ที่มาและความสำคัญ

ปลิงทะเลมีฤทธิ์ป้องกันโรคความเสื่อมของระบบประสาทและฟื้นฟูเซลล์ประสาทจากการถูกทำลาย โดยมีสารออกฤทธิ์ที่สำคัญคือ 2-Butoxytetrahydrofuran (2-BTHF) โดยสามารถลดการเกาะกลุ่มและความเป็นพิษของ amyloid-beta และ alpha-synuclein ในโรคความเสื่อมระบบประสาทได้ การศึกษาวิจัยนี้จึงได้ศึกษาการออกฤทธิ์ของ 2-BTHF ต่อโรคพาร์กินสันที่เกิดจากการทำลายเซลล์ประสาทโดปามีน

Abstract

Holothuria scabra (H. scabra), a marine organism traditionally known for its health benefits, has been utilized in both food and medicine. Our previous studies indicated that 2-butoxytetrahydrofuran (2-BTHF), which is isolated from H. scabra, possesses the potential to alleviate amyloid-β and α-synuclein accumulations associated with Alzheimer’s and Parkinson’s diseases (AD and PD), respectively. However, the mechanisms through which 2-BTHF mitigates PD-related neurotoxicity remain unclear. In this study, we investigated the effects of 2-BTHF on a 6-hydroxydopamine (6-OHDA)-induced Caenorhabditis elegans (C. elegans) model. Our results demonstrated that 2-BTHF recovered dopaminergic (DAergic) neurons from degeneration and restored dopamine-related behaviors. Furthermore, 2-BTHF reduced reactive oxygen species (ROS) production, preserved mitochondrial fluorescence, and decreased both mitochondrial and cytoplasmic unfolded protein responses (UPRmt and UPRcyto) activation. Transcriptome sequencing analysis revealed the critical roles of various systems, including the immune system, nervous system, glutathione (GSH) metabolism, xenobiotics, terpenoids, energy metabolism, cell growth and death, and aging-related longevity pathways. Additionally, 2-BTHF showed potential interactions with stress resistance and detoxification transcription factors, promoting the nuclear translocation of DAF-16 and SKN-1, which in turn activated their targets, including SOD-3, CTL-2, GCS-1, and GST-4. Moreover, 2-BTHF increased total GSH levels and reduced the ced-3-related cascade. This study demonstrates that 2-BTHF holds promise as a therapeutic agent for treating 6-OHDA-induced DAergic neurodegeneration in the C. elegans model.

ในการศึกษาครั้งนี้ คณะผู้วิจัยได้ทำการตรวจสอบการปรากฏและการกระจายของ GABA และ DA ในระยะตัวอ่อนขั้นต่างๆของ H. scabra โดยได้เน้นศึกษาเป็นพิเศษที่อวัยวะที่ช่วยในการลงเกาะ (settlement organs, SOs) ซึ่งพบว่า มีการปรากฏ และการกระจายแบบจำเพาะของเซลล์และเส้นใย GABA และ DA ในโครงสร้าง SO หลายโครงสร้าง รวมถึง ซีเลียรีแบนด์ (ciliary bands, CiBs), เทนทาเคิล (tentacle, Ts) และโพเดีย (PDs) ของตัวอ่อน H. scabra นอกจากนี้ คณะผู้วิจัยยังพบว่า จำนวนเซลล์และความเข้มของ GABA-ir และ DA-ir-positive ใน CiBs, Ts และ PDs ในระหว่างขั้นตอนการพัฒนาของตัวอ่อนในระยะต่างๆนั้น มีการเปลี่ยนแปลงที่จำเพาะไปตามระยะการพัฒนาของตัวอ่อน ในการศึกษานี้ คณะผู้วิจัยเป็นกลุ่มแรกแรกที่ได้ศึกษาเกี่ยวกับการปรากฏ ของระบบ GABAergic และ dopaminergic ในโครงสร้างที่เกี่ยวข้องกับการลงเกาะของปลิงทะเล H. scabra ซึ่งการค้นพบนี้ได้ให้ข้อมูลบ่งชี้ที่สำคัญที่บ่งชี้ถึงบทบาทและกลไกที่สำคัญของสารสื่อประสาททั้งสองนี้ในการควบคุมการลงเกาะของปลิงทะเล H. scabra

In the sea cucumber, Holothuria scabra, the competent larvae require main settlement organs (SOs), including the ciliary bands, tentacles, and podia, as well as cues from major neurotransmitters, including gamma-aminobutyric acid (GABA) and dopamine (DA), for successful settlement. However, the presence and differential expression of DA during the developmental stages in H. scabra, have not yet been investigated. Additionally, the exact location for the production and secretion of these two neurotransmitters in the SO needs to be characterized so that this important knowledge may help increase the production of this sea cucumber species. In the present study, we investigated and mapped the presence and distribution of GABA and DA in several organs that may be involved in mediating the settlement of H. scabra larvae, with special emphasis on SOs, during different developmental stages. Our findings shed important light on the anatomical localization and potential implications of the two neurotransmitter systems in SOs to control the larval settlement process in this sea cucumber species.

Abstract

In the sea cucumber, Holothuria scabra, the competent larvae require main settlement organs (SOs), including the ciliary bands (CiBs), tentacles (Ts), podia (PDs), and cues from neurotransmitters, including gamma-aminobutyric acid (GABA) and dopamine (DA), for successful settlement. In the present study, we investigated the spatial distribution of GABA and DA in the developmental stages of H. scabra, with special emphasis on SOs by detecting immunoreactivity (-ir) against these two neurotransmitters. Strong GABA-ir and DA-ir cells and fibers were specifically detected in several SO structures, including CiBs, CiB cells (CiBCs), and long cilia (LCi), of H. scabra larvae. Additionally, we found intense GABA-ir and DA-ir cells in the epithelial lining of bud-papillae (BP) and mesothelium (Me) in the stem (S) region of Ts in larvae and juveniles. Intense GABA-ir and DA-ir were observed in the epineural nerve plexus (ENP) and hyponeural nerve plexus (HNP) of Ts in H. scabra pentactula and juvenile stages. Staining for these two neurotransmitters was particularly intense in the PDs and their nerve fibers. We also found significant changes in the numbers of GABA-ir and DA-ir-positive cells and intensities in the CiBs, Ts, and PDs during the developmental stages. Taken together, we are the first to report on the existence and distribution of GABAergic and dopaminergic systems in structures associated with the settlement. Our findings provide new and important insights into the possible functions of these two neurotransmitters in regulating the settlement of this sea cucumber species.

Abstract

Extracts from a sea cucumber, Holothuria scabra, have been shown to exhibit various pharmacological properties including anti-oxidation, anti-aging, anti-cancer, and anti-neurodegeneration. Furthermore, certain purified compounds from H. scabra displayed neuroprotective effects against Parkinson’s and Alzheimer’s diseases. Therefore, in the present study, we further examined the anti-aging activity of purified H. scabra compounds in a Caenorhabditis elegans model. Five compounds were isolated from ethyl acetate and butanol fractions of the body wall of H. scabra and characterized as diterpene glycosides (holothuria A and B), palmitic acid, bis (2-ethylhexyl) phthalate (DEHP), and 2-butoxytetrahydrofuran (2-BTHF). Longevity assays revealed that 2-BTHF and palmitic acid could significantly extend lifespan of wild type C. elegans. Moreover, 2-BTHF and palmitic acid were able to enhance resistance to paraquat-induced oxidative stress and thermal stress. By testing the compounds’ effects on longevity pathways, it was shown that 2-BTHF and palmitic acid could not extend lifespans of daf-16age-1sir-2.1jnk-1, and skn-1 mutant worms, indicating that these compounds exerted their actions through these genes in extending the lifespan of C. elegans. These compounds induced DAF-16::GFP nuclear translocation and upregulated the expressions of daf-16hsp-16.2sod-3 mRNA and SOD-3::GFP. Moreover, they also elevated protein and mRNA expressions of GST-4, which is a downstream target of the SKN-1 transcription factor. Taken together, the study demonstrated the anti-aging activities of 2-BTHF and palmitic acid from H. scabra were mediated via DAF-16/FOXO insulin/IGF and SKN-1/NRF2 signaling pathways.

ที่มาและความสำคัญ
The white sea cucumber, Holothuria scabra, is among the highest-priced economic marine species in the commercial fisheries of the tropical Indo-Pacific region, including Thailand (Hamel et al., 2001; Purcell et al., 2012). This animal has the high nutrient quality and richness in medicinal substances (Demeuldre and Eeckhaut, 2012). There is a high risk of overfishing this species which greatly affects the wild population. In the past few years, attempts have been made to produce this species by aquaculture. However, the breeding of H. scabra broodstocks in captivity is partially successful because their gonads fail to fully develop, and only a limited quantity of gametes can be obtained with a prolonged spawning period (Chiyamoon et al., 2020b; Nontunha et al., 2021). Therefore, the identification of some important hormonal factors, that regulate gonadal development and gametogenesis, is important and necessary for increasing the production of gametes for mass production of this sea cucumber species by aquaculture. However, important questions remain whether PGE2 plays a critical role in the reproduction of the sea cucumber H. scabra. Therefore, we characterized Hs-cPGES3, a key enzyme involved in PGE2 biosynthesis, and further examined the presence and distribution of PGES and PGE2 in the gonadal tissues. Functionally, we then investigated the roles of PGE2 in gonadal development and gametogenesis of H. scabra by using bioassays.

Abstract 
Prostaglandin E2 (PGE2) plays important roles in several physiological processes, including reproduction of several marine invertebrate species. The aims of the present study were to characterize the putative prostaglandin E synthase (PGES), an enzyme that is involved in the PGE2 biosynthetic pathway, in the sea cucumber, H. scabra. We then examined the presence and distribution of PGES and PGE2 in gonadal tissues using RT-qPCR and immunohistochemistry, and further investigated the effects of PGE2 on gonadal development and germ cell proliferation by using functional assays. Transcriptomic data and molecular cloning analyses from gonadal tissues of H. scabra were performed, and these tissues were found to have the PGES in H. scabra, namely Hs-cPGES3. By using RT-qPCR, the Hs-cPGES3 mRNA expression levels were higher at the early stages than the late stages of the gonadal cycle. In addition, the presence of PGE2 and its corresponding enzyme, Hs-cPGES3, was detected in the gonads with the highest intensities appearing in gamete cytoplasm during the early stages than those at the late stages. Functionally, PGE2, at the doses of 50 and 500 ng/g BW, significantly promoted gonadal development, gonado-somatic index (GSI), gonadal tube diameters, and increased germ cell proliferation, compared with those of the control animals (P < 0.05). Taken together, our findings are the first to demonstrate the presence of PGE2 and its biosynthetic-associated enzymes in the gonadal tissues, and its effects on gonadal development and germ cell proliferation in this sea cucumber. The knowledge gained would improve understanding of PGE2 mechanism in controlling gonadal development, which could be useful in enhancing the production of seeds for aquaculture of H. scabra.

ปลิงทะเลขาวมีคุณค่าทางอาหารและมีประโยชน์ต่อสุขภาพมายาวนาน โดยผลงานวิจัยเมื่อเร็วๆ นี้พบว่าสารสกัดหยาบของปลิงทะเลขาวมีฤทธิ์ป้องกันเซลล์ประสาทในโรคพาร์กินสันได้ ดังนั้นการศึกษาวิจัยนี้จึงได้ทำการสกัดแยกสาร diterpene glycosides จากปลิงทะเลขาวและนำมาทดสอบศักยภาพในการลดการสะสมของโปรตีน a-synuclein และป้องกันเซลล์ประสาทจากการถูกทำลายด้วยโปรตีน a-synuclein ที่สะสมมากเกินไป ซึ่งเป็นสาเหตุหลักในการเกิดโรคพาร์กินสัน โดยใช้ C. elegans เป็นสัตว์ทดลองต้นแบบ ผลการศึกษาพบว่าสาร diterpene glycosides จากปลิงทะเลขาวสามารถลดการสะสมของโปรตีน a-synuclein เพิ่มอัตราการเคลื่อนไหวและอายุขัยของสัตว์ทดลองที่สูญเสียไปจากโปรตีน a-synuclein ที่สะสม นอกจากนี้สาร diterpene glycosides ยังสามารถป้องกันเซลล์ประสาทสร้างโดปามีนจากพิษที่เกิดจากโปรตีน a-synuclein ที่สะสมมากเกินไป และฟื้นฟูพฤติกรรมของสัตว์ทดลองจากระดับโดปามีนที่เพิ่มสูงขึ้นหลังจากได้รับสาร diterpene glycosides ผลการวิจัยเชิงลึกยังพบว่าสาร diterpene glycosides สามารถเพิ่มการแสดงออกของยีนที่เกี่ยวข้องกับกลไก autophagy เช่น beclin-1/bec-1, lc-3/lgg-1 และ atg-7/atg-7 ซึ่งเมื่อยืนยันด้วยการศึกษาโดย RNA interference พบว่าทั้ง lc-3/lgg-1 และ atg-7/atg-7 เกี่ยวข้องกับการสลายโปรตีน a-synuclein ที่สะสมและป้องกันเซลล์ประสาทสร้างโดปามีนจากการถูกทำลายด้วยโปรตีน a-synuclein ที่สะสมมากเกินไป

Abstract

Ethnopharmacological relevance

Holothuria (Metriatyla) scabra Jaeger (H. scabra), sea cucumber, is the marine organism that has been used as traditional food and medicine to gain the health benefits since ancient time. Although our recent studies have shown that crude extracts from H. scabra exhibited neuroprotective effects against Parkinson’s disease (PD), the underlying mechanisms and bioactive compounds are still unknown.

Aim of the study

In the present study, we examined the efficacy of purified compounds from H. scabra and their underlying mechanism on α-synuclein degradation and neuroprotection against α-synuclein-mediated neurodegeneration in a transgenic Caenorhabditis elegans PD model.

Material and methods

The H. scabra compounds (HSEA-P1 and P2) were purified and examined for their toxicity and optimal dose-range by food-clearance and lifespan assays. The α-synuclein degradation and neuroprotection against α-synuclein-mediated neurodegeneration were determined using transgenic C. elegans model, Punc-54::α-syn and Pdat-1:: α-syn; Pdat-1::GFP, respectively, and then further investigated by determining the behavioral assays including locomotion rate, basal slowing rate, ethanol avoidance, and area-restricted searching.

The underlying mechanisms related to autophagy were clarified by quantitative PCR and RNAi experiments.

Results

Our results showed that HSEA-P1 and HSEA-P2 significantly diminished α-synuclein accumulation, improved motility deficits, and recovered the shortened lifespan. Moreover, HSEA-P1 and HSEA-P2 significantly protected dopaminergic neurons from α-synuclein toxicity and alleviated dopamine-associated behavioral deficits, i.e., basal slowing, ethanol avoidance, and area-restricted searching. HSEA-P1 and HSEA-P2 also up-regulated autophagy-related genes, including beclin-1/bec-1, lc-3/lgg-1, and atg-7/atg-7. RNA interference (RNAi) of these genes in transgenic α-synuclein worms confirmed that lc-3/lgg-1 and atg-7/atg-7 were required for α-synuclein degradation and DAergic neuroprotection activities of HSEA-P1 and HSEA-P2. NMR and mass spectrometry analysis revealed that the HSEA-P1 and HSEA-P2 contained diterpene glycosides.

Conclusion

These findings indicate that diterpene glycosides extracted from H. scabra decreases α-synuclein accumulation and protects α-synuclein-mediated DAergic neuronal loss and its toxicities via lgg-1 and atg-7.

ที่มาและความสำคัญ
The sea cucumber, Holothuria scabra, is a high-value economic animal in Thailand for local consumption and exportation. One major drawback in aquaculture of this species is that their larvae have low settlement and survival rates during the early developmental period. Previous reports have shown that several potential inducers, including neurotransmitters (e.g., GABA, dopamine), have potential role to promote larval settlement in marine invertebrates. Until now, the investigation of roles and possible mechanisms of these neurotransmitters on larval settlement in H. scabra has not yet been investigated. Therefore, important knowledge gained here provides regarding the role and possible mechanisms of these neurotransmitters in regulating the larval settlement, and they could further be used to enhance high settlement, metamorphosis, and survival rates, which will have high impact on aquaculture of this sea cucumber species.

Abstract 
The sea cucumber, Holothuria scabra, is a high-value economic animal in Thailand for local consumption and exportation. One major drawback in aquaculture of this species is that their larvae have low settlement and survival rates during the early developmental period. In the present study, we investigated the effects of various neurotransmitters, including GABA, dopamine (DA), L-DOPA, and monosodium glutamate (MSG) on larval settlement in both laboratory and practical models. In laboratory models, treatments of doliolaria with GABA or MSG, at a dose of 10−1 mM for 72 h could induce larval settlement at about 90% and 93%, respectively, compared with the control group (about 20%) (P < 0.05), while treatments with DA or L-DOPA at a dose of 10−2 mM for 12 h, were capable of inducing 100% settlement, compared with the control group (P < 0.05). In practical models, at day 37 after treatment, GABA or MSG significantly induced larvae to settle and survive at approximately 6% and 4%, respectively, while L-DOPA induced at about 3%, compared with the control group (about 0.2%) (P < 0.05). At day 37, all neurotransmitters-treated groups exhibited faster growth rates, while at 51 days, the growth rates of groups treated with DA, L-DOPA and MSG were faster, than the control group. GABA- and DA-treated groups also attained juvenile features earlier than MSG and L-DOPA-treated groups, and control groups. Taken together, this study shows significant effects of neurotransmitters on inducing settlement, increased metamorphosis and survival rates of the larvae of this sea cucumber, which could potentially be applied to enhance the production of this sea cucumber species.

ที่มาและความสำคัญ
The gonadal development and production of seeds remain the main bottlenecks in aquaculture of the sea cucumber, Holothuria scabra. Hence, investigation and application of certain neurohormonal factors could reduce these problems. The TRH was first identified in the sea urchin, S. purpuratus, and it was believed to be related to the GnRH-type peptide. However, there is a current lack of important knowledge regarding this neuropeptide in H. scabra. This led us to hypothesize that TRH/GnRH neuropeptides may exist in this sea cucumber species, and that TRH/GnRH-like peptide may be involved in controlling reproduction, particularly ovarian development and oocyte maturation of H. scabra resembling GnRH functions. The basic knowledge obtained here contributes important new insights into the evolution and possible mechanisms of TRH/GnRH neuropeptide system in H. scabra, and these neuropeptides could be applied for promoting the seed and larval productivity in sea cucumber aquaculture.

Abstract 
Thyrotropin/Gonadotropin-releasing hormone (TRH/GnRH) is a neurohormone that plays important roles in regulation of several physiological processes of invertebrate species, including echinoderms. The aims of this study were to characterize for the first time a TRH/GnRH-like peptide in the sea cucumber, Holothuria scabra. We have isolated a full-length cDNA for a TRH/GnRH preprohormone gene from the radial nerve cord of H. scabra. We found that H. scabra (hs)TRH/GnRH precursor comprised two types of peptides, namely, the short TRH/GnRH-like peptides and the long TRH/GnRH-like peptides. The expression of hsTRH/GnRH gene, was intense in radial nerve cord, nerve ring and mature ovary by using RT-PCR. By in situ hybridization the transcripts of hsTRH/GnRH was detected at a high level within the late stages (oocyte stages 3 and 4). In the functional assays, injections of the short TRH/GnRH-like peptides could significantly accelerate gonadal development as reflected by increased gonadal-somatic index (GSI), and stimulated gametogenesis, as reflected by significantly increased oocyte proliferation. Taken together, our new discovery of the TRH/GnRH-like peptides in H. scabra contributes important new insights into the evolution of TRH/GnRH neuropeptide system in this sea cucumber species, and these neuropeptides could be potentially applied in the sea cucumber aquaculture.

Abstract 
Sea cucumber, Holothuria scabra, is a well-known traditional Asian medicine that has been used for suppressing inflammation, promoting wound healing, and improving immunity. Moreover, previous studies demonstrated that the extract from H. scabra contains many bioactive compounds and has potent on inhibiting tumor cells survival and progression. However, the effect of the methanolic extract from the body wall of H. scabra (BWMT) on human prostate cancer cells has not yet been investigated. In this study, we aimed to investigate the effects and underlying mechanism of BWMT on prostate cancer cell viability and metastasis. BWMT was obtained by maceration with methanol. The effect of BWMT on cell viability was assessed by MTT and colony formation assays. The intracellular ROS accumulation was evaluated using a DCFH-DA fluorescence probe. Hoechst 33342 staining and Annexin V-FITC/PI staining were used to examine the apoptotic-inducing effect of the extract. A transwell migration assay was performed to determine the anti-metastasis effect. BWMT significantly reduced cell viability, triggered cellular apoptosis by accumulating intracellular ROS and resulting in the upregulation of JNK and p38 signaling pathways. In addition, BWMT also inhibited the invasion of PC3 cells by downregulating MMP-2/-9 expression via the ERK pathway. Consequently, our study provides BWMT from H. scabra as a putative therapeutic agent that could be applicable for prostate cancer progression.

Abstract

Introduction
Cancer cells utilize the modified glucose metabolism known as Warburg effect, with lactate production as the end product. In the search for alternative therapy, the body wall of sea cucumbers contains various substances with pharmacological activities. Herein, we investigate the effect of Holothuria scabra extract on the viability and Warburg effect of aggressive breast cancer cells.

Methods
Body wall of H. scabra was extracted using 95% ethanol. Triple-negative breast cancer cells, MDA-MB-231, were treated with the extract at various concentrations under normoglycemic and hyperglycemic conditions. Cytotoxicity test was performed using MTT assay. Apoptotic proteins were quantified using Western blot. Apoptotic cells were stained with Hoechst 33342. Lactate production was determined using L-lactate assay kit.

Results
By MTT assay, H. scabra extract suppressed the viability of breast cancer cells in a dose-dependent and time-dependent manner by enhancing apoptosis, indicated by a marked increase of proapoptotic Bax and pro-caspase three expressions, and decreased expression of anti-apoptotic Bcl-2. The extract could reduce hexokinase II expression, leading to reduced lactate production by blocking the Akt/mTOR/HIF-1 axis.

Discussion
Overall findings indicated that H. scabra extract could be a possible therapeutic against breast cancer progression in patients with hyperglycemia, for instance, diabetes mellitus.

สารสกัดหยาบปลิงทะเลขาว Holothuria scabra (HS) มีคุณสมบัติที่ต้านการอักเสบ ต้านอนุมูลอิสระ และต้านมะเร็ง รวมทั้งมีศักยภาพในการป้องกันการเสื่อมของเซลล์ประสาทที่พบในหนอน Caenorhabditis elegans ที่เป็นโรคพาร์กินสัน การศึกษาวิจัยนี้จึงมุ่งเน้นศึกษาประโยชน์ของสารสกัดปลิงทะเลขาว HS ที่มีคุณสมบัติในการป้องกันและซ่อมแซมเซลล์ประสาทโดปามีนในเซลล์ทดลองและหนู พบว่าการรักษาด้วยวิธีก่อนและหลังให้สารสกัดนี้ในหนูทดลองที่ถูกชักนำให้เกิดโรคพาร์กินสันด้วย 1- methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) ทำให้หนูมีการเคลื่อนไหวแบบ grid walk ดีขึ้น ร่วมกับการเพิ่มจำนวนเซลล์ประสาทโดปามีนและเส้นใยประสาทใน substantia nigra pars compacta และ striatum ส่วนในระดับเซลล์พบว่าสารสกัดนี้ลดการตายแบบ apoptosis ในเซลล์ประสาทที่มีลักษณะคล้ายเซลล์โดปามีนที่ถูกชักนำให้เกิดโรคพาร์กินสันด้วยสาร 1-methyl- 4-phenylpyridinium (MPP+) โดยการเพิ่มการแสดงออกของ Bcl-2, ลดการแสดงออกของ cleaved caspase 3 และป้องกันการเกิด depolarization ของผนังไมโตคอนเดรีย รวมทั้งยังเพิ่มการแสดงออกของ tyrosine hydroxylase และ ยับยั้งการสร้าง α-synuclein protein

Abstract
Extracts from Holothuria scabra (HS) have been shown to possess anti-inflammation, anti-oxidant and anti-cancer activities. More recently, it was shown to have neuroprotective potential in Caenorhabditis elegans PD model. Here, we assessed whether HS has neuroprotective and neurorestorative effects on dopaminergic neurons in both mouse and cellular models of PD. We found that both pre-treatment and post-treatment with HS improved motor deficits in PD mouse model induced with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) as determined by grid walk test. This was likely mediated by HS protective and restorative effects on maintaining the numbers of dopaminergic neurons and fibers in both substantia nigra pars compacta (SNpc) and striatum. In a cellular model of PD, HS significantly attenuated 1-methyl-4-phenylpyridinium (MPP+)-induced apoptosis of DAergic-like neurons differentiated from SH-SY5Y cells by enhancing the expression of Bcl-2, suppressing the expression of cleaved Caspase 3 and preventing depolarization of mitochondrial membrane. In addition, HS could stimulate the expression of tyrosine hydroxylase (TH) and suppressed the formation of α-synuclein protein. Taken together, our in vivo and in vitro findings suggested that HS is an attractive candidate for the neuroprotection rather than neurorestoration in PD.
