Abstract

Sulfated polysaccharides from marine macroalgae are increasingly recognized as high-value bioproducts with diverse biofunctional properties. This study investigated sulfated galactan (SG) extracted from the red alga Gracilaria fisheri (G. fisheri) as a gut-modulating algal biopolymer and evaluated its structural stability, microbial selectivity, and effects on intestinal health. SG exhibited a high molecular weight (259 kDa) that remained largely unchanged during simulated oral, gastric, and intestinal digestion, indicating strong resistance to enzymatic and acidic degradation. In vitro fermentation showed that SG selectively stimulated the growth of the probiotics Lactobacillus casei and Lactobacillus plantarum and enhanced β-galactosidase activity. Molecular docking further demonstrated promising interactions between SG subunits and β-galactosidase catalytic residues. An intestinal permeability assay confirmed that SG did not traverse the Caco-2 monolayer, suggesting that it remains unabsorbed and available for probiotic fermentation upon reaching the colon. Consistent with these in vitro results, SG supplementation in mice revealed improved colonic epithelial proliferation and enhanced mucosal architecture, supporting its benefits to gut barrier integrity. Taken together, these findings identify SG as a promising high-value algal bioproduct derived from G. fisheri with potential applications in gut health.

Abstract

Staphylococcus epidermidis is a leading opportunistic pathogen in medical device-associated infections due to its ability to adhere to abiotic materials and develop biofilms that are difficult to eradicate. This study investigated the antibiofilm potential of an ethanolic extract of the red seaweed Gracilaria fisheri and its purified constituent, N-benzylcinnamamide, against S. epidermidis. Antibacterial activity was determined, and antibiofilm effects were assessed using the crystal violet assay and confocal laser scanning microscopy (CLSM). Early bacterial adhesion on glass and polyurethane (PU) surfaces was measured. The effect on catheter-associated biofilms was evaluated by scanning electron microscopy (SEM). Transcripts of biofilm- and quorum-sensing-associated genes (icaA and luxS) were assessed by semi-quantitative RT-PCR. Cytotoxicity was evaluated by MTT assay. At 200 µg/mL, biofilm biomass decreased to 48.21 ± 5.52% with the extract and to 36.65 ± 6.82% with N-benzylcinnamamide. CLSM time-course imaging showed delayed biofilm maturation and less consolidated, discontinuous structures. Surface exposure to the extract markedly reduced early attachment on both materials. On PU catheter segments, SEM demonstrated that N-benzylcinnamamide markedly reduced surface coverage and disrupted three-dimensional biofilm architecture. At the molecular level, transcription of icaA and luxS was reduced. Both the extract and N-benzylcinnamamide showed minimal cytotoxicity in HeLa cells. These findings support further evaluation of these marine-derived agents as candidates for antibiofilm surface treatments to reduce early medical device colonization.

Abstract

Vibrio harveyi (V. harveyi) is a major bacterial pathogen that significantly impacts shrimp cultivation. This study aimed to evaluate the potential of bioflocs, harvested from shrimp pond systems, as a fishmeal substitute to combat V. harveyi infection in shrimp Penaeus vannamei (P. vannamei). Shrimp were fed with 6 diet formulations: Four experimental diets in which fishmeal was replaced with biofloc meal at 25, 50, 75 and 100 % (designated B25, B50, B75, and B100, respectively), a fishmeal-based control diet without biofloc (B0), and a positive control diet supplemented with β-glucan (BG). Feeding trials were conducted for 30 days, followed by a challenge with V. harveyi. Growth performance, survival rates, total hemocyte count (THC), phagocytic activity, and histopathological changes were evaluated. The results demonstrated that all biofloc-fed groups had growth performance comparable to the B0 and BG groups. Shrimp fed with the B25, B50, and B75 diets exhibited significantly enhanced immune responses, with elevated THC and phagocytic activity. In the V. harveyi challenge, the B25 group showed the highest survival rate among biofloc diets, comparable to that of the BG group. Histological analysis revealed that B25 and B50 diets reduced hepatopancreatic and muscle damage associated with infection. In conclusion, this study provides evidence that substituting fishmeal with biofloc (25 – 50 %) effectively enhances immune function and confers protection against V. harveyi infection in P. vannamei, supporting its potential as a sustainable functional feed ingredient in shrimp aquaculture.

ที่มาและความสำคัญ

มะเร็งท่อน้ำดี (cholangiocarcinoma; CCA) เป็นมะเร็งที่มีความรุนแรงสูง ตรวจพบมักอยู่ในระยะลุกลาม และมีทางเลือกการรักษาจำกัด ทำให้ผลการรักษาและอัตราการรอดชีวิตยังไม่ดี แม้จะมีการพัฒนายามุ่งเป้า แต่ผู้ป่วยจำนวนมากยังดื้อยาและเกิดการกลับเป็นซ้ำได้บ่อย ผู้ป่วย CCA แสดงออก epidermal growth factor receptor (EGFR) มากเกินสัมพันธ์กับการกลับเป็นซ้ำและพยากรณ์โรคที่แย่ อย่างไรก็ตามการรักษาที่มุ่งเป้า EGFR ที่ใช้ในปัจจุบันให้ประสิทธิผลจำกัดใน CCA อาจเนื่องมาจากความหลากหลายของก้อนมะเร็ง การส่งสัญญาณชดเชยของเส้นทางอื่น และความสามารถของเซลล์มะเร็งในการหลบเลี่ยงการยับยั้ง ดังนั้นงานวิจัยนี้จึงมีเป้าหมายเพื่อค้นหา “ตัวเลือกใหม่” ในการยับยั้ง EGFR ที่อาจมีศักยภาพมากกว่าเดิม โดยใช้เทคโนโลยี phage display คัดเลือกแอนติบอดีจากคลัง naïve single-chain variable fragment (scFv) มนุษย์ผ่านกระบวนการ subtractive bio-panning เพื่อให้ได้ scFv ที่จับกับเซลล์ CCA อย่างจำเพาะและลดการจับข้ามกับเซลล์ปกติ/มะเร็งชนิดอื่น แนวทางนี้คาดว่าจะได้แอนติบอดีที่สามารถเข้าสู่เซลล์และยับยั้งโดเมนไทโรซีนไคเนสของ EGFR (EGFR-TK) ได้โดยตรง ซึ่งอาจนำไปสู่การพัฒนาเครื่องมือรักษาแบบแอนติบอดีสำหรับ CCA ที่เป้าหมายคือ EGFR

Abstract

Cholangiocarcinoma (CCA) is an aggressive malignancy with limited treatment options. EGFR overexpression is associated with tumor recurrence and poor prognosis, yet current EGFR-targeted therapies show limited efficacy. To identify alternative therapeutic candidates, we performed subtractive bio-panning of a naïve human single-chain variable fragment (scFv) phage display library against CCA cell lysates. Three novel scFv antibodies-E1, G8, and H2-were selected based on preferential binding to CCA cells with minimal cross-reactivity to unrelated cancers and normal fibroblasts. Surface plasmon resonance and kinase inhibition assays demonstrated that G8 and H2 bound the EGFR tyrosine kinase (EGFR-TK) domain with nanomolar affinities and suppressed kinase activity, whereas E1 showed weak binding and no kinase inhibition. All three scFvs exhibited efficient internalization into EGFR-overexpressing HuCCA-1 cells. Functional analyses revealed distinct effects on cell growth: G8 and H2 reduced EGFR phosphorylation, decreased cell viability, and induced apoptosis accompanied by S/G₂-M phase accumulation, whereas E1 primarily inhibited proliferation through G₁-phase arrest without significantly affecting EGFR phosphorylation. Molecular docking predicted that G8 interacts near the ATP-binding pocket and H2 at the dimerization interface of EGFR-TK. These interactions may contribute to EGFR inhibition in HuCCA-1 cells. These findings suggest that G8 and H2 function as intracellular inhibitors of EGFR-TK activity and are promising candidates for antibody-based therapy in EGFR-driven CCA.

มะเร็งท่อน้ำดี (CCA) รักษายากเพราะมีสภาพแวดล้อมของก้อนมะเร็งซับซ้อนและยังขาดเป้าหมายการรักษาที่ได้ผล งานวิจัยนี้ทดสอบแนวคิด CAR T cell ที่มุ่งเป้าไปที่ mesothelin (MSLN) ซึ่งพบแสดงออกสูงใน CCA โดยพัฒนา CAR T จากแอนติบอดี “กระต่ายแบบ humanized” ที่จับ MSLN ต่างตำแหน่งกัน ผลพบว่า CAR T ที่สร้างจากแอนติบอดี hYP218 (ออกแบบโครงสร้าง/การจัดวางเฉพาะ) สามารถกำจัดก้อนมะเร็งในหนูได้ดี มีความคงอยู่ของเซลล์สูง และมีผลข้างเคียงน้อย ชี้ว่า CAR T targeting MSLN อาจเป็นทางเลือกการรักษาใหม่ที่มีศักยภาพสำหรับ CCA

ที่มาและความสำคัญ

มะเร็งท่อน้ำดี (CCA) เป็นมะเร็งที่รักษายาก เพราะสภาพแวดล้อมของก้อนมะเร็งซับซ้อน มีพังผืดมากทำให้ T cell เข้าไปในก้อนได้ยาก และยังขาด “เป้าหมาย” ที่ยืนยันใช้รักษาได้จริงในคลินิก แม้ CAR T cell จะได้ผลดีในมะเร็งเม็ดเลือด แต่ยังมีข้อจำกัดใน CCA งานวิจัยนี้เลือก Mesothelin (MSLN) เป็นเป้าหมายของการพัฒนา CAR T cell เนื่องจากมีคุณสมบัติสอดคล้องกับหลักเกณฑ์ของ antigen สำหรับการรักษาแบบจำเพาะในมะเร็งท่อน้ำดี (CCA) โดยเฉพาะอย่างยิ่ง MSLN เป็นโปรตีนบนผิวเซลล์ซึ่งเอื้อต่อการจดจำและการทำงานของ CAR T cell และพบการแสดงออกสูงในเนื้อเยื่อ CCA ของผู้ป่วยไทยในสัดส่วนมาก (79%) สะท้อนความเป็นเป้าหมายที่ครอบคลุมผู้ป่วยจำนวนมาก นอกจากนี้ MSLN ถูกจัดอยู่ในกลุ่ม tumor-associated antigen ที่เกี่ยวข้องกับชีววิทยาการดำรงอยู่และความก้าวร้าวของก้อนมะเร็ง จึงมีศักยภาพในการให้ผลเชิงการรักษามากกว่าเป็นเพียงตัวบ่งชี้การเกิดโรค อีกทั้ง CCA ยังขาดเป้าหมายที่ผ่านการยืนยันทางคลินิกอย่างชัดเจน ทำให้การมุ่งพัฒนา CAR T ที่จำเพาะต่อ MSLN เป็นแนวทางเชิงกลยุทธ์เป้าหมายสำหรับ CAR T cell therapy ใน CCA.

Abstract

Background: Chimeric antigen receptor (CAR) T cell therapy has shown promise in treating hematological malignancies. However, the complex tumor microenvironment of cholangiocarcinoma (CCA) poses significant challenges, particularly due to the lack of clinically validated targets and the presence of fibrosis, which hinders T cell infiltration into tumor sites. Mesothelin (MSLN) is highly expressed in CCA. In this study, we aimed to develop CAR T therapy based on a panel of humanized rabbit monoclonal antibodies targeting various epitopes of MSLN, ranging from the N- to the C-terminus, for CCA.

Methods: MSLN expression was assessed in CCA tissue samples obtained from Thai patients. CAR T cells were generated using various single-chain variable fragment (scFv) constructs, engineered in either VH-linker-VL or VL-linker-VH orientation, targeting non-overlapping epitopes of membrane-bound MSLN: hYP218 (proximal region), hYP223 and hYP3 (middle region), and hYP158 (distal region). The cytotoxicity of MSLN-specific CAR T cells was evaluated in 3 preclinical CCA mouse models: Mz-ChA-1, KMCH, and KMBC.

Results: MSLN was strongly expressed in 79% of the CCA specimens. Among the CAR constructs, hYP218-based CAR T cells-with the VL-linker-VH orientation and CD28-derived hinge and transmembrane domains (CD28HTM)-completely eradicated CCA tumors in all 3 CCA xenograft mouse models (Mz-ChA-1, KMCH, and KMBC). Furthermore, hYP218 VLVH CD28HTM CAR T cells showed strong persistence in mice, with low PD-1 expression (a marker of T cell exhaustion) and minimal adverse effects.

Conclusions: Our findings suggest that MSLN is a promising target for CAR T cell therapy in CCA. CAR T cells engineered with hYP218 VLVH CD28HTM, which targets the membrane-proximal epitope of MSLN, may represent a novel therapeutic strategy for the clinical treatment of CCA.

งานวิจัยประเมินซัลเฟต กาแลคแตนชนิดน้ำหนักโมเลกุลต่ำ (LSG) ที่ได้จากสาหร่ายผมนาง Gracilaria fisheri ในแบบจำลองสะเก็ดเงินหนูที่เหนี่ยวนำด้วย imiquimod พบว่า LSG ลดความหนาหนังกำพร้า การสร้างหลอดเลือดใหม่ และการอักเสบของผิว ลดการแทรกซึมของ CD4+ T และ mast cell พร้อมกดไซโตไคน์สำคัญ (TNF-α, IL-17A, IL-23 ฯลฯ) และยับยั้งสัญญาณ JAK2/STAT2/STAT3 โดยไม่มีพิษต่อตับและไต

ที่มาและความสำคัญ

โรคสะเก็ดเงินเป็นโรคผิวหนังอักเสบเรื้อรัง สาเหตุจากความผิดปกติของภูมิคุ้มกันร่วมกับการเพิ่มจำนวนของเคราติโนไซต์มากผิดปกติ ส่งผลให้เกิดผื่นหนา แดง ลอก และมีการแทรกซึมของเซลล์อักเสบในผิวหนังอย่างต่อเนื่อง กลไกสำคัญเกี่ยวข้องกับไซโตไคน์ก่อการอักเสบซึ่งกระตุ้นทั้งการอักเสบและการหนาตัวของหนังกำพร้า การเพิ่มจำนวนและการอยู่รอดของเคราติโนไซต์ รวมถึงการคงอยู่ของวงจรการอักเสบ แม้มีการรักษาหลากหลาย ผู้ป่วยจำนวนหนึ่งยังคงตอบสนองไม่ดี เกิดผลข้างเคียงจากการใช้ยาระยะยาว จึงมีความจำเป็นในการค้นหาทางเลือกใหม่ที่ลดการอักเสบได้หลายมิติและมีความปลอดภัยเหมาะสมต่อการใช้ต่อเนื่อง งานวิจัยจึงเลือกประเมิน low molecular weight sulfated galactan (LSG) ซึ่งเป็นพอลิแซ็กคาไรด์ซัลเฟตจากสาหร่ายแดง Gracilaria fisheri มีศักยภาพการลดอาการโรคสะเก็ดเงินได้หรือไม่ เนื่องจาก LSG มีความเป็นไปได้ในการปรับภูมิคุ้มกัน (immunomodulation) และโมเลกุลน้ำหนักต่ำทำให้สารละลายน้ำได้ดีขึ้นและเอื้อต่อการดูดซึม การกระจายตัวในร่างกายเพื่อโอกาสการพัฒนาต่อเป็นยาในอนาคต

Abstract

Psoriasis is a chronic immune‑mediated inflammatory skin disorder characterized by keratinocyte hyperproliferation and immune cell infiltration. Low molecular weight sulfated galactan (LSG), derived from the red seaweed Gracilaria fisheri, has immunomodulatory potential relevant to psoriasis pathogenesis. The present study investigated the therapeutic potential of LSG using an imiquimod (IMQ)‑induced murine model of psoriasis. BALB/c mice received intraperitoneal administration of LSG once daily, 2 h prior to IMQ application, for 7 consecutive days. Clinical severity was assessed using the Psoriasis Area and Severity Index (PASI). Histopathological and immunohistochemical analyses were performed to evaluate epidermal architecture, vascular change and immune cell infiltration. Expression of keratinocyte proliferation and differentiation markers, proinflammatory cytokines and JAK/STAT pathway components was analyzed using reverse transcription‑quantitative PCR, ELISA and western blotting. Systemic inflammation was assessed by spleen and lymph node size. LSG significantly decreased PASI scores, neovascularization, epidermal thickness and dermal inflammation. LSG downregulated keratinocyte proliferation and differentiation markers (Ki67, keratin 6, 16 and 17, involucrin) and decreased proinflammatory cytokine expression (TNF‑α, IL‑1β, IL‑17A, IL‑23, IL‑6), accompanied by decreased CD4+ T and mast cell infiltration. LSG downregulated JAK2/STAT2/STAT3 signaling and downstream genes (BCL2, CCND1). Furthermore, LSG alleviated systemic inflammation without inducing hepatic or renal toxicity. These findings indicated that LSG effectively ameliorates IMQ‑induced psoriatic inflammation via coordinated reduction of keratinocyte hyperproliferation, cytokine production and immune cell activity. LSG represents a promising marine‑derived therapeutic candidate for psoriasis management.

Abstract

Red seaweeds (Rhodophyta) and their polysaccharides represent promising, sustainable solutions for boosting aquaculture productivity. Rich in bioactive compounds such as carrageenan, agar, and sulfated galactans, these seaweeds have demonstrated the ability to improve growth, health, and disease resistance in various aquaculture species. This review examines the applications, benefits, and limitations of red seaweeds in aquaculture, with particular attention to their roles as feed binders, partial fishmeal replacements, feed supplements, and immunostimulants. Species such as Gracilaria, Kappaphycus, and Porphyra have been shown to enhance immune responses, promote gut health, and mitigate nutrient pollution in aquaculture systems. Despite these advantages, challenges remain, including high cultivation costs, variability in bioactivity, and supply chain constraints. This review highlights the growing potential of red seaweeds for sustainable aquaculture while identifying research needs related to dosage optimization, delivery strategies, and elucidating the molecular mechanisms of their effects. Supplementation with red seaweeds aligns with the United Nations’ Sustainable Development Goals, offering an eco-friendly alternative to antibiotics with the potential to yield both environmental and economic benefits in aquaculture.

โรคข้อเข่าเสื่อมเป็นภาวะเสื่อมเรื้อรังที่พบมากในผู้สูงอายุ เนื้อเยื่อกระดูกอ่อนในข้อต่อถูกทำลาย ส่งผลต่อการเคลื่อนไหวและคุณภาพชีวิต แนวทางการรักษาในปัจจุบันมุ่งบรรเทาอาการแต่ไม่สามารถฟื้นฟูเนื้อเยื่อที่สูญเสียได้อย่างแท้จริง จึงมีความจำเป็นต้องค้นหาสารชีวภาพที่สามารถกระตุ้นการสร้างกระดูกอ่อน งานวิจัยนี้มุ่งศึกษาซัลเฟตกาแลคแตนจากสาหร่ายแดง Gracilaria fisheri ซึ่งมีโครงสร้างคล้ายไกลโคซามิโนไกลแคนในเนื้อเยื่อกระดูกอ่อนเพื่อประเมินศักยภาพในการส่งเสริมการสร้าง Aggrecan และ COL2A1 ในเซลล์กระดูกอ่อนมนุษย์ นำไปสู่การพัฒนาทางเลือกใหม่ในการฟื้นฟูเนื้อเยื่อกระดูกอ่อนสำหรับการรักษาโรคข้อเสื่อมในอนาคต

Abstract

Osteoarthritis (OA) is characterized by progressive cartilage degradation and limited tissue regeneration, commonly affecting older adults. Current treatment strategies mainly alleviate symptoms without effectively restoring cartilage integrity. Sulfated polysaccharides derived from marine algae have attracted attention for their potential biological effects in promoting cartilage repair. This study examined the chondrogenic activity of sulfated galactans (SG) isolated from the marine red alga Gracilaria fisheri in human C28/I2 chondrocytes, using transforming growth factor-beta 3 (TGF-β3) as a comparative control. SG treatment significantly increased the synthesis of essential cartilage extracellular matrix (ECM) components, such as type II collagen and aggrecan. These effects were mediated through the integrin β1/FAK/Akt signaling pathway, as demonstrated by reduced ECM synthesis following treatment with the Akt inhibitor, MK2206. Additionally, SG improved chondrocyte adhesion and proliferation. These results suggest that sulfated galactans from Gracilaria fisheri may support cartilage ECM synthesis and exhibit potential as biologically active components for osteoarthritis management.

ที่มาและความสำคัญ

Abstract

Exposure to oxalate crystals causes cellular injury and dysfunction in the renal tubular epithelium. Sulfated galactan with increased sulfation (SGS) from the red seaweed Gracilaria fisheri exhibits anti‑urolithiasis effects by inhibiting oxalate crystal formation and preventing sodium oxalate (NaOX)‑induced death of renal tubular (HK‑2) cells. However, the effects of SGS on wound healing and adhesion molecule expression in NaOX‑induced HK‑2 cell injury remain unexplored. The present study investigated the effects of SGS on wound healing and the regulation of adhesion molecule expression in NaOX‑induced HK‑2 cell damage. The findings showed that SGS promoted wound healing in HK‑2 cells following a scratch injury under NaOX‑induced conditions. NaOX exposure increased the expression of CD44 and vimentin while decreasing the expression of EpCAM, E‑cadherin, occludin and ZO‑1, as demonstrated by reverse transcription‑quantitative PCR, western blotting and immunofluorescence analysis. Treatment with SGS restored these adhesion molecule expression levels to near normal. Scanning electron microscopy revealed that SGS also reversed NaOX‑induced morphological changes in HK‑2 cells. Additionally, SGS reduced the expression of Akt and p38 while upregulating PI3K and Erk1/2 in NaOX‑treated HK‑2 cells. These results suggested that SGS enhances wound healing and regulates the expression of adhesion molecules, possibly through the PI3K/Akt and MAPK (p38 and Erk1/2) signaling pathways, highlighting the potential of SGS as a promising therapeutic compound for preventing and treating NaOX‑induced renal damage.
