Abstract

Sulfated galactans (SG) isolated from Gracilaria fisheri is partially degraded (DSG), and subsequentially supplemented with octanoyl (DSGO) and sulfate (DSGS) groups. The molecular weights of DSG, DSGO, and DSGS are 7.87, 152.79, and 97.07 kDa, respectively. The modification is confirmed using FTIR and NMR, while in vitro wound healing activity is assessed using scratched wound fibroblasts. The results reveal that DSGO exhibits highest percentage of wound closure in scratched fibroblast L929 cells. Furthermore, DSGO is able to promote proliferation and accelerate migration of scratched fibroblasts, which correspond to the regulation of proteins and mRNA (Ki67, p-FAK, vimentin, and E-cadherin) determined by Western blotting and qPCR analysis. The superior wound healing activity of DSGO is also confirmed in excision wound of rats. The results demonstrate that DSGO significantly enhances the percentage of wound closure, re-epithelialization, and collagen arrangement, increases α-smoth muscle actin (α-SMA) and vimentin expression, and decreases that of tumor necrosis factor-α (TNF-α) at the wound site. The results suggest that degraded SG supplemented with medium-chain fatty acids of octanoyl group may pass through the membrane, subsequently activating the mediators associated with proliferation and migration of fibroblasts, which can potentially lead to the promotion of wound healing activity.

Fig. 1. A schematic diagram showing the proposed protection of S-SG from sodium oxalate induced HK-2 cell damage. S-SG, which is due to high sulfate groups, possibly interacts with oxalate crystal surface, reduces cell–oxalate crystal interaction, and subsequently decreases HK-2 cell damage. Furthermore, S-SG also decreases expression of caspase-3 and increases expression of BCL-2, resulting in reduced cell damage induced by sodium oxalate crystals (created with BioReder.com).

Abstract

Halymenia durvillei (HD), a marine red alga, is believed to have potentials for pharmacological, nutritional and cosmetic applications. However, such potentials are acceptable only when their extracts are devoid of any adverse effects on human health. No previous research has been conducted the toxicity and anti-oxidation capacity of HD. Thus, the aim of this work was to investigate toxicity and anti-oxidation activities of HD extracts. In this study, the toxicity and anti-oxidation capacity of 5 fractions of HD solvent extracts, i.e., ethanol (HDET), hexane (HDHE), ethyl acetate (HDEA), butanol (HDBU), and aqueous (HDAQ) were evaluated. The cytotoxicity was evaluated by MTT and LDH assays on 4 cell types, i.e., fibroblast, macrophage, hepatocyte and keratinocyte. The genotoxicity was evaluated by comet assay and micronucleus test using TK6 lymphoblastoid cell line. The anti-oxidation capacity was investigated by DPPH and ABTS assays. The toxicity studies showed that HDET, HDBU, HDAQ had very low to no toxicity as indicated by cytotoxicity and genotoxicity tests while HDEA, HDHE have some toxicity at high concentrations. HDAQ showed low antioxidant activity while HDET, HDEA, HDHE and HDBU possess relatively high antioxidant activity. Overall, our results indicated that HDET and HDAQ could be consumed as they are not toxic and HDHE, HDEA, and HDBU could be safely consumed at doses lower than 100 μg/mL. Further investigation using in vivo assays are needed to ensure the safety of HD extracts for animal and human consumptions.
