ที่มาและความสำคัญ

แม้การสร้างเอ็นสะบ้า–ต้นขาด้านใน (MPFL; Medial patellofemoral ligament) ร่วมกับเอ็นสะบ้า–หน้าแข้งด้านใน (MPTL; Medial patellotibial ligament) จะมีศักยภาพในการเพิ่มความมั่นคงของกระดูกสะบ้าในช่วงเข่างอมาก แต่หลักฐานทางชีวกลศาสตร์ที่เปรียบเทียบโดยตรงกับการสร้าง MPFL เพียงอย่างเดียวยังมีจำกัด ดังนั้น การศึกษานี้จึงมุ่งเปรียบเทียบผลของการสร้าง MPFL เพียงอย่างเดียวกับการสร้าง MPFL ร่วมกับ MPTL ต่อการเคลื่อนของกระดูกสะบ้าและความดันบริเวณข้อสะบ้า–ต้นขา (patellofemoral joint) ในองศาการงอเข่าที่แตกต่างกัน เพื่อประเมินว่าการเพิ่ม MPTL reconstruction สามารถเพิ่มความมั่นคงของกระดูกสะบ้าได้โดยไม่เพิ่มความดันในข้ออย่างมีนัยสำคัญหรือไม่

Abstract

Introduction/objectives

The medial patellotibial ligament (MPTL) is a secondary patellar stabilizer whose contribution increases with progressive knee flexion. Combined medial patellofemoral ligament (MPFL) and MPTL reconstruction is increasingly used clinically, but biomechanical data comparing it with isolated MPFL reconstruction for patellar translation and contact pressure are lacking. This cadaveric study compared these two techniques and simulated patellar instability, evaluating patellar translation and mean/peak patellofemoral contact pressure at 0°, 30°, 45°, 60°, and 90° of knee flexion. We hypothesized that combined reconstruction would reduce lateral patellar translation to a greater degree than isolated reconstruction without increasing patellofemoral contact pressure.

Methods

Eighteen fresh-frozen cadaveric knees underwent transection of the medial stabilizers to simulate patellar instability, then were randomized to isolated MPFL reconstruction or combined MPFL–MPTL reconstruction using peroneus longus tendon grafts. Three-dimensional patellar kinematics were captured with a six-camera motion capture system, and patellofemoral contact pressures were recorded using Tekscan sensors with the knee held statically at each angle. Patellar translation and contact pressure were compared between techniques.

Results

All specimens showed successful instability induction after medial stabilizer transection. Compared with the injured state, isolated reconstruction reduced lateral translation by 0.03 mm and combined reconstruction by 0.08–0.09 mm at all angles (each P-value < 0.05). The combined group showed 0.08–0.09 mm greater medial translation than isolated reconstruction at all angles (P-value ≤ 0.017), with the largest difference at 60° and 90°. No significant between-group difference was seen in proximal–distal translation. Both techniques restored a physiologic pattern of mean and peak contact pressure with no significant between-group difference in mean pressure at any angle. Peak pressure increased from baseline at 90° for isolated reconstruction (P-value = 0.05) and at 0°, 60° and 90° for combined reconstruction (P-value ≤ 0.032). Peak pressure was significantly higher after combined than isolated reconstruction at full extension (P-value = 0.021) with no other between-technique differences.

Conclusion

Combined MPFL–MPTL reconstruction reduces lateral patellar translation more than isolated MPFL reconstruction, particularly in deeper knee flexion, without increasing patellofemoral contact pressure. These findings support MPTL reconstruction as a biomechanically beneficial adjunct for managing patellar instability.

Abstract

Objective

This study aimed to develop a reproducible immunohistochemistry method for formalin-fixed, paraffin-embedded (FFPE) muscle sections and to assess its diagnostic utility for idiopathic inflammatory myopathies (IIM) in settings where snap-frozen muscle biopsy is unavailable.

Methods

Twenty cases were analysed: dermatomyositis (n = 5), inclusion body myositis (n = 2), immune-mediated necrotizing myopathy (n = 5), overlap myositis (n = 1), unspecified myopathy (n = 3), mitochondrial myopathy (n = 1), LGMD R2 dysferlin-related muscular dystrophy (n = 1), facioscapulohumeral muscular dystrophy (n = 1), and neurogenic muscle change (n = 1). Immunohistochemistry was performed on 3-μm FFPE muscle sections using heat-induced antigen retrieval with citrate/EDTA buffer and overnight incubation at 4°C with antibodies against MHC-I, MHC-II, MAC, MxA, and p62. Frozen sections were analysed in parallel for comparison.

Results

Comparable staining intensities and patterns for MHC-II, MxA, and p62 were achieved in FFPE muscle sections. Although discrepancies between frozen and FFPE sections occurred in eight cases, each involving a single marker, no statistically significant differences in immunohistochemical results were observed for MHC-II (P = 0.4795), MxA (P = 0.4795), and p62 (P = 0.6171). Concordance between methods was high, reaching 90% for MHC-II and MxA and 80% for p62. These isolated discrepancies may reflect variability in antigen expression associated with the segmental and patchy involvement characteristic of IIM. However, MHC-I and MAC staining were unsuccessful in FFPE sections.

Conclusions

Successful immunolabelling of MHC-II, MxA, and p62 was achieved in FFPE muscle sections. These findings indicate that several diagnostically informative markers for IIM can be reliably assessed in FFPE muscle, providing a practical diagnostic option in resource-constrained settings.

ความหลวมคงเหลือ (residual laxity) หลังการผ่าตัดสร้างเอ็นไขว้หลังข้อเข่า (posterior cruciate ligament; PCL) แบบเส้นเอ็นเดี่ยว (single-bundle reconstruction) ยังคงเป็นปัญหาทางคลินิกที่อาจส่งผลกระทบต่อผลลัพธ์ด้านการใช้งานของข้อเข่าหลังผ่าตัดได้ การเสริมด้วยแถบเทปเย็บ (suture tape augmentation) ได้ถูกนำมาใช้เป็นเทคนิค internal brace เพื่อช่วยปกป้องเส้นเอ็นปลูกถ่าย (graft) และลดความหลวมของข้อเข่าในระยะแรกหลังการผ่าตัด

Abstract

Background

Residual laxity after single-bundle posterior cruciate ligament (PCL) reconstruction remains a clinical concern that may compromise functional outcomes. Suture tape augmentation has been introduced as an internal bracing technique to enhance graft protection and reduce early postoperative laxity. This study aimed to determine whether suture tape augmentation reduces posterior tibial translation (PTT) and graft elongation under cyclic loading in single-bundle PCL reconstruction.

Methods

Eight fresh-frozen human cadavers (16 knees) underwent bilateral single-bundle PCL reconstruction. One knee in each pair received suture tape augmentation, while the contralateral knee served as control. Posterior tibial translation was measured at 90° of flexion under four conditions: intact, after partial PCL transection, after reconstruction, and after 1000 loading cycles at 1 Hz. Graft elongation, diameter, and tibial tunnel size were recorded. Paired t-tests compared groups.

Results

No significant differences in PTT were found between groups in the intact, cut, or reconstructed states. After cyclic loading, the suture tape group showed lower PTT than controls (4.60 ± 2.24 mm vs 5.49 ± 1.52 mm, p = 0.06). The increase in PTT from reconstruction to post-cyclic loading was smaller with suture tape (1.09 ± 0.65 mm vs 1.57 ± 0.65 mm, p < 0.05). Graft elongation was similar, while graft diameter and tibial tunnel size were slightly larger with suture tape (p < 0.05).

Conclusion

Suture tape augmentation limited the increase in posterior tibial translation under cyclic loading, indicating improved dynamic stability and potential graft protection in single-bundle PCL reconstruction.

มะเร็งท่อน้ำดี (CCA) รักษายากเพราะมีสภาพแวดล้อมของก้อนมะเร็งซับซ้อนและยังขาดเป้าหมายการรักษาที่ได้ผล งานวิจัยนี้ทดสอบแนวคิด CAR T cell ที่มุ่งเป้าไปที่ mesothelin (MSLN) ซึ่งพบแสดงออกสูงใน CCA โดยพัฒนา CAR T จากแอนติบอดี “กระต่ายแบบ humanized” ที่จับ MSLN ต่างตำแหน่งกัน ผลพบว่า CAR T ที่สร้างจากแอนติบอดี hYP218 (ออกแบบโครงสร้าง/การจัดวางเฉพาะ) สามารถกำจัดก้อนมะเร็งในหนูได้ดี มีความคงอยู่ของเซลล์สูง และมีผลข้างเคียงน้อย ชี้ว่า CAR T targeting MSLN อาจเป็นทางเลือกการรักษาใหม่ที่มีศักยภาพสำหรับ CCA

ที่มาและความสำคัญ

มะเร็งท่อน้ำดี (CCA) เป็นมะเร็งที่รักษายาก เพราะสภาพแวดล้อมของก้อนมะเร็งซับซ้อน มีพังผืดมากทำให้ T cell เข้าไปในก้อนได้ยาก และยังขาด “เป้าหมาย” ที่ยืนยันใช้รักษาได้จริงในคลินิก แม้ CAR T cell จะได้ผลดีในมะเร็งเม็ดเลือด แต่ยังมีข้อจำกัดใน CCA งานวิจัยนี้เลือก Mesothelin (MSLN) เป็นเป้าหมายของการพัฒนา CAR T cell เนื่องจากมีคุณสมบัติสอดคล้องกับหลักเกณฑ์ของ antigen สำหรับการรักษาแบบจำเพาะในมะเร็งท่อน้ำดี (CCA) โดยเฉพาะอย่างยิ่ง MSLN เป็นโปรตีนบนผิวเซลล์ซึ่งเอื้อต่อการจดจำและการทำงานของ CAR T cell และพบการแสดงออกสูงในเนื้อเยื่อ CCA ของผู้ป่วยไทยในสัดส่วนมาก (79%) สะท้อนความเป็นเป้าหมายที่ครอบคลุมผู้ป่วยจำนวนมาก นอกจากนี้ MSLN ถูกจัดอยู่ในกลุ่ม tumor-associated antigen ที่เกี่ยวข้องกับชีววิทยาการดำรงอยู่และความก้าวร้าวของก้อนมะเร็ง จึงมีศักยภาพในการให้ผลเชิงการรักษามากกว่าเป็นเพียงตัวบ่งชี้การเกิดโรค อีกทั้ง CCA ยังขาดเป้าหมายที่ผ่านการยืนยันทางคลินิกอย่างชัดเจน ทำให้การมุ่งพัฒนา CAR T ที่จำเพาะต่อ MSLN เป็นแนวทางเชิงกลยุทธ์เป้าหมายสำหรับ CAR T cell therapy ใน CCA.

Abstract

Background: Chimeric antigen receptor (CAR) T cell therapy has shown promise in treating hematological malignancies. However, the complex tumor microenvironment of cholangiocarcinoma (CCA) poses significant challenges, particularly due to the lack of clinically validated targets and the presence of fibrosis, which hinders T cell infiltration into tumor sites. Mesothelin (MSLN) is highly expressed in CCA. In this study, we aimed to develop CAR T therapy based on a panel of humanized rabbit monoclonal antibodies targeting various epitopes of MSLN, ranging from the N- to the C-terminus, for CCA.

Methods: MSLN expression was assessed in CCA tissue samples obtained from Thai patients. CAR T cells were generated using various single-chain variable fragment (scFv) constructs, engineered in either VH-linker-VL or VL-linker-VH orientation, targeting non-overlapping epitopes of membrane-bound MSLN: hYP218 (proximal region), hYP223 and hYP3 (middle region), and hYP158 (distal region). The cytotoxicity of MSLN-specific CAR T cells was evaluated in 3 preclinical CCA mouse models: Mz-ChA-1, KMCH, and KMBC.

Results: MSLN was strongly expressed in 79% of the CCA specimens. Among the CAR constructs, hYP218-based CAR T cells-with the VL-linker-VH orientation and CD28-derived hinge and transmembrane domains (CD28HTM)-completely eradicated CCA tumors in all 3 CCA xenograft mouse models (Mz-ChA-1, KMCH, and KMBC). Furthermore, hYP218 VLVH CD28HTM CAR T cells showed strong persistence in mice, with low PD-1 expression (a marker of T cell exhaustion) and minimal adverse effects.

Conclusions: Our findings suggest that MSLN is a promising target for CAR T cell therapy in CCA. CAR T cells engineered with hYP218 VLVH CD28HTM, which targets the membrane-proximal epitope of MSLN, may represent a novel therapeutic strategy for the clinical treatment of CCA.

ที่มาและความสำคัญ

รายงานฉบับนี้นำเสนอกรณีศึกษาที่หายากและซับซ้อนของทารกแรกเกิดไทยคนหนึ่ง ซึ่งมีโรคผิดปกติแต่กำเนิดสองตำแหน่งในระบบทางเดินอาหารซึ่งไม่เคยมีรายงานพบร่วมกันมาก่อน ได้แก่ โรค Retroesophageal right subclavian artery และ Congenital colonic stenosis ซึ่งมีอาการคล้ายโรคที่พบได้บ่อย เช่น โรคแพ้โปรตีนนมวัวหรือภาวะท้องอืดจากการติดเชื้อ ส่งผลให้เกิดการวินิจฉัยผิดพลาด

กรณีนี้ได้เน้นย้ำถึงความสำคัญของการตระหนักถึงโรคแต่กำเนิดที่พบได้น้อยแต่มีผลกระทบรุนแรง ซึ่งหากตรวจพบได้ตั้งแต่เนิ่น ๆ จะช่วยหลีกเลี่ยงการวินิจฉัยผิดและเพิ่มโอกาสในการรักษาทารกในกรณีคล้ายกันในอนาคต

Abstract

Background

Double alimentary tract obstruction due to congenital anomalies is a rare clinical occurrence, with limited cases published in medical literature. This article presents a unique case of coexisting retroesophageal right subclavian artery (RRSA) and congenital colonic stenosis (CCS), conditions that have not been previously documented together in pediatric population.

Case presentation

A Thai male newborn was born by cesarean section at gestational age of 41 weeks. One week before birth, intrauterine asphyxia and idiopathic bilateral intracerebral hemorrhage were diagnosed by prenatal ultrasonography. Despite postnatal interventions including a ventriculoperitoneal shunt and subsequent external ventricular drain, the intracerebral hemorrhage recurred and progressed. Concurrently, the patient experienced multiple episodes of post-feeding vomiting, intermittent abdominal distension, and regular defecation without constipation. Sepsis secondary to an infected shunt occurred, accompanied by marked abdominal distension. The physician clinically suspected non-IgE-mediated cow’s milk protein allergy and ileus associated with sepsis. Tragically, the patient succumbed at seven months due to a brain abscess stemming from an infected external ventricular drain. Ultimately, postmortem examination unraveled double alimentary tract obstruction, consisting of RRSA and CCS. The RRSA, originating from proximal thoracic aorta, caused notable esophageal compression and functional stenosis which led to the frequent vomiting and reflux. The CCS involved the distal transverse colon, descending colon and proximal sigmoid colon, accounting for nearly 50% of the colon. The CCS was therefore the exact cause of intermittent abdominal distension. The stenotic colon contained submucosal and myenteric plexuses, excluding Hirschsprung disease.

Conclusion

This case highlights the diagnostic complexities of RRSA and CCS resulting in double gut obstruction and masquerading as non-IgE-mediated cow’s milk protein allergy and sepsis-induced ileus. Awareness of these rare coexisting congenital anomalies can aid in early recognition, prevent misdiagnosis, enable timely management and improve outcomes for affected pediatric patients.

มะเร็งเม็ดเลือดขาวชนิด DLBCL ที่มีการแสดงออกของโปรตีน MYC และ BCL2 จะมีความรุนแรงมากและมีอัตราการรอดชีวิตของผู้ป่วยน้อย คณะวิจัยจึงได้นำข้อมูลทางคลินิกของผู้ป่วยโรคนี้จำนวน 111 คน วิเคราะห์ทางสถิติและพบว่าการเพิ่มการแสดงออกของโปรตีน MYC และ BCL2 ของเซลล์มะเร็งในสูตรของ NCCN-IPI ไม่สามารถเพิ่มประสิทธิภาพของการพยากรณ์โรคในระดับที่ยอมรับได้

Abstract

Background

MYC/BCL2 double expression (DE) is associated with poor prognosis in patients with diffuse large B-cell lymphoma (DLBCL) receiving rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisolone (R-CHOP). This study aimed to determine whether the addition of DE to the National Comprehensive Cancer Network Internal Prognostic Index (NCCN-IPI) could improve the prediction of disease progression in patients with DLBCL treated with R-CHOP.

Methods

This confirmatory prognostic factor study retrospectively recruited patients with newly diagnosed DLBCL between January 1, 2014, and January 31, 2018, at Ramathibodi Hospital (RA) and Thammasat University Hospital (TU). The follow-up period ended on July 1, 2022. Tumors expressing MYC ≥ 40% and BCL2 ≥ 50% were classified as DE. We calculated the hazard ratios (HR) for progression-free survival (PFS) from the date of diagnosis to refractory disease, relapse, or death. Discrimination of the 5-year prediction was based on Cox models using Harrell’s concordance index (c-index).

Results

A total of 111 patients had DE (39%), NCCN-IPI (8%), and disease progression (46%). The NCCN-IPI adjusted HR of DE was 1.6 (95% confidence interval [CI]: 0.9–2.8; P = 0.117). The baseline NCCN-IPI c-index was 0.63. Adding DE to the NCCN-IPI slightly increased Harrell’s concordance index (c-index) to 0.66 (P = 0.119).

Conclusions

Adding DE to the NCCN-IPI may not improve the prognostic value to an acceptable level in resource-limited settings. Multiple independent confirmatory studies from a large cohort of lymphoma registries have provided additional evidence for the clinical utility of DE.

ศัลยแพทย์ปัจจุบันนิยมผ่าตัดรักษาผู้ป่วยโรคหมอนรองกระดูกคอกดทับเส้นประสาทด้วยวิธี posterior endoscopic cervical foraminotomy หรือ discectomy (PECF หรือ PECD) มากขึ้น ทว่าองค์ความรู้เกี่ยวกับจุดสังเกตทางกายวิภาคเพื่อความปลอดภัยในการผ่าตัดนำกระดูกบางส่วนออกเพื่อลดการกดทับเส้นประสาทยังมีอยู่อย่างจำกัด

Abstract

This study investigates the relationships among specific bony structures of cervical vertebra, termed the “V point,” the dural sac, nerve roots, and intervertebral discs. The aim is to improve surgical outcomes and minimize complications in procedures like posterior endoscopic cervical foraminotomy or discectomy (PECF or PECD) for treating cervical radiculopathy. The research, conducted on 10 soft adult cadavers, measured distances between the V point and key anatomical features to provide practical guidance for surgical procedures.
The findings indicate that a 3-4 mm horizontal removal of bone from the V point is sufficient, a 3-4 mm vertical removal of bone in the upper direction is generally adequate at levels C3/4 to C5/6, and a more extensive bone cut of over 4 mm is recommended for levels C6/7 and C7/T1 for effective exposure and neural decompression. This information is valuable for surgeons offering insights that can enhance the efficiency of posterior endoscopic cervical procedures and lower the risk of complications. Surgeons can use these recommended bone removal guidelines to perform these surgeries more effectively, ultimately improving patient outcomes.

Study Design

Cadaveric anatomical studies.

Objective

This study aims to investigate the anatomical relationship between bony landmark “V point”, dural sac, nerve roots, and intervertebral disc for improving operative outcomes and decreasing post-operative complications in posterior endoscopic cervical foraminotomy or discectomy (PECF or PECD).

Method

10 soft adult cadavers were studied. We measured the distance of the V point to the lateral margin of dural sac, V point to the inferior border of intervertebral disc, and the inferior border of cervical nerve root to the inferior border of intervertebral disc. Then we calculated the mean of distance from V point to the inferior border of cervical nerve root.

Result

The mean distance from the V point to the lateral margin of dural sac from C3/4 to C7/T1 ranged from 3.1 ± 1.38 mm to 3.37 ± 1.46 mm. The mean distances from V point to the inferior border of intervertebral disc from C3/4 to C7/T1 were .19 ± 1.16 mm at C3/4, .45 ± 1.23 mm at C4/5, .43 ± 1.01 at C5/6, −.43 ± 1.86 mm at C6/7 and −1.5 ± 1.2 mm at C7/T1. The mean distance between V point and the inferior border of cervical nerve root from C3/4 to C7/T1 showed all positive value, ranging from .06 ± 1.18 mm to 4.45 ± 2.57 mm, increasing caudally.

Conclusion

In performing PECF or PECD, a 3-4 mm radius of bone removal should be enough for exposure and neural decompression at C3/4 to C5/6. At C6/7 and C7/T1 a more extensive bone cut of more than 4 mm is recommended, especially in cranial direction.

Abstract

Background

Soft tissue defects around the ankle are common and must be covered with thin and pliable flaps. A regional flap, particularly from the dorsum of the foot was considered ideal. A neurocutaneous flap, based on the superficial peroneal nerve (SPN) and its branches was designed as a proximally based flap via cadaveric dissection. This study aimed to demonstrate the vascularity and characteristics of the superficial peroneal neurocutaneous (SPNC) flap.

Materials and Methods

The SPNC flap was created in 11 lower limbs (seven cadavers) using a proximally based design. The skin flap was dissected at the dorsum of the foot, followed by injection of diluted methylene blue through the anterior tibial artery, to visualize the vascularity. The flap pedicle above the anterior ankle joint line was dissected along the SPN for anatomical study of perforating branches, paraneural vessels, and flap territory.

Results

The mean distances of the most proximal perforating branches were 1.51 ±1.48 cm from the anterior ankle joint line, and 5.12 ±1.78 cm from the lateral malleolus. The mean distances of the most distal perforating branches were 2.75 ± 1.54 cm from the anterior ankle joint line, and 5.90 ±1.81 cm from the lateral malleolus. The mean number of perforating branches was 3.73 ± 1.49. The mean flap territories were 5.51 ±0.59 cm in length, and 7.15 ± 0.64 cm in width.

Conclusion

The SPNC flap is an alternative method for soft tissue reconstruction around the ankle with a proximally based flap design. The antegrade flow has been shown to offer effective vascularity in flaps prepared via cadaveric dissection.

สารยับยั้ง SRPK1 ลดกระบวนการฟอสโฟรีเลชั่นของ SRSF1 ในเซลล์มะเร็งท่อน้ำดี ลดการเคลื่อนย้าย SRSF1 ไปยังนิวเคลียส เซลล์ที่ได้รับสารยับยั้ง SRPK1 มีระดับโปรตีน VEGFA-165b เพิ่มขึ้น เซลล์เยื่อบุผนังหลอดเลือด (HUVEC) ที่เพาะเลี้ยงด้วยอาหารเลี้ยงเซลล์มะเร็งท่อน้ำดีที่ได้รับสารยับยั้ง SRPK1 ที่มีปริมาณ VEGF-A165b สูง เกิดการสร้างหลอดเลือดใหม่น้อยลง

ที่มาและความสำคัญ

มะเร็งหลายชนิดมีการแสดงออกของโปรตีน SRPK1 ซึ่งเป็นโปรตีนควบคุมการ splicing ของ mRNA มากผิดปกติ บ่งชี้ว่ามีศักยภาพเป็นเป้าหมายการรักษามะเร็ง และมะเร็งระยะแพร่กระจายมีการแสดงออกของ angiogenic isoform ของ vascular endothelial growth factor (VEGF) มาก การศึกษานี้ตรวจสอบการแสดงออกของ SRPK1 และผลของการยับยั้ง SRPK1 ต่อความสามารถในการมีชีวิตและการสร้างหลอดเลือดใหม่ของเซลล์มะเร็งท่อน้ำดีโดยใช้สารยับยั้งต่อ SRPK1 คือ SPHINX31 ซึ่งเป็นโมเลกุลที่ยับยั้ง จำเพาะต่อ SRPK1 โดยมุ่งหวังผลต่อการยับยั้งการ splicing ของ angiogenic isoform ของ VEGF

Inhibition of serine/arginine-rich protein kinase-1 (SRPK1) prevents cholangiocarcinoma cells induced angiogenesis

Abstract

The serine/arginine-rich protein kinase-1 (SRPK1) is an enzyme that has an essential role in regulating numerous aspects of mRNA splicing. SRPK1 has been reported to be overexpressed in multiple cancers, suggesting it as a promising therapeutic target in oncology. No previous studies reported the role of SRPK1 in cholangiocarcinoma (CCA) cells. This study aimed to examine the expression of SRPK1 and the effects of SRPK1 inhibition on the viability and angiogenesis activity of CCA cells using a selective SRPK1 inhibitor, SPHINX31. Here, we demonstrate that SPHINX31 (0.3‐10 μM) had no inhibitory effects on CCA cells’ viability and proliferation. However, SPHINX31 decreased the mRNA expression of pro-angiogenic VEGF-A165a isoform. In addition, SPHINX31 attenuated SRSF1 phosphorylation and nuclear localization, and increased the ratio of VEGF-A165b/total VEGF-A proteins. Moreover, when HUVECs were grown in conditioned medium from SPHINX31-treated CCA cells, migration slowed, and tube formation decreased. The present study demonstrates that targeting SRPK1 in CCA cells effectively attenuates angiogenesis by suppressing pro-angiogenic VEGF-A isoform splicing. These findings suggest a potential therapeutic treatment using SRPK1 inhibitors for the inhibition of angiogenesis in cholangiocarcinoma.
