โรคพาร์กินสัน (Parkinson’s disease: PD) เป็นโรคทางระบบประสาทที่เสื่อมเรื้อรัง มีลักษณะเด่นคือการสูญเสียเซลล์ประสาทโดพามิเนอร์จิกในสมองส่วน substantia nigra pars compacta ซึ่งนำไปสู่การเคลื่อนไหวช้า กล้ามเนื้อแข็งเกร็ง และอาการสั่น สาเหตุของโรคมีความซับซ้อนและเกี่ยวข้องกับหลายปัจจัย โดยเฉพาะความเครียดออกซิเดชัน (oxidative stress) จากการสะสมของอนุมูลอิสระ (ROS) ซึ่งเป็นตัวการสำคัญที่เร่งการเสื่อมของเซลล์ประสาท กลไกสำคัญในการป้องกันความเสียหายจากอนุมูลอิสระในร่างกายคือ เส้นทางสัญญาณ Nrf2 ซึ่งทำหน้าที่กระตุ้นการแสดงออกของยีนที่เกี่ยวข้องกับการต้านอนุมูลอิสระ เช่น SOD1, GST, GPX, HMOX1 และ NQO1 โดยปกติ Nrf2 จะถูกยับยั้งในไซโตพลาสซึม แต่เมื่อเกิดภาวะเครียดออกซิเดชัน Nrf2 จะถูกปลดปล่อยและเคลื่อนเข้าสู่นิวเคลียสเพื่อกระตุ้นยีนเหล่านี้ เวย์โปรตีนไอโซเลต (Whey Protein Isolate: WPI) เป็นโปรตีนบริสุทธิ์สูงจากนมวัว มีคุณสมบัติโดดเด่นในด้านความสามารถในการละลายน้ำ ย่อยง่าย และมีกรดอะมิโนจำเป็นครบถ้วน หลายการศึกษารายงานว่า WPI และเปปไทด์ที่ได้จากการย่อยเวย์มีฤทธิ์ต้านอนุมูลอิสระ ช่วยเสริมการสร้างกลูตาไธโอน และควบคุมสภาวะรีดอกซ์ในเซลล์ รวมถึงอาจลดการอักเสบในระบบต่าง ๆ ของร่างกาย การศึกษานี้มีความสำคัญ เนื่องจากมุ่งประเมินศักยภาพของ WPI ในการปกป้องเซลล์ประสาทจากความเสียหายที่เกิดจาก MPP⁺ ผ่านกลไกการลดระดับ ROS, กระตุ้นการเข้าสู่นิวเคลียสของ Nrf2 และการกระตุ้นยีนต้านอนุมูลอิสระ เพื่อให้เข้าใจถึงกลไกการออกฤทธิ์ของ WPI ในการป้องกันโรคพาร์กินสันระดับเซลล์ ซึ่งอาจเป็นแนวทางใหม่ในการพัฒนาอาหารฟังก์ชันหรือผลิตภัณฑ์เสริมอาหารสำหรับผู้ป่วยโรคพาร์กินสันในอนาคต

Abstract

The pathogenesis of Parkinson’s disease (PD) consists of the apoptosis of dopaminergic neurons in the substantia nigra pars compacta (SNpc) due to oxidative stress. The present study aimed to evaluate the potential antioxidant activity of whey protein isolate (WPI) in PD models, using neurotoxin-exposed SH-SY5Y cells differentiated into dopaminergic-like neurons. Our research shows that WPI’s high glutamic acid, aspartic acid, and leucine contribute to its antioxidant and neuroprotective effects, with glutamic acid crucial for glutathione synthesis. In vitro studies found that WPI, at concentrations of 5–1000 µg/mL, is non-toxic to differentiated SH-SY5Y cells. Notably, the lowest con-centration of WPI (5 µg/mL) significantly decreased intracellular reactive oxygen species (ROS) levels in these cells following a 24 h co-treatment with 1-methyl-4-phenylpyridinium (MPP+). The antioxidant effects of WPI were also confirmed by the increased expression of HO1 and GPx antioxidant enzymes, which are Nrf2 pathway target genes and were evaluated by real-time PCR. Furthermore, Nrf2 nuclear translocation in the differentiated SH-SY5Y cells was also increased when the cells were exposed to 5 µg/mL of WPI with MPP+. These results together suggest that WPI has antioxidant effects on dopaminergic-like neurons in a Parkinson’s disease model.

พยาธิสรีรวิทยาของโรคพาร์กินสันเชื่อมโยงกับความเครียดจากปฏิกิริยาออกซิเดชั่นและการอักเสบในสมองจนกระทั่งทำให้เกิดการตายของเซลล์ประสาทชนิดโดปามิเนอร์จิคในสมองส่วน substantia nigra pars compacta (SNpc) การรักษาโรคพาร์กินสันในปัจจุบันคือการใช้ยาบรรเทาอาการ เช่น ยาเลโวโดปา แต่ยาหลายๆชนิดนี้ยังไม่สามารถรักษาโรคนี้ให้หายขาดได้ นอกจากนี้การใช้ยาในระยะยาวอาจนำไปสู่ผลข้างเคียงได้ ดังนั้นการวิจัยในปัจจุบันจึงเน้นที่จะใช้ผลิตภัณฑ์จากธรรมชาติมาใช้เป็นทางเลือกในการบรรเทาอาการหรือรักษาโรคพาร์กินสันหรือโรคทางระบบประสาทอื่นๆ เนื่องจากอาจจะไม่ก่อให้เกิดผลข้างเคียงหรือเกิดผลข้างเคียงได้น้อย ในระยะหลังๆนี้มีงานวิจัยหลายชิ้นรายงานว่า โปรตีนไฮโดรไลเสตจากปลามีสารต้านอนุมูลอิสระ และคุณสมบัติต่อต้านการอักเสบและลดตายของเซลล์หลายชนิด ซึ่งรวมถึงเซลล์ประสาท ดังนั้นการศึกษานี้มีวัตถุประสงค์เพื่อตรวจสอบฤทธิ์ต้านอนุมูลอิสระและฤทธิ์ต้านการอักเสบของเลือดปาทูน่าเลือดไฮโดรไลเซท (TBH) ในการบรรเทาการตายของเซลล์ประสาทนิวโรบลาสโตมาของมนุษย์ (SH-SY5Y) ที่ได้รับการกระตุ้นการเปลี่ยนแปลงไปเป็นเซลล์ประสาทที่คล้ายเซลล์ประสาทชนิดโดปามิเนอร์จิก (differentiated SH-SY5Y) ในแง่ของการลดการตายของเซลล์ โดยลดอนุมูลอิสระและลดการอักเสบภายในเซลล์

Abstract

The pathophysiology of Parkinson’s disease (PD) is linked to oxidative stress and inflammatory-induced cell death of dopaminergic neurons in substantia nigra pars compacta (SNpc). The current study aims to investigate the effects of tuna blood hydrolysate (TBH) on the neurodegeneration of Parkinson-like disease models using neurotoxin-induced SH-SY5Y cells differentiated to be dopaminergic-like neurons. It was found that TBH at concentrations ranging from 5 to 250 µg/ml can reduce differentiated SH-SY5Y cell death after exposure to 1-methyl-4-phenylpyridinium (MPP+) co-treated with TBH for 24 h. Real-time PCR results indicated that TBH enhanced the expressions of NAD(P)H quinone dehydrogenase 1 (NQO1), catalase (CAT), and thioredoxin reductase (TrxR1), which are Nrf2 downstream antioxidant genes. Bioinformatic analysis and molecular docking revealed that the peptide found in TBH, IPGQPGLPGPPGPPGPPGLG, could penetrate the cell and blood–brain barrier and strongly bind the Keap1-Kelch domain, causing Nrf2 nuclear translocation. Consequently, the upregulation of antioxidant genes ameliorates ROS and reduces dopaminergic neuron death. Another, TBH can reduce differentiated SH-SY5Y cell death and intracellular ROS after exposure to 24 h of co-treatment between TBH and tumor necrosis factor-alpha (TNF-α). Western blot analysis was performed to investigate apoptosis-related proteins. Bcl2 and BDNF are dramatically increased, while BAX, cytochrome c, caspase3, caspase9, and cleaved caspase9 are significantly decreased in co-treating differentiated SH-SY5Y cells with TBH and TNF-α. These findings demonstrated that TBH can suppress the apoptosis of inflammatory differentiated SH-SY5Y cells. Taken together, this study suggests that TBH can alleviate neurodegeneration and has anti-inflammatory effects on dopaminergic-like neurons in the simulation of a PD condition.

โรคพาร์กินสัน เกี่ยวข้องกับการสูญเสียเซลล์โดปามีนและการรวมกันของ alpha-synuclein ซึ่งมีสาเหตุมาจากการสร้างสารอนุมูลอิสระที่มากเกินไปส่งผลให้เกิดการทำงานที่ผิดปกติของไมโตคอนเดรียและกระบวนการออโตฟาจี (autophagy) ตามลำดับ ในปัจจุบันได้มีการศึกษาทางเภสัชวิทยาของสาร Andrographolide หรือ Andro เป็นจำนวนมาก เช่น ฤทธิ์การต้านเบาหวาน ต้านมะเร็ง ต้านการอักเสบ เป็นต้น อย่างไรก็ตามงานวิจัยที่ศึกษาฤทธิ์ของสาร Andro ในการป้องกันสารพิษต่อระบบประสาท (neurotoxin) MMP+ ที่ชักนำในเซลล์ประสาทสายพันธุ์นิวโรบลาสโตมา SH-SY5Y ให้เป็นโรคพาร์กินสันยังไม่พบ ดังนั้นในการศึกษานี้ เราจึงได้ตั้งสมมติฐานว่า สาร Andro น่าจะมีฤทธิ์ป้องกันระบบประสาทต่อสาร MMP+ ที่ชักนำให้เกิดการตายแบบ apoptosis โดยผ่านทางกระบวน mitophagy เพื่อกำจัดไมโตคอนเดรียที่มีการทำงานที่ผิดปกติและการต้านสารอนุมูลอิสระ

Abstract

Parkinson’s disease (PD) is associated with dopaminergic neuron loss and alpha-synuclein aggregation caused by ROS overproduction, leading to mitochondrial dysfunction and autophagy impairment. Recently, andrographolide (Andro) has been extensively studied for various pharmacological properties, such as anti-diabetic, anti-cancer, anti-inflammatory, and anti-atherosclerosis. However, its potential neuroprotective effects on neurotoxin MPP+-induced SH-SY5Y cells, a cellular PD model, remain uninvestigated. In this study, we hypothesized that Andro has neuroprotective effects against MPP+-induced apoptosis, which may be mediated through the clearance of dysfunctional mitochondria by mitophagy and ROS by antioxidant activities. Herein, Andro pretreatment could attenuate MPP+-induced neuronal cell death that was reflected by reducing mitochondrial membrane potential (MMP) depolarization, alpha-synuclein, and pro-apoptotic proteins expressions. Concomitantly, Andro attenuated MPP+-induced oxidative stress through mitophagy, as indicated by increasing colocalization of MitoTracker Red with LC3, upregulations of the PINK1–Parkin pathway, and autophagy-related proteins. On the contrary, Andro-activated autophagy was compromised when pretreated with 3-MA. Furthermore, Andro activated the Nrf2/KEAP1 pathway, leading to increasing genes encoding antioxidant enzymes and activities. This study elucidated that Andro exhibited significant neuroprotective effects against MPP+-induced SH-SY5Y cell death in vitro by enhancing mitophagy and clearance of alpha-synuclein through autophagy, as well as increasing antioxidant capacity. Our results provide evidence that Andro could be considered a potential supplement for PD prevention.

Abstract
Extracts from Holothuria scabra (HS) have been shown to possess anti-inflammation, anti-oxidant and anti-cancer activities. More recently, it was shown to have neuroprotective potential in Caenorhabditis elegans PD model. Here, we assessed whether HS has neuroprotective and neurorestorative effects on dopaminergic neurons in both mouse and cellular models of PD. We found that both pre-treatment and post-treatment with HS improved motor deficits in PD mouse model induced with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) as determined by grid walk test. This was likely mediated by HS protective and restorative effects on maintaining the numbers of dopaminergic neurons and fibers in both substantia nigra pars compacta (SNpc) and striatum. In a cellular model of PD, HS significantly attenuated 1-methyl-4-phenylpyridinium (MPP+)-induced apoptosis of DAergic-like neurons differentiated from SH-SY5Y cells by enhancing the expression of Bcl-2, suppressing the expression of cleaved Caspase 3 and preventing depolarization of mitochondrial membrane. In addition, HS could stimulate the expression of tyrosine hydroxylase (TH) and suppressed the formation of α-synuclein protein. Taken together, our in vivo and in vitro findings suggested that HS is an attractive candidate for the neuroprotection rather than neurorestoration in PD.
