การศึกษาผลของการออกฤทธิ์ของกลูตาเมทและซีโรโทนิน ต่อการกินอาหาร แอคติวิตี้ของเอนไซม์การย่อยอาหาร และระบบภูมิคุ้มกัน ในกุ้งก้ามกรามเพศเมีย

ในการศึกษาครั้งนี้ คณะผู้วิจัยได้ทำการศึกษาการปรากฎของกลูตาเมท (glutamate, Glu) และซีโรโทนิน (serotonin, 5-HT) ในอวัยวะย่อยอาหาร และทำการศึกษาผลของการออกฤทธิ์ของกลูตาเมท และซีโรโทนิน ต่อการกินอาหาร แอคติวิตี้ของเอนไซม์การย่อยอาหาร และระบบภูมิคุ้มกัน ในกุ้งก้ามกรามเพศเมีย (Macrobrachium rosenbergii) คณะผู้วิจัยพบว่า Glu และ 5-HT มีการปรากฎใน foregut, midgut, hindgut, และ hepatopancreas และมีระดับความเข้มข้นสูงในระยะเต็มวัยของรังไข่ ในขณะที่สารสื่อประสาททั้งสองตัวนี้ พบระดับความเข้มข้นลดลง ในช่วงของรังไข่ระยะอ่อน โดยใช้วิธี high-performance liquid chromatography (HPLC) นอกจากนี้ ยังพบการปรากฏของ Glu-immunoreactivity (-ir) และ 5-HT-ir ในอวัยวะทางเดินอาหาร โดยใช้วิธี immunohistochemistry นอกจากนี้ กุ้งก้ามกรามที่ถูกฉีดด้วย Glu และ 5-HT ที่ระดับความเข้มข้น 2.5 × 10−6 และ 2.5 × 10−7 mol/prawn ส่งผลต่อ food consumption, gastric emptying midgut index, และเพิ่ม digestive enzymes อาทิ amylase, protease, และ lipase activities เมื่อเปรียบเทียบกับกลุ่มควบคุม อย่างมีนัยสำคัญทางสถิติ (P < 0.05). Glu และ 5-HT ยังสามารถกระตุ้น epithelial cell height, villi height, crypt depth และสามารถเพิ่มปฏิกิริยาของ PAS/AB ในอวัยวะทางเดินอาหาร นอกจากนี้ Glu และ 5-HT สามารถเพิ่มระดับของ M. rosenbergii NPF (MrNPF) และ immune-related transcripts อย่างมีนัยสำคัญทางสถิติ (P < 0.05) จากผลการวิจัยนี้ คณะผู้วิจัยได้ค้นพบองค์ความรู้พื้นฐานใหม่ที่สำคัญอย่างมาก ที่อาจจะสามารถนำสารสื่อประสาท Glu และ 5-HT มาประยุกต์ใช้เพื่อที่จะเพิ่มอัตราการกินอาหาร และการเพิ่มภูมิคุ้มกัน ในส่วนของการเพาะเลี้ยงกุ้งก้ามกรามเพศเมียในอนาคตต่อไปได้

ที่มาและความสำคัญ

Currently, prawn aquaculture with high-density populations faces several challenges, including diseases and infections, which cause stress and lead to diminished appetite and reduced feeding (Hooper et al., 2023). These factors directly impact gonadal development, particularly in the ovaries, leading to a decrease in the quantity and quality of eggs and, consequently, reduced offspring production. Feeding is affected by external factors such as water temperature, light, and food composition and taste properties (Kestemont and Baras, 2001; Volkoff and Rønnestad, 2020). In addition, feeding is regulated by internal factors, including various neurotransmitters, e.g., glutamate (Glu), serotonin (5-HT), and catecholamines (Scannell et al., 2008; Tierney, 2020). These factors are crucial in controlling feeding by activating the digestive and nervous systems, which is essential for assuring enough nutrient ingestion to maintain energy balance in the body (Celi et al., 2017). In female crustacean broodstocks, feeding is a crucial physiological process to acquire sufficient nutrients that are utilized to produce energy for ovarian development, oocyte production, and vitellogenesis. The quantity and quality of oocytes influence the successful development of fertilized eggs, which subsequently impacts crustacean production (Fernández-Palacios et al., 2011; Feng et al., 2023). Considering the importance of Glu and 5-HT in physiological functions, such as feeding, digestive enzyme activity, and immunity in aquatic animals like fish and decapod crustaceans, we question whether these neurotransmitters display alterations and regulatory functions in the digestive organs as essential modulators of feeding and immunity in female M. rosenbergii during the ovarian cycle.

Abstract

This study investigated the presence of glutamate (Glu) and serotonin (5-HT) in the digestive organs and their effects on feeding performance, digestive enzyme activity, and immunity throughout the ovarian cycle in female giant freshwater prawns (Macrobrachium rosenbergii). By high-performance liquid chromatography (HPLC), Glu and 5-HT were detected in the foregut, midgut, hindgut, and hepatopancreas, at the highest concentrations in the late ovarian stage and the lowest concentrations in the early ovarian stage. Through immunohistochemistry, the presence of Glu-immunoreactivity (-ir) and 5-HT-ir were observed in all digestive organs, and their intensities were shown to correlate with the levels detected by those observed via HPLC. Prawns injected with Glu and 5-HT at the doses of 2.5 × 10−6 and 2.5 × 10−7 mol/prawn, markedly increased food consumption, gastric emptying, and midgut index as well as enhanced amylase, protease, and lipase activities, compared with the control group (P < 0.05). Glu and 5-HT also enhanced epithelial cell height, villi height, and crypt depth and promoted the reactivity of PAS/AB in the digestive organs. In addition, treatment with Glu or 5-HT significantly upregulated M. rosenbergii NPF (MrNPF) and immune-related transcripts (P < 0.05). Collectively, these findings demonstrated the promising effects of Glu and 5-HT at a dose of 2.5 × 10−6 mol/prawn in stimulating feeding, digestive activities, and immunity, which could potentially be used to enhance feeding, growth, and maintain the health status of female prawn in aquaculture.

คณะผู้วิจัยได้ทำการศึกษาประสาทกายวิภาคศาสตร์เปรียบเทียบ ของระบบ M. rosenbergii neuropeptide F (MrNPF) ในระบบประสาทตลอดช่วงระยะการพัฒนาของรังไข่ของกุ้งก้ามกรามเพศเมีย ซึ่งพบว่า MrNPF transcripts มีความสัมพันธ์กับวงจรการพัฒนาของรังไข่ โดยระดับของ MrNPF จะค่อยๆเพิ่มจากระยะเริ่มต้น และมีระดับสูงสุดที่ระยะที่สามของวงจรการพัฒนาของไข่ และค่อยๆลดลงในระยะที่สี่ ในก้านตา สมอง ปมประสาท subesophageal และปมประสาท ในส่วนของจำนวน MrNPF-ir cells และความเข้มข้น ของ fluorescence intensity จะแปรผันตรงกับกัพัฒนาของรังไข่ ดังนั้นการศึกษานี้ได้รับองค์ความรู้พื้นฐานใหม่ที่สำคัญถึงระบบ MrNPF ในสมองส่วนกลาง ซึ่งเป็น neuropeptide ที่มีความควบคุมกับ sensory stimuli, รวมถึง photo-, chemo-, and gustatory receptions, ที่ควบคุมการกินอาหารและการสืบพันธุ์ ในกุ้งก้ามกรามเพศเมีย

ที่มาและความสำคัญ
There is still a lack of fundamental knowledge on the comparative neuroanatomical mapping and changes in the expression levels of MrNPF in the CNS during the ovarian cycle in the female prawn, M. rosenbergii. Therefore, in the present study, we investigated the comparative neuroanatomical distribution and alterations in the expression levels of MrNPF in the CNS of the female prawn, M. rosenbergii during the ovarian cycle, to provide new insights into MrNPF neural circuits, and MrNPF expression levels that could modulate the feeding process and ovarian development of this female prawn.

Abstract 
Neuropeptide F (NPF) plays critical roles in controlling the feeding and reproduction of prawns. In the present study, we investigated changes in the expression levels of Macrobrachium rosenbergii neuropeptide F (MrNPF), and its neuroanatomical distribution in eyestalk (ES), brain (BR), subesophageal ganglion (SEG), thoracic ganglia (TG), and abdominal ganglia (AG), during the ovarian cycle of female prawn. By qRT-PCR, the amount of MrNPF transcripts exhibited a gradual increase in the ES, BR, and combined SEG and TG from stages I and II, to reach a maximum level at stage III, and slightly declined at stage IV, respectively. The highest to lowest expression levels were detected in combined SEG and TG, BR, ES, and AG, respectively. MrNPF immunolabeling was observed in several neuronal clusters, associated fibers, and neuropils of these central nervous system (CNS) tissues. MrNPF-ir was more intense in neurons and neuropils of SEG and TG than those found in other parts of the CNS. The number of MrNPF-ir neurons and intensity of MrNPF-ir were higher in the ES, BR, SEG, and TG at the late stages than those at the early stages of the ovarian cycle, while those in AG exhibited insignificant change. Taken together, there is a correlation between changes in the neuroanatomical distribution of MrNPF and stages of the ovarian cycle, implying that MrNPF may be an important neuropeptide that integrates sensory stimuli, including photo-, chemo-, and gustatory receptions, to control feeding and reproduction, particularly ovarian development, of this female prawn, M. rosenbergii.
