รังสีอัลตราไวโอเลต เป็นปัจจัยสำคัญที่ก่อให้เกิดความเครียดออกซิเดชัน (oxidative stress) ในเซลล์ผิวหนัง ส่งผลให้เกิดการสะสมของอนุมูลอิสระ กระตุ้นการอักเสบ การสลายตัวของคอลลาเจน การเสื่อมสภาพของผิวหนัง และเพิ่มความเสี่ยงต่อการเกิดโรคผิวหนังและมะเร็งผิวหนัง สาหร่ายพวงองุ่น (Caulerpa lentillifera) เป็นสาหร่ายสีเขียวที่อุดมด้วยสารออกฤทธิ์ทางชีวภาพที่มีคุณสมบัติต้านอนุมูลอิสระ งานวิจัยนี้จึงมุ่งศึกษาศักยภาพของสารสกัดจากสาหร่ายพวงองุ่นในการปกป้องเซลล์ผิวหนังจากรังสีอัลตราไวโอเลต และการศึกษากลไกระดับโมเลกุลผ่านการกระตุ้นเส้นทาง Nrf2/HMOX1 และการยับยั้งเอนไซม์ Matrix Metalloproteinases ซึ่งเป็นกลไกสำคัญในการรักษาสมดุลของระบบต้านอนุมูลอิสระและโครงสร้างของผิวหนัง

Abstract

Caulerpa lentillifera is an edible green seaweed widely consumed in Southeast Asia and has been recognized for its nutritional and antioxidant properties. Previous studies demonstrated that its ethyl acetate extract (CLEA) exhibits strong free radical-scavenging and metabolic regulatory activities; however, its potential protective role against ultraviolet (UV)-induced oxidative stress has not yet been elucidated. Therefore, this study aimed to evaluate the photoprotective effects of CLEA on UV-induced cellular damage in keratinocytes (HaCaT) and fibroblasts (L929), and explore its underlying mechanisms. The results revealed that CLEA significantly improved cell viability following UV irradiation and attenuated intracellular reactive oxygen species (ROS) accumulation in both keratinocytes and fibroblasts. Mechanistically, CLEA enhanced nuclear translocation of nuclear factor erythroid 2–related factor 2 (Nrf2) and upregulated the mRNA expression of downstream antioxidant enzymes, particularly heme oxygenase-1 (HMOX1). In addition, CLEA suppressed UV-induced expression of matrix metalloproteinases, MMP1 and MMP3, while promoting procollagen synthesis, suggesting its role in preserving skin integrity. Collectively, these findings suggest that CLEA confers photoprotective effects against UV-induced oxidative stress and extracellular matrix degradation through activation of the Nrf2/HMOX1 pathway and inhibition of MMPs. This study highlights the potential of C. lentillifera extract as a natural photoprotective agent, warranting further in vivo and clinical investigations to validate its efficacy and safety.

Abstract

Neurotoxic agents such as 6-hydroxydopamine (6-OHDA) and reserpine are reported to cause neurodegenerative and neuroendocrine effects, respectively. Using the Caenorhabditis elegans (C. elegans) model, this study performs comparative transcriptomic profiling to elucidate molecular pathways that contribute to their neurotoxicity. Differentially expressed genes (DEGs) were identified following 6-OHDA and reserpine exposure, with subsequent analyses for Gene Ontology (GO) and KEGG pathway enrichment. Notably, 6-OHDA mainly disrupted ribosomal/proteasomal activity and neurodegeneration-related pathways, while reserpine primarily affected the extracellular compartment, neuropeptide-based signaling, and metabolic processes. Meta-analysis of the top 3,000 DEGs, combined with comparative enrichment network analysis of the 2 neurotoxicants using Metascape, identified critical regulatory processes, including cell development, reproduction, cytoskeletal organization, cell cycle control, metabolic pathways, and post-transcriptional regulation. In addition, protein-protein interaction (PPI) network analysis revealed eight distinct Molecular Complex Detection (MCODE) modules, which correspond to biological clusters such as regulation of translation, fatty acid metabolism, glutamate/glutamine metabolism, ceramide metabolism, sphingolipid metabolism, porphyrin metabolism, and post-transcriptional regulation. Moreover, reserpine exposure altered neurobehavioral function in the ethanol avoidance assay without inducing dopaminergic (DAergic) neurodegeneration in C. elegans. Overall, these findings suggest a transcriptomic framework that reports both common and unique molecular signatures of neurotoxicant-mediated neurodegeneration and behavioral alterations. These results provide valuable insights for future studies on regulatory pathways involved in PD pathogenesis.

Abstract

The black sea cucumber Holothuria leucospilota, an Asian traditional food and medicine, has been explored for health benefits, including neuroprotective effects. Although its anti-Parkinson’s effects have been demonstrated, its potential anti-Alzheimer’s effects remain unexplored. Therefore, this study aimed to investigate the effects and underlying mechanisms of H. leucospilota extracts on amyloid-β (Aβ) proteotoxicity in a Caenorhabditis elegans model of Alzheimer’s disease (AD). All fractions of H. leucospilota extracts were proven to be non-toxic to C. elegans. Among them, the ethyl acetate fraction (HLEA) at 500 μg/mL exhibited the greatest efficacy due to its ability to delay Aβ-induced paralysis in muscular Aβ-expressing C. elegans. Moreover, HLEA at 500 μg/mL rescued the chemotaxis defects in neuronal Aβ-expressing worms. The levels of Aβ oligomers and deposits declined after HLEA treatment of AD worms. Correspondingly, HLEA alleviated Aβ toxicity by lowering ROS levels and extending the lifespan of AD worms. Furthermore, differential RNA sequencing analysis revealed that the lysosome, proteasome, and antioxidant pathways were activated in HLEA-treated AD worms. RT-qPCR assays confirmed that HLEA upregulated ubiquitin-proteasome system, autophago-lysosomal, and antioxidant genes. Moreover, HLEA treatment promoted the nuclear localization of key transcription factors, SKN-1 and DAF-16, along with upregulation of their downstream antioxidant genes, indicating activation of SKN-1 and DAF-16 signaling pathways. Lastly, mass spectrometry analysis revealed that HLEA was predominantly composed of triterpenoid saponins. In summary, HLEA at 500 μg/mL diminished Aβ proteotoxicity by activating the protein clearance mechanism and SKN-1 and DAF-16 stress resistance pathways in the C. elegans AD model.

Abstract

Cathepsin L1H has been identified as a potential vaccine candidate against fasciolosis. The present study was conducted to evaluate the immunoprophylactic efficacy of recombinant proFgCatL1H (rproFgCatL1H) antigen in goats subsequently challenged with the parasite. Immunization of goats with 200 µg of rproFgCatL1H formulated in Montanide™ ISA 61 VG conferred statistically significant reduction in worm recovery compared with both the infected control and adjuvant control groups, corresponding to reductions of 37.55 % and 34.07 %, respectively (p < 0.05). A negative correlation was observed between worm burden reduction and serum IgG levels (OD₄₅₀), measured both at the time of infection and at the study endpoint, although the association was not statistically significant. Scanning Electron Microscopy (SEM) analysis of adult F. gigantica recovered from vaccinated goats demonstrated pronounced tegumental damage, characterized by extensive swelling, surface erosion, and the presence of cell-like structures adhering to the tegumental surface. These findings suggest that the recombinant protein rproFgCatL1H holds promise as a potential vaccine candidate for the control of fasciolosis in goats.

Abstract

The antiobesity effect of extracts from the marine algae, Halymenia durvillei (HD), was investigated in a glucose-induced Caenorhabditis elegans model of obesity. Total fat accumulation, triglyceride levels, lifespan, intracellular ROS levels, and the potential mechanism of action of H. durvillei extracts were examined. The present study demonstrated that the ethanol fraction of H. durvillei (HDET) and ethyl acetate fraction of H. durvillei (HDEA) extracts led to a significant reduction in fat accumulation, triglyceride levels, the GFP-labeled dhs-3, a marker for lipid droplets, and the intracellular ROS levels. H. durvillei extracts significantly extended the lifespan of glucose-induced worms. In addition, the mRNA expression of lipolysis-related genes, atgl-1 and hosl-1, showed significant upregulation following treatment with H. durvillei extracts. This finding was supported by RNA interference (RNAi) of atgl-1 and hosl-1, which resulted in disrupting the effect of the H. durvillei extracts on lowering fat accumulation. Furthermore, transcriptomic analysis revealed diverse metabolic activities in glucose-induced worms treated with HDEA, affecting fatty acid metabolism. The results suggested that these extracts provide an antiobesity effect mediated through the lipolysis genes, atgl-1 and hosl-1H. durvillei-derived extracts may offer valuable insights as functional food ingredients for use in the prevention of obesity.

Abstract

Holothuria scabra (H. scabra), a marine organism traditionally known for its health benefits, has been utilized in both food and medicine. Our previous studies indicated that 2-butoxytetrahydrofuran (2-BTHF), which is isolated from H. scabra, possesses the potential to alleviate amyloid-β and α-synuclein accumulations associated with Alzheimer’s and Parkinson’s diseases (AD and PD), respectively. However, the mechanisms through which 2-BTHF mitigates PD-related neurotoxicity remain unclear. In this study, we investigated the effects of 2-BTHF on a 6-hydroxydopamine (6-OHDA)-induced Caenorhabditis elegans (C. elegans) model. Our results demonstrated that 2-BTHF recovered dopaminergic (DAergic) neurons from degeneration and restored dopamine-related behaviors. Furthermore, 2-BTHF reduced reactive oxygen species (ROS) production, preserved mitochondrial fluorescence, and decreased both mitochondrial and cytoplasmic unfolded protein responses (UPRmt and UPRcyto) activation. Transcriptome sequencing analysis revealed the critical roles of various systems, including the immune system, nervous system, glutathione (GSH) metabolism, xenobiotics, terpenoids, energy metabolism, cell growth and death, and aging-related longevity pathways. Additionally, 2-BTHF showed potential interactions with stress resistance and detoxification transcription factors, promoting the nuclear translocation of DAF-16 and SKN-1, which in turn activated their targets, including SOD-3, CTL-2, GCS-1, and GST-4. Moreover, 2-BTHF increased total GSH levels and reduced the ced-3-related cascade. This study demonstrates that 2-BTHF holds promise as a therapeutic agent for treating 6-OHDA-induced DAergic neurodegeneration in the C. elegans model.

ที่มาและความสำคัญ

รายงานฉบับนี้นำเสนอกรณีศึกษาที่หายากและซับซ้อนของทารกแรกเกิดไทยคนหนึ่ง ซึ่งมีโรคผิดปกติแต่กำเนิดสองตำแหน่งในระบบทางเดินอาหารซึ่งไม่เคยมีรายงานพบร่วมกันมาก่อน ได้แก่ โรค Retroesophageal right subclavian artery และ Congenital colonic stenosis ซึ่งมีอาการคล้ายโรคที่พบได้บ่อย เช่น โรคแพ้โปรตีนนมวัวหรือภาวะท้องอืดจากการติดเชื้อ ส่งผลให้เกิดการวินิจฉัยผิดพลาด

กรณีนี้ได้เน้นย้ำถึงความสำคัญของการตระหนักถึงโรคแต่กำเนิดที่พบได้น้อยแต่มีผลกระทบรุนแรง ซึ่งหากตรวจพบได้ตั้งแต่เนิ่น ๆ จะช่วยหลีกเลี่ยงการวินิจฉัยผิดและเพิ่มโอกาสในการรักษาทารกในกรณีคล้ายกันในอนาคต

Abstract

Background

Double alimentary tract obstruction due to congenital anomalies is a rare clinical occurrence, with limited cases published in medical literature. This article presents a unique case of coexisting retroesophageal right subclavian artery (RRSA) and congenital colonic stenosis (CCS), conditions that have not been previously documented together in pediatric population.

Case presentation

A Thai male newborn was born by cesarean section at gestational age of 41 weeks. One week before birth, intrauterine asphyxia and idiopathic bilateral intracerebral hemorrhage were diagnosed by prenatal ultrasonography. Despite postnatal interventions including a ventriculoperitoneal shunt and subsequent external ventricular drain, the intracerebral hemorrhage recurred and progressed. Concurrently, the patient experienced multiple episodes of post-feeding vomiting, intermittent abdominal distension, and regular defecation without constipation. Sepsis secondary to an infected shunt occurred, accompanied by marked abdominal distension. The physician clinically suspected non-IgE-mediated cow’s milk protein allergy and ileus associated with sepsis. Tragically, the patient succumbed at seven months due to a brain abscess stemming from an infected external ventricular drain. Ultimately, postmortem examination unraveled double alimentary tract obstruction, consisting of RRSA and CCS. The RRSA, originating from proximal thoracic aorta, caused notable esophageal compression and functional stenosis which led to the frequent vomiting and reflux. The CCS involved the distal transverse colon, descending colon and proximal sigmoid colon, accounting for nearly 50% of the colon. The CCS was therefore the exact cause of intermittent abdominal distension. The stenotic colon contained submucosal and myenteric plexuses, excluding Hirschsprung disease.

Conclusion

This case highlights the diagnostic complexities of RRSA and CCS resulting in double gut obstruction and masquerading as non-IgE-mediated cow’s milk protein allergy and sepsis-induced ileus. Awareness of these rare coexisting congenital anomalies can aid in early recognition, prevent misdiagnosis, enable timely management and improve outcomes for affected pediatric patients.

Abstract

Aggregative α-synuclein and incurring oxidative stress are pivotal cascading events, leading to dopaminergic (DAergic) neuronal loss and contributing to clinical manifestations of Parkinson’s disease (PD). Our previous study demonstrated that 2-butoxytetrahydrofuran (2-BTHF), isolated from Holothuria scabra (H. scabra), could inhibit amyloid-β aggregation and its ensuing toxicity, which leads to Alzheimer’s disease. In the present study, we found that 2-BTHF also attenuated the aggregative and oxidative activities of α-synuclein and lessened its toxicity in a transgenic Caenorhabditis elegans (C. elegans) PD model. Such worms treated with 100 μM of 2-BTHF showed substantial reductions in α-synuclein accumulation and DAergic neurodegeneration. Mechanistically, 2-BTHF, at this concentration, significantly decreased aggregation of monomeric α-synuclein and restored locomotion and dopamine-dependent behaviors. Molecular docking exhibited potential bindings of 2-BTHF to HSF-1 and DAF-16 transcription factors. Additionally, 2-BTHF significantly increased the mRNA transcripts of genes encoding proteins involved in proteostasis, including the molecular chaperones hsp-16.2 and hsp-16.49, the ubiquitination/SUMOylation-related ubc-9 gene, and the autophagy-related genes atg-7 and lgg-1. Transcriptomic profiling revealed an additional mechanism of 2-BTHF in α-synuclein-expressing worms, which showed upregulation of PPAR signaling cascades that mediated fatty acid metabolism. 2-BTHF significantly restored lipid deposition, upregulated the fat-7 gene, and enhanced gcs-1-mediated glutathione synthesis in the C. elegans PD model. Taken together, this study demonstrated that 2-BTHF could abrogate aggregative and oxidative properties of α-synuclein and attenuate its toxicity, thus providing a possible therapeutic application for the treatment of α-synuclein-induced PD.

Abstract

Seaweed has attracted attention as a bioactive source for preventing different chronic diseases, including liver injury and non-alcoholic fatty liver disease, the leading cause of liver-related mortality. Caulerpa lentillifera is characterized as tropical edible seaweed, currently being investigated for health benefits of its extracts and bioactive substances. This study examined the effects of C. lentillifera extract in ethyl acetate fraction (CLEA) on controlling lipid accumulation and lipid metabolism in HepG2 cells induced with oleic acid through the in vitro hepatic steatosis model. Gas chromatography-mass spectrometry (GC-MS) analysis indicated that CLEA contained diverse organic compounds, including hydrocarbons, amino acids, and carboxylic acids. Docked conformation of dl-2-phenyltryptophane and benzoic acid, two major bioactive CLEA components, showed high affinity binding to SIRT1 and AMPK as target molecules of lipid metabolism. CLEA reduced lipid accumulation and intracellular triglyceride levels in HepG2 cells stimulated with oleic acid. The effect of CLEA on regulating expression of lipid metabolism-related molecules was investigated by qPCR and immunoblotting. CLEA promoted expression of the SIRT1 gene in oleic acid-treated HepG2 cells. CLEA also reduced expression levels of SREBF1, FAS, and ACC genes, which might be related to activation of AMPK signaling in lipid-accumulated HepG2 cells. These findings suggest that CLEA contains bioactive compounds potentially reducing triglyceride accumulation in lipid-accumulated HepG2 hepatocytes by controlling lipid metabolism molecules.

Abstract

Cry5B, a crystal protein produced by Bacillus thuringiensis (Bt), is a bionematicide with potent nematicidal activity against various plant-parasitic and free-living nematodes. This protein, however, is susceptible to destruction by ultraviolet light, proteolytic enzymes, and high temperatures. This study aims to produce Cry5B protein for bionematicidal use and improve its stability and nematicidal efficacy by loading it intoArthrobotrys oligospora-mediated sulfur nanoparticles (AO-SNPs). Based on the mortality assay, the Cry5B protein exhibited dose-dependent nematicidal activity against the model organismCaenorhabditis elegans. The nematicidal activity, thermal stability, and pathogenic effects of Cry5B-loaded AO-SNPs (Cry5B-SNPs) were compared to those of free Cry5B. After 3 h of exposure to heat at 60 °C, Cry5B-SNPs had greater nematicidal activity than free Cry5B protein, indicating the effective formulation of Cry5B-SNPs that could be used as an alternative to current nematicide delivery strategies.
