ที่มาและความสำคัญ

ในปัจจุบัน ความสนใจด้านการออกกำลังกายและสุขภาพร่างกายที่ดีเพิ่มสูงขึ้นอย่างต่อเนื่อง ส่งผลให้มีความต้องการผลิตภัณฑ์อาหารฟังก์ชันและอาหารเสริมที่ช่วยฟื้นฟูกล้ามเนื้อและเพิ่มสมรรถภาพทางกายมากขึ้น โดยภาวะเมื่อยล้าของกล้ามเนื้อและการสูญเสียมวลกล้ามเนื้อมักเป็นปัญหาที่เกิดขึ้นจากการออกกำลังกายอย่างหนัก ซึ่งมีสาเหตุหลักจากการสลายตัวของโปรตีนในกล้ามเนื้อร่วมกับการสะสมของอนุมูลอิสระ (ROS) ภายในเซลล์กล้ามเนื้อ การผลิต ROS ในปริมาณมากส่งผลให้เกิดความเครียดออกซิเดชัน ซึ่งทำให้การทำงานของกล้ามเนื้อแย่ลงและเพิ่มความเสี่ยงต่อการบาดเจ็บของเนื้อเยื่อกล้ามเนื้อ ดังนั้น การพัฒนากลยุทธ์เพื่อลดความเครียดออกซิเดชันและเร่งกระบวนการฟื้นฟูกล้ามเนื้อจึงเป็นสิ่งสำคัญ โดยเฉพาะสำหรับนักกีฬา ผู้ที่ออกกำลังกายเป็นประจำ และผู้ที่ต้องการรักษามวลกล้ามเนื้อ
ปัจจุบันเลือดปลาทูน่าเป็นของเสียจากอุตสาหกรรมที่ยังไม่ได้ใช้ประโยชน์ ทั้งที่อุดมด้วยกรดอะมิโนและแร่ธาตุสำคัญ อีกทั้งยังมีรายงานว่าเปปไทด์จากน้ำต้มปลาทูน่าสามารถยับยั้งเอนไซม์ ACE และกระตุ้นการเกิด apoptosis ในเซลล์มะเร็งเต้านม อย่างไรก็ตาม ยังไม่มีการศึกษาอย่างชัดเจนเกี่ยวกับศักยภาพของไฮโดรไลเซตจากเลือดปลาทูน่าในการเป็นวัตถุดิบเชิงหน้าที่เพื่อการฟื้นฟูกล้ามเนื้อ งานวิจัยนี้จึงมุ่งพัฒนาไฮโดรไลเซตจากเลือดปลาทูน่าเพื่อใช้เป็นส่วนผสมในผลิตภัณฑ์เพื่อสุขภาพกล้ามเนื้อ พร้อมส่งเสริมการใช้ทรัพยากรอย่างคุ้มค่าและยั่งยืน

ภาวะความล้าของกล้ามเนื้อจากการออกกำลังกายแบบทนทาน (exercise-induced muscle fatigue) เป็นภาวะที่ส่งผลให้สมรรถนะในการออกกำลังกายลดลง โดยมีปัจจัยสำคัญ ได้แก่ การลดลงของแหล่งพลังงาน การสะสมของสารเมแทบอไลต์ และภาวะ oxidative stress ทั้งนี้ AMPK/SIRT1/PGC-1α signaling pathway มีบทบาทสำคัญในการควบคุมสมดุลพลังงาน การสร้างไมโทคอนเดรีย และการผลิตพลังงาน ซึ่งมีส่วนช่วยในการคงสมรรถนะของกล้ามเนื้อและชะลอการเกิดความล้า

ปัจจุบันมีความสนใจในการนำสารออกฤทธิ์ทางชีวภาพจากแหล่งธรรมชาติมาพัฒนาเพื่อช่วยลดความล้าจากการออกกำลังกาย ขณะเดียวกัน ผลพลอยได้จากอุตสาหกรรมอาหารยังเป็นแหล่งของโปรตีนและสารออกฤทธิ์ทางชีวภาพที่มีศักยภาพแต่ยังไม่ได้รับการใช้ประโยชน์อย่างเต็มที่ โดยเฉพาะอุตสาหกรรมปลาทูน่าที่ก่อให้เกิดผลพลอยได้จำนวนมาก เช่น หัว กระดูก หนัง อวัยวะภายใน และเลือด การนำ เลือดปลาทูน่า ซึ่งเป็นผลพลอยได้ที่ยังมีการใช้ประโยชน์ค่อนข้างจำกัดมาเพิ่มมูลค่า จึงสามารถช่วยลดของเสียและผลกระทบต่อสิ่งแวดล้อมได้

การย่อยโปรตีนจากเลือดปลาทูน่าด้วยเอนไซม์ทำให้ได้ tuna blood hydrolysate (TBH) ซึ่งประกอบด้วยกรดอะมิโนและ bioactive peptides ที่มีศักยภาพทางชีวภาพ อย่างไรก็ตาม แม้จะมีรายงานว่า TBH มีฤทธิ์ต้านอนุมูลอิสระและต้านการอักเสบ รวมทั้งมีองค์ประกอบที่เกี่ยวข้องกับ energy metabolism แต่ข้อมูลเกี่ยวกับฤทธิ์ลดความล้าในสัตว์ทดลองและกลไกที่เกี่ยวข้องกับการควบคุมพลังงานและการทำงานของไมโทคอนเดรียยังมีจำกัด

ดังนั้น การศึกษานี้จึงมุ่งประเมินศักยภาพของ TBH ในการเพิ่มความทนทานและลดความล้าจากการออกกำลังกาย รวมถึงศึกษากลไกผ่าน AMPK/SIRT1/PGC-1α signaling pathway และวิเคราะห์ bioactive peptides ที่เกี่ยวข้อง เพื่อเป็นข้อมูลพื้นฐานสำหรับการพัฒนา TBH เป็น functional food หรือ nutritional supplement ตลอดจนส่งเสริมการเพิ่มมูลค่าและการใช้ประโยชน์จากผลพลอยได้ของอุตสาหกรรมปลาทูน่าอย่างยั่งยืน

Abstract

Exercise-induced muscle fatigue is associated with energy depletion, metabolite accumulation, and oxidative stress. These processes are closely linked to molecular mechanisms involving the AMPK/SIRT1/PGC-1α signaling pathway, which plays a crucial role in regulating energy homeostasis and mitochondrial biogenesis in response to exercise and nutritional interventions. Tuna blood hydrolysate (TBH) is a potential source of bioactive peptides that may regulate energy metabolism and alleviate exercise-induced muscle fatigue. Therefore, this study aimed to evaluate the effects of TBH as a nutritional supplement on muscle endurance and its anti-fatigue properties in vivo, as well as to identify its bioactive peptides using LC-MS/MS. The findings indicated that TBH significantly increased the time to exhaustion and also had no adverse effects. The liver and skeletal muscle glycogen were significantly increased after administration. TBH induced gene expression that associated with mitochondrial biogenesis. Additionally, the TBH significantly increased the expression of AMPK/SIRT1/PGC-1α signaling proteins. LC-MS/MS analysis identified over 14,000 peptides, with ATFSPYRDRIPLQIVR as the most abundant, and several peptides predicted to exhibit antioxidant and anti-inflammatory activities.

โรคพาร์กินสันเป็นโรคทางระบบประสาทเสื่อมที่พบมากในผู้สูงอายุ ส่งผลให้เกิดความผิดปกติของการเคลื่อนไหว โดยมีสาเหตุหลักจากการเสื่อมของเซลล์ประสาทโดปามิเนอร์จิกในสมอง แม้ว่าพยาธิสภาพของโรคจะเกี่ยวข้องกับการสะสมของโปรตีนผิดปกติ เช่น Lewy bodies แต่ปัจจุบันพบว่าเมแทบอลิซึมของไขมัน โดยเฉพาะกรดไขมันไม่อิ่มตัวเชิงซ้อนก็มีบทบาทสำคัญต่อการเกิดโรค

งานวิจัยที่ผ่านมาแสดงให้เห็นการเปลี่ยนแปลงของไขมันในสมองผู้ป่วยพาร์กินสัน อย่างไรก็ตาม เทคนิคที่ใช้ยังไม่สามารถแสดงการกระจายตัวของไขมันในเนื้อเยื่อได้อย่างชัดเจน ดังนั้น การใช้เทคนิค MALDI-MSI ซึ่งสามารถวิเคราะห์ตำแหน่งของไขมันในเนื้อเยื่อโดยตรง จึงมีความสำคัญในการศึกษาการเปลี่ยนแปลงของไขมันในบริเวณ motor cortex ซึ่งเกี่ยวข้องกับการควบคุมการเคลื่อนไหว

Abstract


Parkinson’s disease (PD) is a neurodegenerative disorder caused by the death of dopaminergic neurons in the substantia nigra pars compacta (SNc). Lipid metabolism, especially phospholipids, has been reported to be altered in PD. The purpose of this study is to investigate the temporal expression and spatial distribution of phospholipids in the motor cortex and striatum at different time points of PD using matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) in a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced parkinsonian mouse model. Mice were injected with saline (NSS) or MPTP at two different time points to create acute and subacute models. Motor analysis was performed at 0, 3, 7, 14, and 21 days post-injection. Tyrosine hydroxylase (TH) staining revealed progressive damage of neurons in the substantia nigra compacta (SNc) and reduced striatal fibers in MPTP-treated animals. By using MALDI-MSI, we identified changes in phosphatidylcholine (PC) profiles in the brains of MPTP-treated animals. Polyunsaturated PCs, including PC 36:4 (16:0/20:4), PC 38:6 (16:0/22:6), and PC 40:8 (18:2/22:6), were decreased in the MPTP-treated groups. These reductions were time-dependent and were more pronounced in the subacute MPTP-treated group. The loss of dopamine neurons caused by MPTP may be associated with the selective loss of polyunsaturated PCs in brain membranes, indicating that lipid metabolism and membrane structural alterations may contribute to the pathology of PD.

Abstract

The pathogenesis of Parkinson’s disease (PD) consists of the apoptosis of dopaminergic neurons in the substantia nigra pars compacta (SNpc) due to oxidative stress. The present study aimed to evaluate the potential antioxidant activity of whey protein isolate (WPI) in PD models, using neurotoxin-exposed SH-SY5Y cells differentiated into dopaminergic-like neurons. Our research shows that WPI’s high glutamic acid, aspartic acid, and leucine contribute to its antioxidant and neuroprotective effects, with glutamic acid crucial for glutathione synthesis. In vitro studies found that WPI, at concentrations of 5–1000 µg/mL, is non-toxic to differentiated SH-SY5Y cells. Notably, the lowest con-centration of WPI (5 µg/mL) significantly decreased intracellular reactive oxygen species (ROS) levels in these cells following a 24 h co-treatment with 1-methyl-4-phenylpyridinium (MPP+). The antioxidant effects of WPI were also confirmed by the increased expression of HO1 and GPx antioxidant enzymes, which are Nrf2 pathway target genes and were evaluated by real-time PCR. Furthermore, Nrf2 nuclear translocation in the differentiated SH-SY5Y cells was also increased when the cells were exposed to 5 µg/mL of WPI with MPP+. These results together suggest that WPI has antioxidant effects on dopaminergic-like neurons in a Parkinson’s disease model.

This study developed hydrolyzed tuna blood powder (HTBP) from canned tuna industry by-products and demonstrated its antioxidant and anti-apoptotic effects on C2C12 muscle cells under oxidative stress. HTBPs, particularly those produced with alcalase and trypsin, reduced ROS and NO levels, suppressed apoptosis-related proteins, and improved cell survival. These findings suggest HTBP as a promising functional ingredient for muscle recovery and sustainable resource utilization.

ปัจจุบันเลือดปลาทูน่าเป็นของเสียจากอุตสาหกรรมที่ยังไม่ได้ใช้ประโยชน์ ทั้งที่อุดมด้วยกรดอะมิโนและแร่ธาตุสำคัญ อีกทั้งยังมีรายงานว่าเปปไทด์จากน้ำต้มปลาทูน่าสามารถยับยั้งเอนไซม์ ACE และกระตุ้นการเกิด apoptosis ในเซลล์มะเร็งเต้านม อย่างไรก็ตาม ยังไม่มีการศึกษาอย่างชัดเจนเกี่ยวกับศักยภาพของไฮโดรไลเซตจากเลือดปลาทูน่าในการเป็นวัตถุดิบเชิงหน้าที่เพื่อการฟื้นฟูกล้ามเนื้อ งานวิจัยนี้จึงมุ่งพัฒนาไฮโดรไลเซตจากเลือดปลาทูน่าเพื่อใช้เป็นส่วนผสมในผลิตภัณฑ์เพื่อสุขภาพกล้ามเนื้อ พร้อมส่งเสริมการใช้ทรัพยากรอย่างคุ้มค่าและยั่งยืน

Abstract

This study aimed to develop hydrolyzed tuna blood powders (HTBP) with potential anti-apoptotic and muscle recovery properties. HTBP was produced alcalase, trypsin, and pepsin at concentrations of 0.5%, 1.0%, and 1.5%. The physical and chemical properties of HTBP were evaluated. The results showed no significant difference in yield (p > 0.05), with a range of 7.08% ± 0.78% to 9.26 % ± 1.74%. HTBP produced with alcalase at all concentrations yielded a higher amount of protein (76.86% ± 0.5%–79.1% ± 1.10%) compared to trypsin and pepsin. Glutamic acid, cysteine, and aspartic acid were identified as the main amino acids in HTBP. The water solubility index of all HTBP was high (>90%). Antioxidant values increased with higher protein content. Hydrolyzed tuna blood with 0.5% alcalase (HTBPA) and 0.5% trypsin were selected for further investigation of their biological activities, assessed using C2C12 muscle cells under oxidative stress conditions induced by hydrogen peroxide (H2O2). In H2O2-induced C2C12 cells, HTBPA and HTBPT reduced nitric oxide (NO) secretion and intracellular reactive oxygen species (ROS) levels. Furthermore, HTBPA and HTBPT significantly decreased the percentage of apoptotic cells in these H2O2-induced C2C12 cells. They also inhibited H2O2-induced apoptosis by modulating apoptosis-related proteins, including NF-κB, Bax, and caspase-3. Additionally, HTBPA and HTBPT suppressed the phosphorylation levels of key signaling proteins such as Akt, ERK1/2, and p38, thereby contributing to enhanced cell survival. These findings suggest that hydrolyzing tuna blood, waste of the canned tuna industry, can create a valuable ingredient for promoting muscle health.

เมื่อวันอังคารที่ 11 มีนาคม พ.ศ. 2568 ภาควิชากายวิภาคศาสตร์ คณะวิทยาศาสตร์ มหาวิทยาลัยมหิดล ได้จัดการบรรยายพิเศษในหัวข้อ “Unraveling the Molecular Mechanisms of Cell Migration: From Tissue Morphogenesis to Cancer Progression” ณ ห้อง AN1-202 ระหว่างเวลา 9.30-10.30 น.

การบรรยายครั้งนี้ได้รับเกียรติจาก ผู้ช่วยศาสตราจารย์ ดร.โคจิ สึทสึมิ (Koji Tsutsumi, Ph.D.) อาจารย์อาวุโสจาก Department of Biosciences, School of Science, Kitasato University ประเทศญี่ปุ่น ซึ่งเป็นนักวิจัยผู้เชี่ยวชาญด้านกลไกระดับโมเลกุลของการเคลื่อนที่ของเซลล์ โดยมีเนื้อหาครอบคลุมบทบาทสำคัญของการเคลื่อนที่ของเซลล์ในกระบวนการทางชีววิทยาต่าง ๆ เช่น การสร้างเนื้อเยื่อ (tissue morphogenesis) และการแพร่กระจายของมะเร็ง (cancer metastasis)

ภายในการบรรยาย รองศาสตราจารย์ ดร.สึทสึมิ ได้อธิบายถึงบทบาทของโครงสร้าง cytoskeleton โดยเฉพาะ แอคตินฟิลาเมนต์ (actin filaments) ที่มีความสำคัญต่อการควบคุมรูปร่างและการเคลื่อนที่ของเซลล์ รวมถึงการตอบสนองต่อสัญญาณภายนอก นอกจากนี้ยังกล่าวถึงโปรตีน FilGAP ซึ่งเป็นตัวควบคุมสำคัญของ actin cytoskeleton และมีบทบาทในการกำหนดทิศทางการเคลื่อนที่ของเซลล์ (front-rear polarity) รวมถึงการรักษาความสมบูรณ์ของเนื้อเยื่อ (epithelial integrity)

การค้นพบล่าสุดของทีมวิจัยพบว่า FilGAP มีบทบาทสำคัญในการเคลื่อนที่ของเซลล์มะเร็งผ่านเส้นทางฟอสโฟรีเลชันที่แตกต่างกัน รวมถึงการควบคุมการเติบโตของสเฟียรอยด์ในมะเร็งสมอง (glioma) โดยมีปฏิสัมพันธ์กับ mTOR pathway ซึ่งเป็นเส้นทางที่แตกต่างจากบทบาทเดิมของ FilGAP ในการควบคุม actin dynamics ผลลัพธ์เหล่านี้ช่วยให้เข้าใจถึงการควบคุมกลไกของเซลล์ต่อสิ่งเร้าในสภาพแวดล้อมที่ซับซ้อน และมีศักยภาพในการพัฒนาเป็นเป้าหมายใหม่ในการรักษามะเร็งและโรคทางพันธุกรรมที่เกี่ยวข้อง

ที่มาและความสำคัญ

Endometriosis หรือเยื่อบุโพรงมดลูกเจริญผิดที่ เป็นภาวะที่มีเซลล์เยื่อบุโพรงมดลูกไปเจริญภายนอกมดลูก ภาวะนี้ส่งผลให้เกิดปัญหาทางสุขภาพของผู้หญิงได้แก่การปวดประจำเดือนรุนแรง ภาวะมีบุตรยากและการกลับเป็นซ้ำหลังการรักษาหรือแม้แต่กระทั่งสามารถกลายเป็นมะเร็งได้ ปัจจุบันยังไม่สามารถหาสาเหตุของการเกิดเยื่อบุโพรงมดลูกเจริญผิดที่ที่แน่ชัดได้ ด้วยเหตุนี้การรักษาเยื่อบุโพรงมดลูกเจริญผิดที่จึงเป็นการรักษาอาการและเพื่อแก้ไขปัญหาของผู้ป่วย เช่น การใช้ยาฮอร์โมนต่าง ๆ เพื่อลดอาการปวด และการผ่าตัดซึ่งสามารถช่วยให้แก้ไขภาวะมีบุตรยาก อย่างไรก็ตามในปัจจุบันภาวะนี้ยังไม่มียาหรือการรักษาที่ได้ผลดีเนื่องจากผู้ป่วยส่วนใหญ่จะมีการกลับมาเป็นซ้ำได้ หลังจากการให้ยาหรือการผ่านำรอยโรคออกมาแล้ว ดังนั้นการตรวจพบผู้ที่มีความเสี่ยงแต่เนิ่น ๆ รวมถึงการจัดการที่เหมาะสมอย่างทันท่วงที จึงเป็นสิ่งที่สำคัญในการป้องกันการเจริญของเยื่อบุโพรงมดลูกเจริญผิดที่ เพราะฉะนั้นการศึกษานี้จึงเป็นการศึกษารูปแบบการปวดประจำเดือน ผลของการใช้ยา และวิเคราะห์ความเสี่ยงในการเป็นเยื่อบุโพรงมดลูกเจริญผิดที่ในกลุ่มนักศึกษาแพทย์และนักศึกษาพยาบาลจากประเทศที่มีรายได้สูง และประเทศที่มีรายได้ปานกลาง

Abstract

Research question

Do patterns of the menstrual cycle, menstrual pain and the use of medication for menstrual pain differ between young women from high-income countries (HIC) and middle-income countries (MIC)?

Design

A multinational, multicentre, cross-sectional study using pen-and-paper questionnaires was conducted between 2016 and 2021 to assess patterns of the menstrual cycle, menstrual pain and the use of medication for menstrual pain. Various parameters were evaluated to identify high-risk factors for severe menstrual pain in women from two HIC (n = 1550) and nine MIC (n = 7139).

Results

From a total of 9114 young women, 4920 medical students (HIC n = 696, MIC n = 4224) and 3769 nursing students (HIC n = 854, MIC n = 2915) were included in this study. Compared with those from HIC, a significantly higher proportion of medical and nursing students from MIC reported cyclic pain (83.9% and 86.8%, respectively) and acyclic pain (33.8% and 31.9%, respectively) (both P < 0.001). Multivariate regression analysis revealed that low body mass index and early onset of menarche were independent risk factors for severe cyclic/acyclic pain among women from HIC, and a family history of menstrual pain was a risk factor for severe cyclic/acyclic pain among women from HIC and MIC.

Conclusions

Differential patterns of the menstrual cycle, menstrual pain and use of medication for menstrual pain were found between young women from HIC and MIC. A proper educational programme may be necessary for these women and healthcare providers to understand the consequences of intractable cyclic/acyclic pain, in order to facilitate early detection and timely management of menstrual pain and its negative consequences, such as endometriosis.

Abstract

Background: Coffee leaves are a major source of bioactive components and are used as ethnomedicine. However, despite their traditional medicinal use, information about their effects on antihyperlipidemia remains limited. Methods: The aims of this study were to evaluate the main components of leaf extracts from Arabica and Robusta coffees and to examine the potential of these coffee leaves in reducing lipid digestion and absorption in vitro. Results: Coffee leaf extracts from Arabica coffee contain a high amount of caffeine, whereas extracts from Robusta coffee contain high amounts of chlorogenic acid (CGA) and caffeine. Additionally, leaf extracts from Arabica and Robusta coffee demonstrated the inhibition of pancreatic lipase, decreased micellar cholesterol solubility, and reduced bile acid binding. Furthermore, these extracts resulted in a reduction in cholesterol uptake in Caco-2 cells. Molecular docking experiments supported this discovery, showing CGA and caffeine binding to Niemann–Pick C1-like 1 (NPC1L1), a key protein in cholesterol absorption. The results indicated that CGA and caffeine can competitively bind to NPC1L1 at the cholesterol binding pocket, reducing its cholesterol binding rate. These findings suggest that coffee leaves might help suppress lipid absorption and digestion, highlighting their potential use in preventing and treating hyperlipidemia.

Abstract

Parkinson’s disease (PD) is classified as one type of neurodegenerative disorder. Movement disorder, which includes resting tremors and slowness of movement, is a common clinical symptom in PD patients. Neuroinflammation is one of the most important processes involved in the pathogenesis of PD. An inflammatory response in the brain can induce neuronal cell death. Microglia, a type of immune cell, plays a crucial role in neuroinflammation. In this review, we discussed the information on microglia-activated neuroinflammation, its relationship with PD, and therapeutic approaches for neuroinflammation in PD. Under normal conditions, microglia in their inactive state (M0) act as surveillance agents in the brain to investigate potential invasions. They regulate neuron production, remodel synapses, and secrete growth factors to protect the neurons. Under pathological conditions, the M0 transforms into active phenotypes, dividing into pro-inflammatory (M1) and anti-inflammatory (M2) microglia. The M1 and M2 microglia exhibit opposite functions, where M1 microglia promote pro-inflammatory responses, and M2 microglia promote anti-inflammatory responses. This dichotomy of functions is essential for maintaining a healthy level of inflammation in the brain. Presently, multiple therapeutic strategies are available for PD, encompassing anti-inflammatory drugs, neuroprotective compounds, antioxidants, nanoparticles targeting neuroinflammation, stem cell interventions, lifestyle adjustments, and microglia-focused treatments. These treatments improve patients’ movement, allowing them to have lifestyles like others, consequently benefiting their mental and emotional well-being. Preventing microglia from polarising into the M1 phenotype and promoting their polarisation into the M2 phenotype could be a challenging and promising approach for treating PD.

ฟอลลิเคิล (follicle) ในรังไข่เป็นโครงสร้างสำคัญที่จำเป็นสำหรับการเจริญเติบโตและการสุกของเซลล์ไข่ ในระหว่างการพัฒนาฟอลลิเคิลของรังไข่ การสร้างเส้นเลือดใหม่มีบทบาทสำคัญในการเพิ่มปริมาณเลือดไปยังฟอลลิเคิลของรังไข่ Vascular epithelial growth factor หรือ VEGF หรือที่เรียกอีกอย่างว่า VEGFA เป็นตัวควบคุมหลักของการสร้างเส้นเลือดใหม่ในรังไข่ VEGFA ประกอบด้วยไอโซฟอร์มหลายชนิด ไอโซฟอร์มที่สำคัญคือ VEGF165 ช่วยทำให้มีกระกระตุ้นให้มีอัตราการเกิด antral formation มากขึ้นและกระตุ้นการผลิตเซลล์ไข่ที่โตเต็มที่ได้ ซึ่ง VEGF165 จะมีตัวยับยั้งการทำงานคือ VEGF165b อย่างไรก็ตาม ยังไม่มีการศึกษากลไกของ VEGF ในการเปลี่ยนจาก VEGF165b เป็น VGF165 ในระหว่างการพัฒนาฟอลลิเคิลของรังไข่ ดังนั้นวัตถุประสงค์ของการวิจัยนี้คือเพื่อตรวจสอบบทบาทหน้าที่ของ VEGF165a, VEGF165b และตัวรับ VEGFR ในการพัฒนาฟอลลิเคิลของรังไข่ของวัว

Abstract

Objective: 

To investigate the role of vascular endothelial growth factor (VEGF)165a, VEGF165b, and VEGF receptor (VEGFR) in the development of bovine follicles.

Methods: 

We cultured follicular cells that were collected from small, medium, and large sized bovine follicles with estrogen and measured the expression of VEGF, VEGFR2 and VEGF165b by Western blot analysis and immunofluorescence.

Results: 

The expression of VEGF165 increased in all follicle sizes and the expression of VEGF165b was increased in the small and large follicles after culturing in an estrogen containing medium. The expression of VEGFR2 was increased in the medium and large follicles after culturing with estrogen for 96 h. VEGF165 was activated at 100 ng/mL estrogen in the large follicles for 96 h. In addition, VEGFR2 was upregulated in the medium and large follicles after treated with 100 ng/mL estrogen for 96 h.

Conclusions: 

This evidence suggests that the expression of VEGF165 and VEGFR is associated with estrogen stimulation during the development of bovine follicles and in an autocrine or paracrine manner. This reveals an advantage during oocyte maturation in vitro.

พยาธิสรีรวิทยาของโรคพาร์กินสันเชื่อมโยงกับความเครียดจากปฏิกิริยาออกซิเดชั่นและการอักเสบในสมองจนกระทั่งทำให้เกิดการตายของเซลล์ประสาทชนิดโดปามิเนอร์จิคในสมองส่วน substantia nigra pars compacta (SNpc) การรักษาโรคพาร์กินสันในปัจจุบันคือการใช้ยาบรรเทาอาการ เช่น ยาเลโวโดปา แต่ยาหลายๆชนิดนี้ยังไม่สามารถรักษาโรคนี้ให้หายขาดได้ นอกจากนี้การใช้ยาในระยะยาวอาจนำไปสู่ผลข้างเคียงได้ ดังนั้นการวิจัยในปัจจุบันจึงเน้นที่จะใช้ผลิตภัณฑ์จากธรรมชาติมาใช้เป็นทางเลือกในการบรรเทาอาการหรือรักษาโรคพาร์กินสันหรือโรคทางระบบประสาทอื่นๆ เนื่องจากอาจจะไม่ก่อให้เกิดผลข้างเคียงหรือเกิดผลข้างเคียงได้น้อย ในระยะหลังๆนี้มีงานวิจัยหลายชิ้นรายงานว่า โปรตีนไฮโดรไลเสตจากปลามีสารต้านอนุมูลอิสระ และคุณสมบัติต่อต้านการอักเสบและลดตายของเซลล์หลายชนิด ซึ่งรวมถึงเซลล์ประสาท ดังนั้นการศึกษานี้มีวัตถุประสงค์เพื่อตรวจสอบฤทธิ์ต้านอนุมูลอิสระและฤทธิ์ต้านการอักเสบของเลือดปาทูน่าเลือดไฮโดรไลเซท (TBH) ในการบรรเทาการตายของเซลล์ประสาทนิวโรบลาสโตมาของมนุษย์ (SH-SY5Y) ที่ได้รับการกระตุ้นการเปลี่ยนแปลงไปเป็นเซลล์ประสาทที่คล้ายเซลล์ประสาทชนิดโดปามิเนอร์จิก (differentiated SH-SY5Y) ในแง่ของการลดการตายของเซลล์ โดยลดอนุมูลอิสระและลดการอักเสบภายในเซลล์

Abstract

The pathophysiology of Parkinson’s disease (PD) is linked to oxidative stress and inflammatory-induced cell death of dopaminergic neurons in substantia nigra pars compacta (SNpc). The current study aims to investigate the effects of tuna blood hydrolysate (TBH) on the neurodegeneration of Parkinson-like disease models using neurotoxin-induced SH-SY5Y cells differentiated to be dopaminergic-like neurons. It was found that TBH at concentrations ranging from 5 to 250 µg/ml can reduce differentiated SH-SY5Y cell death after exposure to 1-methyl-4-phenylpyridinium (MPP+) co-treated with TBH for 24 h. Real-time PCR results indicated that TBH enhanced the expressions of NAD(P)H quinone dehydrogenase 1 (NQO1), catalase (CAT), and thioredoxin reductase (TrxR1), which are Nrf2 downstream antioxidant genes. Bioinformatic analysis and molecular docking revealed that the peptide found in TBH, IPGQPGLPGPPGPPGPPGLG, could penetrate the cell and blood–brain barrier and strongly bind the Keap1-Kelch domain, causing Nrf2 nuclear translocation. Consequently, the upregulation of antioxidant genes ameliorates ROS and reduces dopaminergic neuron death. Another, TBH can reduce differentiated SH-SY5Y cell death and intracellular ROS after exposure to 24 h of co-treatment between TBH and tumor necrosis factor-alpha (TNF-α). Western blot analysis was performed to investigate apoptosis-related proteins. Bcl2 and BDNF are dramatically increased, while BAX, cytochrome c, caspase3, caspase9, and cleaved caspase9 are significantly decreased in co-treating differentiated SH-SY5Y cells with TBH and TNF-α. These findings demonstrated that TBH can suppress the apoptosis of inflammatory differentiated SH-SY5Y cells. Taken together, this study suggests that TBH can alleviate neurodegeneration and has anti-inflammatory effects on dopaminergic-like neurons in the simulation of a PD condition.
