyloid-beta oligomers ที่เป็นพิษต่อเซลล์ ขณะที่ปริมาณ monomers ไม่มีการเปลี่ยนแปลง และยังส่งผลฟื้นฟูการทำงานของเซลล์ประสาทให้ดีขึ้น รวมถึงการลดปริมาณอนุมูลอิสระที่สะสมด้วย การศึกษานี้จึงแสดงให้เห็นถึงศักยภาพของสาร frondoside A ในการป้องกันโรค Alzheimer และนำไปสู่การวิจัยพัฒนาต่อยอดในอนาคตต่อไป

Abstract
Oligomeric assembly of Amyloid-β (Aβ) is the main toxic species that contribute to early cognitive impairment in Alzheimer’s patients. Therefore, drugs that reduce the formation of Aβ oligomers could halt the disease progression. In this study, by using transgenic Caenorhabditis elegans model of Alzheimer’s disease, we investigated the effects of frondoside A, a well-known sea cucumber Cucumaria frondosa saponin with anti-cancer activity, on Aβ aggregation and proteotoxicity. The results showed that frondoside A at a low concentration of 1 µM significantly delayed the worm paralysis caused by Aβ aggregation as compared with control group. In addition, the number of Aβ plaque deposits in transgenic worm tissues was significantly decreased. Frondoside A was more effective in these activities than ginsenoside-Rg3, a comparable ginseng saponin. Immunoblot analysis revealed that the level of small oligomers as well as various high molecular weights of Aβ species in the transgenic C. elegans were significantly reduced upon treatment with frondoside A, whereas the level of Aβ monomers was not altered. This suggested that frondoside A may primarily reduce the level of small oligomeric forms, the most toxic species of Aβ. Frondoside A also protected the worms from oxidative stress and rescued chemotaxis dysfunction in a transgenic strain whose neurons express Aβ. Taken together, these data suggested that low dose of frondoside A could protect against Aβ-induced toxicity by primarily suppressing the formation of Aβ oligomers. Thus, the molecular mechanism of how frondoside A exerts its anti-Aβ aggregation should be studied and elucidated in the future.
