ในปัจจุบันได้มีพัฒนาเทคนิคใหม่ในการป้องกันกุ้งจากไวรัส โดยใช้วิธี RNA interference (RNAi) โดยการฉีดหรือผสมในอาหารที่ค่อยข้างยุ่งยาก ทีมวิจัยจึงได้พัฒนาการบรรจุสารพันธุกรรม dsRNA ลงใน อนุภาคเลียนแบบไวรัส (VLPs) แล้วให้กุ้งดูดซึมผ่านการแช่น้ำ แทนการฉีดเข้าไปในตัวกุ้งซึ่งทำได้ง่ายขึ้นต่อเกษตรกรผู้เพาะเลี่บงกุ้ง

Abstract

One of the most successful applications to protect aquatic animals from viral infection is the use of RNA interference (RNAi) in the form of double-stranded RNA (dsRNA). However, its administration into animals is usually via injection route (laboratory scale) or mixing feed pellets with bacterial extracts (containing therapeutic compounds); the outcome of which remains to be improved. In this study, we encapsulated purified dsRNA into a virus-like particle (VLP) and delivered it into the prawn tissues by a submersion route using interiorly modified Macrobrachium rosenbergii nodavirus derived VLP with deca-arginine peptide (10R-MrN-VLP) to encapsulate a large scale (gram-level) of dsRNA against RNA2 gene of MrNV capsid. For tracing purposes of submersion administration, GFP plasmid-loaded VLP successfully delivered and expressed green fluorescent protein in prawn-targeted tissues. Delivery efficiency at 12-h submersion was about 50% of the prawn population and became 100% within 72 h, and they remained GFP-positive for up to 7 days during submersion. The protective effect of dsRNA-VLP submersion was tested with a large group (10,000) of post-larvae, which were naturally challenged with MrNV-positive dead prawn. Transcriptome analysis revealed up- and down-regulations of gene clusters involved in metabolism and signal transduction related to an antiviral immune system. The survival rate of prawn treated with dsRNA-VLP reached 80% at Day 7 post-MrNV challenge, significantly different from that of the MrNV-challenged group (control). PCR screening revealed an apparent decrease of MrNV genes and the copy number of MrNV in prawn sampling tissues. Together, we believe that the integration between the chimeric nanocontainer, RNAi technology and submersion application should be foreseen as one of the practical, powerful means to fight against prawn viral infection in field application.

โควิด-19 ซึ่งเกิดจากเชื้อไวรัส SARS-CoV-2 ส่งผลกระทบหลักต่อปอด ทำให้เกิดโรคปอดบวมและทำความเสียหายต่อเนื้อเยื่อปอดได้ ไวรัสนี้เข้าสู่เซลล์โดยใช้โปรตีนหนาม (spike protein) ที่จับกับตัวรับ ACE2 ซึ่งพบได้บ่อยในเซลล์ของปอดและเนื้อเยื่ออื่น ๆ ในร่างกาย การศึกษานี้ได้สร้างอนุภาคคล้ายไวรัส (VLPs) หรือไวรัสที่มีเฉพาะโครงภายนอกไม่สามารถติดเชื้อได้ จากไวรัสของกุ้งก้ามกราม โดยออกแบบให้อนุภาคมีโปรตีนขนาดเล็กที่เรียกว่า ACE2tp ซึ่งสามารถจับกับตัวรับ ACE2 บนผิวเซลล์ได้ โดยให้ชื่อว่า ACE2tp-MrNV-VLPs การเติมโปรตีน ACE2tp เข้าไปบนผิวของอนุภาคไม่ได้ส่งผลกระทบต่อโครงสร้างและรูปทรงปกติของอนุภาคคล้ายไวรัสเหล่านี้ แต่พบว่าโปรตีน ACE2tp ที่เพิ่มเข้าไป กลับช่วยให้อนุภาคจับและเข้าสู่เซลล์มนุษย์ที่มีตัวรับ ACE2 ได้อย่างมีประสิทธิภาพ นอกจากนี้แล้วยังพบว่า อนุภาค ACE2tp-MrNV-VLPs สามารถยับยั้งการจับและเข้าสู่เซลล์ของไวรัส SARS-CoV-2 สายพันธุ์ดั้งเดิม (Wuhan) และสายพันธุ์กลายพันธุ์ เช่น Omicron BA.2 และ Delta ได้ การวิจัยนี้แสดงให้เห็นว่าอนุภาค ACE2tp-MrNV-VLPs อาจถูกนำมาใช้คล้ายวัคซีนเพื่อป้องกันการติดเชื้อจาก SARS-CoV-2 สายพันธุ์ต่าง ๆ และอาจใช้เป็นตัวนำส่งยารักษาโรคไปยังเซลล์ที่ติดเชื้อในอนาคตได้อีกด้วย

Abstract

SARS-CoV-2, the cause of COVID-19, primarily targets lung tissue, leading to pneumonia and lung injury. The spike protein of this virus binds to the common receptor on susceptible tissues and cells called the angiotensin-converting enzyme-2 (ACE2) of the angiotensin (ANG) system. In this study, we produced chimeric Macrobrachium rosenbergii nodavirus virus-like particles, presenting a short peptide ligand (ACE2tp), based on angiotensin-II (ANG II), on their outer surfaces to allow them to specifically bind to ACE2-overexpressing cells called ACE2tp-MrNV-VLPs. Replacing the ACE2tp at the protruding domains (P-domain) of the MrNV capsid proteins did not affect their normal assembly into icosahedral VLPs. The presentation of the ACE2tp on the P-domains significantly improved the binding and internalization of ACE2tp-MrNV-VLPs to hACE2-overexpressing HEK293T cells in a concentration-dependent manner. Furthermore, ACE2tp-MrNV-VLPs exhibited the ability to block the binding and infection of SARS-CoV-2 pseudovirus variants, including Wuhan, BA.2 Omicron, and Delta subtypes. Our results suggest that chimeric ACE2tp-MrNV-VLPs can serve as a blocking agent against various SARS-CoV-2 mutated variants and could also potentially serve as target-specific nano-containers to carry therapeutic agents to combat SARS-CoV-2 infections in the future.

ปัจจุบัน ได้มีหลักฐานของการใช้บรรจุภัณฑ์นาโนที่บรรจุสารพันธุกรรมออกฤทธิ์ (dsRNA) เพื่อลดการติดเชื้อไวรัสในกุ้งที่สามารถหวังผลสัมฤทธิ์ได้สูง ในการศึกษาครั้งนี้ เราจึงทดสอบการใช้บรรจุภัณฑ์นาโนเพื่อบรรจุสารพันธุกรรมออกฤทธิ์ 2 ชนิดพร้อมกัน (Co-encapsulation) คือ สารพันธุกรรมออกฤทธิ์ต่อโปรตีน VP28 และ VP37 ของไวรัสก่อโรคตัวแดงดวงขาวในกุ้ง (WSSV) จำนวนอย่างละ 5 ไมโครกรัม เพื่อทดสอบความสามารถในการลดปริมาณเชื้อไวรัสในกุ้ง ผลการทดลองพบว่า สารพันธุกรรมออกฤทธิ์ต่อโปรตีน VP37 สามารถบรรจุเข้าไปในช่องว่างของบรรจุภัณฑ์นาโนได้มากกว่าสารพันธุกรรมออกฤทธิ์ต่อโปรตีน VP28 และให้ผลในการป้องกันเชื้อไวรัส WSSV ได้ดีกว่าการบรรจุสารพันธุกรรมออกฤทธิ์เพียงชนิดเดียว โดยทดสอบจากปริมาณของยีนและจำนวนตัวไวรัส WSSV ที่หลงเหลืออยู่ในเนื้อเยื่อเหงือกของกุ้ง นอกจากนี้ยังช่วยเพิ่มปริมาณของเม็ดเลือดขาว และสารต่อต้านเชื้อโรคชนิดโปรอ็อกซิเดส (Pro-oxidase, PO) และกระตุ้นการแสดงออกของยีน PrePO ในเนื้อเยื่อของกุ้งให้สูงขึ้น โดยมีผลในการต่อสู้กับเชื้อไวรัส ทำให้ลดอัตราการตายของกุ้งได้อย่างมีประสิทธิภาพมากกว่าการใช้บรรจุภัณฑ์นาโนที่บรรจุสารพันธุกรรมออกฤทธิ์เพียงชนิดเดียว ผลการทดลองนี้น่าจะนำไปสู่กรรมวิธีในการออกแบบผลิตภัณฑ์ที่ใช้ในการป้องกันการติดเชื้อไวรัสในเชิงเพาะเลี้ยงต่อไป

Abstract

Accumulative evidence of using double stranded (ds) RNA encapsulated into virus like particle (VLP) nanocarrier has open feasibility to fight against shrimp viral infection in aquaculture field. In this study, we co-encapsulated VP37 and VP28 dsRNA into hypodermal and hematopoietic necrosis virus (IHHNV) like particle and investigated its protection against white spot syndrome virus (WSSV). Five micrograms of each dsRNA were used as starting materials to load into VLP, while the loading efficiency was slightly different, i.e, VP37 dsRNA had somewhat a better load into VLP’s cavity. It was apparent that co-encapsulation of dual dsRNA showed a superior WSSV silencing ability than the single dsRNA counterpart as evidence by the lower WSSV gene expression and its copy number in the gill tissues. Besides, we also demonstrated that co-encapsulated dual dsRNA into IHHNV-VLP stimulated the increased number of hemocytes and the corresponding PO activity as well as up-regulated proPO gene expression in hemocytes to resist viral invasion after an acute stage of WSSV infection. This synergistic action of dual dsRNA encapsulated into IHHNV-VLPs could thus act to delay time of shrimp death and reduced shrimp cumulative mortality greater than the single, naked dsRNA treatment and positive control groups. The obtaining results would encourage the feasibility to use it as a new weapon to fight WSSV infection in shrimp aquaculture.
