คณะผู้วิจัยได้ทำการศึกษาผลของกลูตาเมท (Glutamate, Glu) ต่อการกระตุ้นการกินอาหาร การเปลี่ยนแปลงของเนื้อเยื่อวิทยาของลำไส้ การเจริญเติบโต และระบบแอนตี้ออกซิแดนซ์ภูมิคุ้มกัน ในลูกกุ้งก้ามกรามระยะจูวิไนล์ โดยพบว่า การให้อาหารที่ผสมด้วย Glu ที่ระดับความเข้มข้น 55 และ 550 mg/kg feed สามารถกระตุ้นการกินอาหาร โดยการเพิ่ม food consumption, digestive enzyme activities, และกระตุ้น feeding behaviors นอกจากนี้ยังพบการเปลี่ยนแปลงของเนื้อเยื่อวิทยาของลำไส้ เมื่อเปรียบเทียบกับกลุ่มควบคุม (P < 0.05) สารสื่อประสาท Glu สามารถกระตุ้นการเจริญเติบโต โดยการเพิ่มขนาด, weight gain (WG), growth rate (GR), specific growth rate (SGR), และaverage daily gain (ADG) ในช่วงสัปดาห์ที่ 8 ถึง 12 เมื่อเปรียบเทียบกับกลุ่มควบคุม อย่างมีนัยสำคัญทางสถิติ (P < 0.05). นอกจากนี้ อาหารที่ผสม Glu สามารถเพิ่มการแสดงออกของ appetite/feeding-related gene อาทิ neuropeptide F (NPF) และ muscle growth-, immune-related genes ตลอดจน immune ต่างๆ รวมทั้ง พบอัตราการอยู่รอด (Survival rate) ที่สูงกว่ากลุ่มควบคุม (P < 0.05) จากผลการวิจัยนี้ คณะผู้วิจัยได้ค้นพบองค์ความรู้พื้นฐานใหม่ที่สำคัญมาก ที่แสดงให้เห็นว่า Glu สามารถกระตุ้นการกินอาหาร การเจริญเติบโต และภูมิคุ้มกัน ในลูกกุ้งก้ามกรามระยะจูวิไนล์ นอกจากนี้ ยังค้นพบกลไกการควบคุม Glu ที่สำคัญต่อการกินอาหาร และ/หรือ การเจริญเติบโต ที่อาจจะทำงานร่วมกัน (interplay) กับ NPF ในการควบคุมสรีระวิทยาเหล่านี้ โดยองค์ความรู้ใหม่ที่ได้รับนี้ อาจจะสามารถนำ Glu ซึ่งมีราคาไม่แพง นำมาประยุกต์ใช้โดยการผสมอาหาร (functional feed) เพื่อเพิ่มอัตราการกินอาหาร การเจริญเติบโต ตลอดจนเพิ่มภูมิคุ้มกัน ในการเพาะเลี้ยงลูกกุ้งก้ามกรามได้ในอนาคต

ที่มาและความสำคัญ

The giant freshwater prawn (Macrobrachium rosenbergii) culturing in Thailand currently faces significant challenges, notably the high-density captive breeding system, which increases stress and retards growth rate of juvenile prawns. Stress leads to diminished appetite, decreased food intake, stunted growth, heightened vulnerability to diseases, and elevated mortality rates. Feeding is an important process for acquiring nutrients essential to energy production and growth in juvenile prawns. Numerous factors influence feeding, including food type, temperature, light, hormones, and neurotransmitters (Volkoff, 2016; Volkoff & Rønnestad, 2020; Tierney, 2020). These factors interact through complex regulatory mechanisms that control digestive function for nutrient absorption, energy balance, and growth. Increasing feeding rates can also lead to higher growth, improved health, as well as enhanced production and economic value of cultured prawns. Glutamate (Glu) is a neurotransmitter and a versatile non-essential amino acid. Glu in intestinal cells stimulates neural networks, consequently influencing brain regions through nutrient sensing. This indicates that Glu is a significant regulator of food consumption, digestion, and nutrient absorption. However, the effects and underlying regulatory mechanisms of Glu on feed consumption, digestibility, growth, appetite-related peptides, and antioxidant–immune gene expression in juvenile freshwater prawn, M. rosenbergii, remain unclear. Given its physiological importance, Glu may serve as a key regulator of feeding, growth, and immune responses in juvenile prawns.

Abstract

The aim of the study is to explore the effects of glutamate (Glu) supplementation on feed consumption, digestibility, intestinal histology, growth, gut barrier, and the antioxidant–immune-related genes during the juvenile stage of freshwater prawn (Macrobrachium rosenbergii). The supplementation of Glu at doses of 55 and 550 mg/kg feed significantly enhanced feed consumption, digestion, digestive enzyme activities, and gut histology (P < 0.05). Feeds supplemented with these two doses of Glu improved growth, demonstrated by increased body size, weight gain (WG), growth rate (GR), specific growth rate (SGR), and average daily gain (ADG) during weeks 8–12 (P < 0.05). Additionally, juvenile prawns fed diets supplemented with both doses of Glu exhibited significant upregulation of appetite-related gene, as well as muscle growth-related and immune-related genes (P < 0.05). Survival rate (SR) was significantly higher for juvenile prawns receiving diets with Glu supplementation at weeks 8 and 12. Collectively, these results indicate that Glu function as an appetite stimulant, growth enhancer, and immune modulator; therefore, this neurotransmitter may be a potential food additive for improving the production of juvenile prawns raised in an aquaculture system.

ที่มาและความสำคัญ

Cultivation of freshwater prawn (Macrobrachium rosenbergii) has expanded considerably, thus emerging as a substantial contributor to Thailand’s aquaculture sector (New and Nair 2012; Tinikul et al., 2023; FAO, 2024). The global production of M. rosenbergii has increased significantly, reaching more than 300,000 tons. Meanwhile, demand has also grown significantly in both local and international markets. However, freshwater prawn farming also faces considerable obstacles, particularly disease outbreaks and hormonal imbalances in prawns caused by stressful situations, such as high-density culture, global warming, and overcrowded agricultural habitats (Hooper et al., 2023; Bhassu et al., 2024). These environmental stressors can lead to decreased feeding, nutritional deficiency, high mortality rate, and retarded growth, ultimately resulting in reduced productivity and significant economic losses (Pillai and Bonami, 2012; Kamaruding and Abdullah, 2021; Calado et al., 2022; Paul et al., 2025). Hence, feeding is a crucial behavior that ensures the intake of sufficient nutrients to maintain energy balance for these physiological processes (Celi et al., 2017). Feed constitutes the main expense in the aquaculture and ensures the quality, quantity, and good health of the prawns being raised (Yu et al., 2021). Thus, it is important to develop a suitable feed that optimizes feeding rates, promotes growth, and enhances immunity during the key developmental stages of this prawn. Neuropeptide F (NPF) is an important neuropeptide that potentially regulates feeding, foraging, metabolism, development, and reproduction, in insects and crustaceans (Fadda et al., 2019; Zhao et al., 2023). Considering the critical roles of NPF in regulating feeding, digestive enzyme activity, development and immunity in insects and crustaceans, we question whether M. rosenbergii NPF (MrNPF) could promote feeding, digestive functions, growth and immunity in juvenile prawns. This important knowledge of MrNPF supplementation may be developed as a viable feed additive in prawn aquaculture to improve feeding, growth performance, muscle mass, and health status of juvenile prawns, M. rosenbergii.

Abstract

Neuropeptide F (NPF) regulates feeding, growth, development, and reproduction in insects and crustaceans. In this study, we examined the effects of Macrobrachium rosenbergii NPF (MrNPF) on food intake, digestive functions, growth performance, and immunity in juvenile freshwater prawns (M. rosenbergii). Supplementing MrNPF at 5 and 0.5 μg/g of feed significantly stimulated food intake and digestive functions by enhancing food consumption, hepatosomatic index, and feeding behaviors; increasing digestive amylase, protease, and lipase activities; and improving gut histomorphology (P < 0.05). In addition, both doses of MrNPF-supplemented feed promoted growth performance by increasing mean weight gain (WG), specific growth rate, and daily WG from weeks 6 to 12 compared with the control groups (P < 0.05). Survival rate was significantly higher in the high-dose MrNPF group than in controls only at week 9 (P < 0.05). Furthermore, MrNPF supplementation significantly upregulated the expression of muscle growth-related genes, including MrTORelF4B, and LIM mRNA. Finally, MrNPF significantly enhanced the expression of immune-related genes. Collectively, our findings suggest that MrNPF is an essential neuropeptide that may serve as a potential regulator of feeding, growth, and immunity in juvenile M. rosenbergii. This knowledge could be used to enhance the aquaculture productivity of this crustacean species.

ที่มาและความสำคัญ

Currently, prawn aquaculture with high-density populations faces several challenges, including diseases and infections, which cause stress and lead to diminished appetite and reduced feeding (Hooper et al., 2023). These factors directly impact gonadal development, particularly in the ovaries, leading to a decrease in the quantity and quality of eggs and, consequently, reduced offspring production. Feeding is affected by external factors such as water temperature, light, and food composition and taste properties (Kestemont and Baras, 2001; Volkoff and Rønnestad, 2020). In addition, feeding is regulated by internal factors, including various neurotransmitters, e.g., glutamate (Glu), serotonin (5-HT), and catecholamines (Scannell et al., 2008; Tierney, 2020). These factors are crucial in controlling feeding by activating the digestive and nervous systems, which is essential for assuring enough nutrient ingestion to maintain energy balance in the body (Celi et al., 2017). In female crustacean broodstocks, feeding is a crucial physiological process to acquire sufficient nutrients that are utilized to produce energy for ovarian development, oocyte production, and vitellogenesis. The quantity and quality of oocytes influence the successful development of fertilized eggs, which subsequently impacts crustacean production (Fernández-Palacios et al., 2011; Feng et al., 2023). Considering the importance of Glu and 5-HT in physiological functions, such as feeding, digestive enzyme activity, and immunity in aquatic animals like fish and decapod crustaceans, we question whether these neurotransmitters display alterations and regulatory functions in the digestive organs as essential modulators of feeding and immunity in female M. rosenbergii during the ovarian cycle.

Abstract

This study investigated the presence of glutamate (Glu) and serotonin (5-HT) in the digestive organs and their effects on feeding performance, digestive enzyme activity, and immunity throughout the ovarian cycle in female giant freshwater prawns (Macrobrachium rosenbergii). By high-performance liquid chromatography (HPLC), Glu and 5-HT were detected in the foregut, midgut, hindgut, and hepatopancreas, at the highest concentrations in the late ovarian stage and the lowest concentrations in the early ovarian stage. Through immunohistochemistry, the presence of Glu-immunoreactivity (-ir) and 5-HT-ir were observed in all digestive organs, and their intensities were shown to correlate with the levels detected by those observed via HPLC. Prawns injected with Glu and 5-HT at the doses of 2.5 × 10−6 and 2.5 × 10−7 mol/prawn, markedly increased food consumption, gastric emptying, and midgut index as well as enhanced amylase, protease, and lipase activities, compared with the control group (P < 0.05). Glu and 5-HT also enhanced epithelial cell height, villi height, and crypt depth and promoted the reactivity of PAS/AB in the digestive organs. In addition, treatment with Glu or 5-HT significantly upregulated M. rosenbergii NPF (MrNPF) and immune-related transcripts (P < 0.05). Collectively, these findings demonstrated the promising effects of Glu and 5-HT at a dose of 2.5 × 10−6 mol/prawn in stimulating feeding, digestive activities, and immunity, which could potentially be used to enhance feeding, growth, and maintain the health status of female prawn in aquaculture.

การศึกษาลักษณะและการเปลี่ยนแปลงทางกายวิภาค สัณฐานวิทยา และเนื้อเยื่อวิทยาของระบบทางเดินอาหาร ในช่วงการพัฒนาของรังไข่ของกุ้งก้ามกรามเพศเมีย

ในการศึกษาครั้งนี้ คณะผู้วิจัยได้ทำการศึกษาลักษณะและการเปลี่ยนแปลงทางกายวิภาค สัณฐานวิทยา และเนื้อเยื่อวิทยา ของระบบทางเดินอาหารส่วน foregut (FG), midgut (MG), and hindgut (HG) ของกุ้งก้ามกรามเพศเมีย Macrobrachium rosenbergii ในช่วงการพัฒนาของรังไข่ ในส่วนของ FG ประกอบไปด้วย หลอดอาหาร (esophagus, ESO), cardia (CD), `และ pylorus (PY) ซึ่งมีลักษณะ epithelium (Ep) บุด้วยชั้น cuticle (Cu) ซึ่งหนา และมีต่อม (ESO glands) อยู่บริเวณนี้  ส่วน CD มีกล้ามเนื้อที่หนา และบริเวณ PY นั้นมีชั้นกล้ามเนื้อ inner longitudinal (LM) และ the outer circular (CM) รวมทั้งพบ villi จำนวนมาก ในส่วนของ MG จะไม่พบ cuticle แต่จะพบ microvilli และ subapical vacuoles บริเวณ apical surface ของเซลล์บุผิว ส่วนชั้นนอกสุด ของ MG จะพบ a dense strip ของ elastic fibers และกลุ่มของ collagen fibers รวมตัวกันอยู่ ในส่วนของ HG ประกอบด้วยชั้น cuticle บางกว่า FG และจะพบ HG glands คล้าย rosette-like structure ใน HG นั้น ประกอบด้วย CM และ the LM fibers นอกจากนี้ reactivity เมื่อทำการย้อมสีด้วย Periodic Acid Schiff/Alcian และการแสดงออกของ mucin พบว่า มีการเปลี่ยนแปลงไปตามระยะการพัฒนาของรังไข่ ซึ่งจากผลการวิจัยนี้ ได้ให้องค์ความรู้พื้นฐานใหม่ที่สำคัญอย่างมากในส่วนของลักษณะทางกายวิภาคและเนื้อเยื่อวิทยา ของระบบทางเดินอาหารในช่วงการพัฒนาของรังไข่ ของกุ้งก้ามกรามเพศเมีย M. rosenbergii ซึ่งสามารถนำองค์ความรู้ที่ได้นี้ไปประยุกต์ใช้ ในการพัฒนาการกินอาหารและการพัฒนาของรังไข่ในการเพาะเลี้ยงกุ้งก้ามกรามเพศเมีย ได้ในอนาคต

ที่มาและความสำคัญ

Basic understanding of the anatomy and physiology of the digestive system in decapod crustaceans is essential for comprehending the nutrition uptake and processing of this prawn M. rosenbergii species, and other decapod species. This will help to further develop aquaculture for this prawn species. However, the detailed morphological, histological, and histochemical characteristics of the FG, MG, and HG of the female prawn, M. rosenbergii during the ovarian cycle have not yet been investigated. Thus, we question whether there are morphological and histochemical changes in digestive organs that may be related to the ovarian maturation cycle in this female M. rosenbergii. These changes may reflect the functional connection between feeding and nutritional processing by the digestive tract which may serve ovarian maturation in this crustacean species.

Abstract

The anatomical, histological, and histochemical characteristics of the foregut (FG), midgut (MG), and hindgut (HG), as well as their alterations during the ovarian cycle in female prawns, Macrobrachium rosenbergii, were investigated. The esophagus (ESO), cardia (CD), and pylorus (PY) are the main components of the FG. An epithelium (Ep) with thick cuticle (Cu) layers lining the ESO, and the ESO is encircled by the ESO glands. The CD has a thick musculature, whereas the Ep of the PY are characterized by numerous villi and columnar Ep cells with a thinner layer of Cu. The inner longitudinal (LM) and the outer circular (CM) muscles were both present in the PY. The MG is lined by Ep cells which are connected to the basement membrane, and it lacks Cu. Microvilli, and subapical vacuoles are visible on the apical surface of Ep cells of the MG. The outermost layer is characterized by a dense strip of elastic fibers and a cluster of collagen fibers. The HG has the Ep cells with a thin Cu layer, and the HG glands form a rosette-like structure. The HG is surrounded by the CM and the LM fibers. The reactivities of Periodic Acid Schiff and Alcian Blue in these digestive organs altered throughout the ovarian cycle, and this was supported by the increased expression of mucin levels as ovarian maturation progressed. Our results offer novel and significant insights into the anatomical and histochemical structures of these digestive organs, and demonstrate a significant correlation between ovarian development and feeding in the female prawn, M. rosenbergii.

ในการศึกษาครั้งนี้ เราได้ตรวจสอบการเปลี่ยนแปลงของระดับการแสดงออกและการกระจายของ M. rosenbergii neuropeptide F (MrNPF) ในอวัยวะทางเดินอาหารในช่วงการพัฒนาของรังไข่ของกุ้งก้ามกรามเพศเมีย M. rosenbergii คณะผู้วิจัยได้ทำการตรวจสอบพบว่า MrNPF มีการแสดงออกอย่างจำเพาะเจาะจงในแต่ละส่วนของทางเดินอาหาร และระดับของMrNPF มีการเปลี่ยนแปลงไปตามช่วงการพัฒนาของรังไข่ จากนั้นคณะผู้วิจัยได้ทำการศึกษาต่อไปถึงความเป็นไปได้ของ mode of action ของ MrNPF ในการควบคุมกิจกรรมของเซลล์ในระบบทางเดินอาหารและอาจจะเป็นไปได้ถึงการควบคุมการกินอาหาร จากการค้นพบครั้งนี้ ทำให้เราได้รับองค์ความรู้ใหม่ที่สำคัญเกี่ยวกับความสัมพันธ์ของ MrNPF กับการกินอาหารในช่วงการพัฒนารังไข่ในกุ้ง M. rosenbergii ตัวเมีย และยังทำให้เราเข้าใจมากขึ้นเกี่ยวกับกลไกการควบคุมของ MrNPF ต่อการกิจกรรมของเซลล์ในระบบทางเดินอาหาร ซึ่งองค์ความรู้ใหม่ที่ได้รับนี้อาจจะนำไปประยุกต์ใช้ต่อยอดในการศึกษาเพิ่มเติมเกี่ยวกับบทบาทและการออกฤทธิ์ของ NPF ในการควบคุมการกินอาหารและกระบวนการทางสรีรวิทยาอื่นๆ ของระบบทางเดินอาหารต่อไปได้

ที่มาและความสำคัญ

Neuropeptide F (NPF) is a key hormone that controls feeding in invertebrates, including decapod crustaceans. In female M. rosenbergii, there may be a correlation between feeding and nutritional status with ovarian development, thus, the expression levels of M. rosenbergii NPF (MrNPF) and its anatomical distribution in various digestive organs during the ovarian cycle in female M. rosenbergii were examined. In the present study, we investigated changes in the levels, differential expression, and distribution of MrNPF in the digestive organs of mature M. rosenbergii females during the ovarian cycle. We further examined whether NPF is expressed in neurons that may reside in the digestive tract by colocalizing MrNPF with the two neuronal markers, including protein gene product 9.5 (PGP 9.5/UCHL-1) and choline acetyltransferase (ChAT), which is important to understand the possible mode of action of NPF to control digestive cell activities. The findings of this study shed important light on the differential expression of MrNPF in the digestive organs and its correlation with feeding and ovarian development in M. rosenbergii females. The knowledge acquired provides the impetus for further research into the possible role of NPF in the regulation of the feeding and other physiological processes of the digestive tract of this commercially important crustacean species.

Abstract

Neuropeptide F is a key hormone that controls feeding in invertebrates, including decapod crustaceans. We investigated the differential expression of Macrobrachium rosenbergii neuropeptide F (MrNPF) in the digestive organs of female prawns, M. rosenbergii, during the ovarian cycle. By using RT-qPCR, the expression of MrNPF mRNA in the esophagus (ESO), cardia (CD), and pylorus (PY) of the foregut (FG) gradually increased from stage II and peaked at stage III. In the midgut (MG), hindgut (HG), and hepatopancreas (HP), MrNPF mRNA increased from stage I, reaching a maximal level at stage II, and declined by about half at stages III and IV (P < 0.05). In the ESO, CD, and PY, strong MrNPF-immunoreactivities were seen in the epithelium, muscle, and lamina propria. Intense MrNPF-ir was found in the MG cells and the muscular layer. In the HG, MrNPF-ir was detected in the epithelium of the villi and gland regions, while MrNPF-ir was also more intense in the F-, R-, and B-cells in the HP. However, we found little colocalization between the MrNPF and PGP9.5/ChAT in digestive tissues, implying that most of the positive cells might not be neurons but could be digestive tract-associated endocrine cells that produce and secrete MrNPF to control digestive organ functions in feeding and utilizing feed. Taken together, our first findings indicated that MrNPF was differentially expressed in digestive organs in correlation with the ovarian cycle, suggesting an important link between MrNPF, the physiology of various digestive organs in feeding, and possibly ovarian maturation in female Mrosenbergii.

In the sea cucumber, Holothuria scabra, the competent larvae require main settlement organs (SOs), including the ciliary bands, tentacles, and podia, as well as cues from major neurotransmitters, including gamma-aminobutyric acid (GABA) and dopamine (DA), for successful settlement. However, the presence and differential expression of DA during the developmental stages in H. scabra, have not yet been investigated. Additionally, the exact location for the production and secretion of these two neurotransmitters in the SO needs to be characterized so that this important knowledge may help increase the production of this sea cucumber species. In the present study, we investigated and mapped the presence and distribution of GABA and DA in several organs that may be involved in mediating the settlement of H. scabra larvae, with special emphasis on SOs, during different developmental stages. Our findings shed important light on the anatomical localization and potential implications of the two neurotransmitter systems in SOs to control the larval settlement process in this sea cucumber species.

Abstract

In the sea cucumber, Holothuria scabra, the competent larvae require main settlement organs (SOs), including the ciliary bands (CiBs), tentacles (Ts), podia (PDs), and cues from neurotransmitters, including gamma-aminobutyric acid (GABA) and dopamine (DA), for successful settlement. In the present study, we investigated the spatial distribution of GABA and DA in the developmental stages of H. scabra, with special emphasis on SOs by detecting immunoreactivity (-ir) against these two neurotransmitters. Strong GABA-ir and DA-ir cells and fibers were specifically detected in several SO structures, including CiBs, CiB cells (CiBCs), and long cilia (LCi), of H. scabra larvae. Additionally, we found intense GABA-ir and DA-ir cells in the epithelial lining of bud-papillae (BP) and mesothelium (Me) in the stem (S) region of Ts in larvae and juveniles. Intense GABA-ir and DA-ir were observed in the epineural nerve plexus (ENP) and hyponeural nerve plexus (HNP) of Ts in H. scabra pentactula and juvenile stages. Staining for these two neurotransmitters was particularly intense in the PDs and their nerve fibers. We also found significant changes in the numbers of GABA-ir and DA-ir-positive cells and intensities in the CiBs, Ts, and PDs during the developmental stages. Taken together, we are the first to report on the existence and distribution of GABAergic and dopaminergic systems in structures associated with the settlement. Our findings provide new and important insights into the possible functions of these two neurotransmitters in regulating the settlement of this sea cucumber species.

คณะผู้วิจัยได้พบองค์ความรู้พื้นฐานใหม่ที่สำคัญคือ short neuropeptide F (Mro-sNPF) มีผลต่อการพัฒนาของรังไข่และการตกไข่ในกุ้งก้ามกรามเพศเมีย Macrobrachium rosenbergii โดยพบว่า เปปไทด์ Mro-sNPF ทำให้ระยะเวลาการพัฒนาของรังไข่สั้นลง เพิ่มดัชนีโกนาโดโซมาติก (GSI) เพิ่มขนาดเส้นผ่านศูนย์กลางไข่ (OD) และการเพิ่มจำนวนการแบ่งเซลล์ เมื่อเปรียบเทียบกับกลุ่มควบคุม (P<0.05) นอกจากนี้พบว่า เปปไทด์ Mro-sNPF เพิ่มระดับความเข้มข้นของ hemolymph vitellogenin และการเพิ่มระดับของ MrEcR เมื่อเปรียบเทียบกับกลุ่มควบคุม อย่างมีนัยสำคัญทางสถิติ (P<0.05) เปปไทด์ Mro-sNPF ยังคงเหนี่ยวนำให้เกิดการตกไข่ได้เร็วขึ้นโดยลดระยะเวลาลงครึ่งหนึ่ง เมื่อเทียบกับระยะเวลาการตกไข่ของกลุ่มควบคุม (P<0.05) เปปไทด์ Mro-sNPF มีศักยภาพในการกระตุ้นการผลิตเซลล์สืบพันธุ์ การพัฒนาของไข่ และการตกไข่ในกุ้งก้ามกราม ตลอดจนน่าจะสามารถนำไปการประยุกต์ใช้ในระบบเพาะเลี้ยงต่อไป

ที่มาและความสำคัญ

In decapod crustaceans, eyestalk ablation has been practiced by aquaculturists to enhance commercial production through the induction of ovarian maturation; however, this technique eventually leads to undesirable effects of decline in egg quality and quantity, precocious development of gonads, and early death of spawners. In recent years, hormonal manipulation has proven a more sustainable alternative to successfully promote ovarian development, and this approach has the potential to replace eyestalk ablation. Understanding of hormones controlling the development of gonads, which produce quality gametes for larval production, is essential to the success of crustacean aquaculture. Thus, in this study, we are the first to report the potent effect and regulatory mechanisms of short neuropeptide F (Mro-sNPFs) on the induction of ovarian maturation and spawning, and associated regulatory mechanisms in M. rosenbergii females. Therefore, Mro-sNPF treatment could be applied to solve prawn aquaculture problems by increasing gamete production of this crustacean species for aquaculture.

Abstract

  In the present study, we investigated the effects of two short neuropeptide F (Mro-sNPF) mature sequences, namely APALRLRFamide and DRTPALRLRFamide, on ovarian maturation and spawning in Macrobrachium rosenbergii females. At doses of 10−5 and 10−6 mol/prawn, both Mro-sNPF peptides significantly shortened the ovarian developmental period (P < 0.05), but enhanced the gonadosomatic index (GSI), oocyte diameter (OD), and cell proliferation, compared with control values (P < 0.05). However, at the lowest dose (10−7 mol/prawn), neither peptide significantly affected ovarian maturation, GSI, nor OD compared with the control values. Females treated with the two peptides at the dose of 10−5 or 10−6 mol/prawn attained mature oocyte stages faster than did control females. Treatment with both peptides significantly elevated hemolymph vitellogenin concentrations and upregulated MrEcR transcripts, compared with the control treatment (P < 0.05). Finally, both Mro-sNPFs induced spawning by shortening its duration by half (∼1.5 days) compared with the control duration (∼3.1 days) (P < 0.05). The number of eggs per spawn was approximately 15.4 × 104 eggs in the experimental group and in the control group (∼15.35 × 104 eggs), albeit without significance. Therefore, among the treatment groups, using APALRLRFamide at 10−5 mol/prawn showed promising effects on the induction of ovarian maturation and spawning. Taken together, Mro-sNPF peptides can promote ovarian development and spawning, indicating their potential application to stimulate gamete and larval production in this important crustacean aquaculture species.

ในการศึกษานี้ คณะผู้วิจัยได้ค้นพบและทำการศึกษา Transcriptome sequencing ของปลิงทะเลขาวกลุ่ม Holothuroidea Holothuria scabra และได้ศึกษาคุณลักษณะของ putative neuropeptides ใน H. scabra และ H. graberrima transcriptomes. นอกจากนี้ยังได้ทำการศึกษาและวิเคราะห์เปรียบเทียบ และศึกษาการแสดงออกของ holothurian specific neuropeptides.

ที่มาและความสำคัญ
ในปัจจุบัน ได้มีความสนใจในการบริโภคปลิงทะเลเพิ่มขึ้น เนื่องจากว่า คุณสมบัติทางโภชนาการของปลิงทะเลมีประโยชน์ต่อสุขภาพอย่างมาก รวมทั้งการป้องกันหรือรักษาโรคต่างๆ ดังนั้น การศึกษาที่ผ่านมามุ่งเน้นไปที่การทำความเข้าใจชีววิทยาของปลิงทะเลเพื่อปรับปรุงการเพาะเลี้ยงสัตว์น้ำ อาทิ การใช้ neuropeptides เพื่อเพิ่มการเจริญเติบโตและการสืบพันธุ์ อย่างไรก็ตาม ได้มีการสำรวจ neuropeptidome ของปลิงทะเล Apostichopus japonicus ซึ่งมีการวิเคราะห์และระบุการถอดรหัส แต่ยังไม่มีการศึกษาใน H. scabra เลย ดังนั้นในการศึกษานี้ เราทำการวิเคราะห์ in silico ของ H. scabra transcriptomes ที่ได้มาจาก วงแหวนประสาท เส้นประสาทเรเดียล รังไข่ และ RNA ของเนื้อเยื่ออัณฑะ นอกจากนี้เรายังเปรียบเทียบการค้นพบของเราจาก H. scabra กับข้อมูลที่ได้จากสายพันธุ์ที่เกี่ยวข้องอย่างใกล้ชิด H. glaberrima และนำมาเปรียบเทียบกับข้อมูลจาก echinoderms และ bilaterians อื่น ๆ ทำให้สามารถระบุถึงการกระจายของสายวิวัฒนาการอย่างกว้างขวางซึ่งอาจเป็นเอกลักษณ์ของ echinoderms อื่นๆโดยเฉพาะในปลิงทะเล

Abstract 
Neuropeptides synthesized and released by neuronal cells play important roles in the regulation of many processes, e.g. growth, feeding, reproduction, and behavior. In the past decade, next-generation sequencing technologies have helped to facilitate the identification of multiple neuropeptide genes in a variety of taxa, including arthropods, molluscs and echinoderms. In this study, we extend these studies to Holothuria scabra, a sea cucumber species that is widely cultured for human consumption. In silico analysis of H. scabra neural and gonadal transcriptomes enabled the identification of 28 transcripts that encode a total of 26 bilaterian and echinoderm-specific neuropeptide precursors. Furthermore, publicly available sequence data from another sea cucumber, Holothuria glaberrima, allowed a more in-depth comparative investigation. Interestingly, two isoforms of a calcitonin-type peptide precursor (CTPP) were deduced from the H. scabra transcriptome – HscCTPP-long and HscCTPP-short, likely the result of alternative splicing. We also identified a sea cucumber relaxin-type peptide precursor, which is of interest because relaxin-type peptides have been shown to act as gonadotropic hormones in starfish. Two neuropeptides that appear to be holothurian-specific are GLRFA, and GN-19. In H. scabra, the expression of GLRFA was restricted to neural tissues, while GN-19 expression was additionally found in the longitudinal muscle and intestinal tissues. In conclusion, we have obtained new insights into the neuropeptide signaling systems of holothurians, which will facilitate physiological studies that may enable advances in the aquaculture of sea cucumbers.

คณะผู้วิจัยได้ทำการศึกษาประสาทกายวิภาคศาสตร์เปรียบเทียบ ของระบบ M. rosenbergii neuropeptide F (MrNPF) ในระบบประสาทตลอดช่วงระยะการพัฒนาของรังไข่ของกุ้งก้ามกรามเพศเมีย ซึ่งพบว่า MrNPF transcripts มีความสัมพันธ์กับวงจรการพัฒนาของรังไข่ โดยระดับของ MrNPF จะค่อยๆเพิ่มจากระยะเริ่มต้น และมีระดับสูงสุดที่ระยะที่สามของวงจรการพัฒนาของไข่ และค่อยๆลดลงในระยะที่สี่ ในก้านตา สมอง ปมประสาท subesophageal และปมประสาท ในส่วนของจำนวน MrNPF-ir cells และความเข้มข้น ของ fluorescence intensity จะแปรผันตรงกับกัพัฒนาของรังไข่ ดังนั้นการศึกษานี้ได้รับองค์ความรู้พื้นฐานใหม่ที่สำคัญถึงระบบ MrNPF ในสมองส่วนกลาง ซึ่งเป็น neuropeptide ที่มีความควบคุมกับ sensory stimuli, รวมถึง photo-, chemo-, and gustatory receptions, ที่ควบคุมการกินอาหารและการสืบพันธุ์ ในกุ้งก้ามกรามเพศเมีย

ที่มาและความสำคัญ
There is still a lack of fundamental knowledge on the comparative neuroanatomical mapping and changes in the expression levels of MrNPF in the CNS during the ovarian cycle in the female prawn, M. rosenbergii. Therefore, in the present study, we investigated the comparative neuroanatomical distribution and alterations in the expression levels of MrNPF in the CNS of the female prawn, M. rosenbergii during the ovarian cycle, to provide new insights into MrNPF neural circuits, and MrNPF expression levels that could modulate the feeding process and ovarian development of this female prawn.

Abstract 
Neuropeptide F (NPF) plays critical roles in controlling the feeding and reproduction of prawns. In the present study, we investigated changes in the expression levels of Macrobrachium rosenbergii neuropeptide F (MrNPF), and its neuroanatomical distribution in eyestalk (ES), brain (BR), subesophageal ganglion (SEG), thoracic ganglia (TG), and abdominal ganglia (AG), during the ovarian cycle of female prawn. By qRT-PCR, the amount of MrNPF transcripts exhibited a gradual increase in the ES, BR, and combined SEG and TG from stages I and II, to reach a maximum level at stage III, and slightly declined at stage IV, respectively. The highest to lowest expression levels were detected in combined SEG and TG, BR, ES, and AG, respectively. MrNPF immunolabeling was observed in several neuronal clusters, associated fibers, and neuropils of these central nervous system (CNS) tissues. MrNPF-ir was more intense in neurons and neuropils of SEG and TG than those found in other parts of the CNS. The number of MrNPF-ir neurons and intensity of MrNPF-ir were higher in the ES, BR, SEG, and TG at the late stages than those at the early stages of the ovarian cycle, while those in AG exhibited insignificant change. Taken together, there is a correlation between changes in the neuroanatomical distribution of MrNPF and stages of the ovarian cycle, implying that MrNPF may be an important neuropeptide that integrates sensory stimuli, including photo-, chemo-, and gustatory receptions, to control feeding and reproduction, particularly ovarian development, of this female prawn, M. rosenbergii.

การศึกษาการออกฤทธิ์ และการแสดงออก ของ prostaglandin E2 ในปลิงทะเลขาว Holothuria scabra

ในการศึกษานี้ คณะผู้วิจัยได้ค้นพบและทำการศึกษาคุณลักษณะของ prostaglandin synthase (PGES) ในปลิงทะเลขาว H. scabra ชื่อว่า Hs-cPGES3 และได้ทำการศึกษาการแสดงออกของ Hs-cPGES3 mRNA expression levels พบว่า มีระดับสูงในอวัยวะสืบพันธุ์ระยะแรก (early stages) และค่อยๆลดลงในระยะสุดท้าย (late gonadal stages) กลุ่มที่ฉีดด้วย prostaglandin E2 (PGE2) กระตุ้นการพัฒนาของรังไข่ อัณฑะ และกระตุ้นการพัฒนาของเซลล์สืบพันธุ์ อย่างมีนัยสำคัญทางสถิติเมื่อเทียบกับกลุ่มควบคุม ดังนั้นผลการวิจัยที่นี้ได้รับองค์ความรู้พื้นฐานใหม่ที่สำคัญเกี่ยวกับบทบาทและการแสดงออกของ PGE2 และ PGES ในอวัยวะสืบพันธุ์ และอาจจะนำไปประยุกต์ใช้ในการช่วยเพิ่มการผลิตของปลิงทะเลขาวซึ่งเป็นสัตว์น้ำเศรษฐกิจที่สำคัญชนิดนี้ได้

ที่มาและความสำคัญ
The white sea cucumber, Holothuria scabra, is among the highest-priced economic marine species in the commercial fisheries of the tropical Indo-Pacific region, including Thailand (Hamel et al., 2001; Purcell et al., 2012). This animal has the high nutrient quality and richness in medicinal substances (Demeuldre and Eeckhaut, 2012). There is a high risk of overfishing this species which greatly affects the wild population. In the past few years, attempts have been made to produce this species by aquaculture. However, the breeding of H. scabra broodstocks in captivity is partially successful because their gonads fail to fully develop, and only a limited quantity of gametes can be obtained with a prolonged spawning period (Chiyamoon et al., 2020b; Nontunha et al., 2021). Therefore, the identification of some important hormonal factors, that regulate gonadal development and gametogenesis, is important and necessary for increasing the production of gametes for mass production of this sea cucumber species by aquaculture. However, important questions remain whether PGE2 plays a critical role in the reproduction of the sea cucumber H. scabra. Therefore, we characterized Hs-cPGES3, a key enzyme involved in PGE2 biosynthesis, and further examined the presence and distribution of PGES and PGE2 in the gonadal tissues. Functionally, we then investigated the roles of PGE2 in gonadal development and gametogenesis of H. scabra by using bioassays.

Abstract 
Prostaglandin E2 (PGE2) plays important roles in several physiological processes, including reproduction of several marine invertebrate species. The aims of the present study were to characterize the putative prostaglandin E synthase (PGES), an enzyme that is involved in the PGE2 biosynthetic pathway, in the sea cucumber, H. scabra. We then examined the presence and distribution of PGES and PGE2 in gonadal tissues using RT-qPCR and immunohistochemistry, and further investigated the effects of PGE2 on gonadal development and germ cell proliferation by using functional assays. Transcriptomic data and molecular cloning analyses from gonadal tissues of H. scabra were performed, and these tissues were found to have the PGES in H. scabra, namely Hs-cPGES3. By using RT-qPCR, the Hs-cPGES3 mRNA expression levels were higher at the early stages than the late stages of the gonadal cycle. In addition, the presence of PGE2 and its corresponding enzyme, Hs-cPGES3, was detected in the gonads with the highest intensities appearing in gamete cytoplasm during the early stages than those at the late stages. Functionally, PGE2, at the doses of 50 and 500 ng/g BW, significantly promoted gonadal development, gonado-somatic index (GSI), gonadal tube diameters, and increased germ cell proliferation, compared with those of the control animals (P < 0.05). Taken together, our findings are the first to demonstrate the presence of PGE2 and its biosynthetic-associated enzymes in the gonadal tissues, and its effects on gonadal development and germ cell proliferation in this sea cucumber. The knowledge gained would improve understanding of PGE2 mechanism in controlling gonadal development, which could be useful in enhancing the production of seeds for aquaculture of H. scabra.
