การใช้ยาปฏิชีวนะในพื้นที่เลี้ยงสัตว์ส่งผลกระทบต่อการเพิ่มจำนวนแบคทีเรียดื้อยาที่ก่อให้เกิดปัญหาด้านสุขภาพสาธารณะ งานวิจัยนี้มุ่งศึกษาแบคทีเรียดื้อยาที่พบในดินที่มีส่วนผสมของเพอร์ไลต์ในประเทศไทย โดยเปรียบเทียบระหว่างพื้นที่ที่ได้รับและไม่ได้รับผลกระทบจากการเลี้ยงสัตว์ ผลการศึกษาพบว่า Bacillus spp. ซึ่งดื้อยาซีเฟพีม (cefepime) เป็นสายพันธุ์ที่พบมากที่สุดทั้งในพื้นที่ที่ได้รับและไม่ได้รับผลกระทบ นอกจากนี้ ยังพบ Bacillus cereus ที่ดื้อต่อยาหลายชนิดในพื้นที่ที่ไม่ได้รับผลกระทบ การศึกษานี้เน้นให้เห็นว่าคุณสมบัติทางกายภาพและชุมชนจุลินทรีย์ในดินมีบทบาทสำคัญต่อการดื้อยาในดินมากกว่าการใช้ยาปฏิชีวนะในสัตว์เลี้ยง

Abstract

Antibiotic resistant bacteria are present in various environments and are concerning for public health. Antibiotic resistance genes in soil may proliferate throughout different ecosystems and be acquired by pathogens that pose health risks to humans and animals. This study investigated the antibiotic resistance of cultivable bacteria isolated from livestock-exposed and unexposed areas in a former perlite mining area in the Fa-La-Mee Mountain range, Lopburi, Thailand. Soil samples were collected in five different locations. Soil bacteria were isolated and tested for susceptibility against seven antibiotics, followed by 16S rRNA gene-based identification. A total of 56 bacterial isolates were isolated from the perlite-rich soil samples, including 34 isolates from the livestock-exposed areas and 22 isolates from the unexposed areas. Most of the isolates were resistant to cefepime (26.5% in exposed areas and 22.7% in unexposed areas) and ampicillin (23.5% in exposed areas and 22.7% in unexposed areas). 16S rRNA gene sequences revealed that most of the resistant bacteria isolated from perlite-rich soil were Bacillus spp. A multidrug-resistant isolate of Bacillus cereus resistant to ampicillin, trimethoprim/ sulfamethoxazole, and cefepime was found in a livestock-unexposed area. The majority of the culturable antibiotic-resistant bacteria isolated from perlite-rich soil were cefepime-resistant Bacillus. Interestingly, the pattern of antibiotic resistance in exposed and unexposed areas was not different, which indicated that antibiotic resistance in perlite-rich soil is likely caused by other factors, such as physicochemical characteristics and/or the microbial population of the soil rather than by antibiotics used in livestock husbandry.

การศึกษาก่อนหน้ารายงานว่า Ag-NPs สามารถป้องกันการติดเชื้อ Vibrio sp. ในกุ้งได้ ประสิทธิภาพของอนุภาคไบเมทัลลิคนาโน Au/Ag (BNPs) ต่อการติดเชื้อจาก V. parahaemolyticus ในกุ้งยังไม่เคยถูกศึกษามาก่อน นอกจากนี้ยังไม่มีรายงานเกี่ยวกับคุณสมบัติของ Au/Ag-NPs ที่สังเคราะห์กับซัลเฟตกาแล็กแตน (SG) ในการต้านแบคทีเรีย การศึกษาครั้งนี้มีวัตถุประสงค์เพื่อรวม BNPs กับ SG และประเมินประสิทธิภาพในการต้านการเจริญและป้องกันฟิล์มแบคทีเรีย V. parahaemolyticus และ V. harveyi รวมทั้งประสิทธิผลในการป้องกันการติดเชื้อจาก V. parahaemolyticus ในกุ้ง Penaeus vannamei ผ่านการเสริมอาหาร งานวิจัยนี้เป็นการศึกษาครั้งแรกที่ศึกษาผลของ BNPs ที่รวมกับ SG ต่อเชื้อแบคทีเรีย V. parahaemolyticus ในกุ้ง

Abstract

Bimetallic (Au/Ag) nanoparticles (BNPs) have shown enhanced antibacterial activity compared to their monometallic counterparts. Sulfated galactans (SG) are a naturally occurring polymer commonly found in red seaweed Gracilaria fisheri. They are biocompatible and biodegradable and environmentally friendly. In this study, we utilized SG in combination with BNPs to develop composite materials that potentially enhance antibacterial activity against shrimp pathogens Vibrio parahaemolyticus and Vibrio harveyi, compared to BNPs or SG alone. BNPs were coated with sulfated galactan (SGBNPs) and characterized using UV–vis spectroscopy, Fourier transform infrared (FTIR) spectroscopy, zeta potential, and transmission electron microscopy (TEM). UV–vis spectroscopy analysis revealed that the surface plasmon peaks of BNPs and SGBNPs appeared at 530 nm and 532 nm, respectively. Zeta potential measurements showed that SGBNPs had a negative charge of −32.4 mV, while the BNPs solution had a positive charge of 38.7 mV. TEM images demonstrated the spherical morphology of both BNPs and SGBNPs with narrow size distributions (3–10 nm). Analysis of the FTIR spectra indicated that SG maintained its backbone structure in SGBNPs, but some functional groups were altered. Notably, SGBNPs showed superior antimicrobial and antibiofilm activities against V. parahaemolyticus and V. harveyi compared to SG and BNPs. Furthermore, treatment with SGBNPs significantly down-regulated the expression of virulence-related genes (toxRcpsQ, and mfpA) for V. parahaemolyticus 3HP compared to the respective control, bacteria treated with BNPs or SG. Diets supplemented with SGBNPs, BNPs, or SG showed no detrimental impact on the growth of shrimp Penaeus vannamei. Shrimp fed with SGBNPs-supplemented feed showed significantly higher survival rates than those fed with BNPs-supplemented feed when infected with 3HP after being on the supplemented feed for seven days and a subsequent number of fifteen days. These findings collectively demonstrate the benefit of using SG capped Au–Ag BNPs as an antibacterial agent for the prevention and control of Vibrio sp. Infection in shrimp while reducing the risk of environmental contamination.
