Abstract

Tilapia tilapinevirus or tilapia lake virus (TiLV) is an emerging virus that inflicts significant mortality on farmed tilapia globally. Previous studies reported detection of the virus in multiple organs of the infected fish; however, little is known about the in-depth localization of the virus in the central nervous system. Herein, we determined the distribution of TiLV in the entire brain of experimentally infected Nile tilapia. In situ hybridization (ISH) using TiLV-specific probes revealed that the virus was broadly distributed throughout the brain. The strongest positive signals were dominantly detected in the forebrain (responsible for learning, appetitive behaviour and attention) and the hindbrain (involved in controlling locomotion and basal physiology). The permissive cell zones for viral infection were observed mostly to be along the blood vessels and the ventricles. This indicates that the virus may productively enter into the brain through the circulatory system and widen broad regions, possibly through the cerebrospinal fluid along the ventricles, and subsequently induce the brain dysfunction. Understanding the pattern of viral localization in the brain may help elucidate the neurological disorders of the diseased fish. This study revealed the distribution of TiLV in the whole infected brain, providing new insights into fish–virus interactions and neuropathogenesis.

Abstract

Betulinic acid (BA) is a pentacyclic triterpene usually isolated from botanical sources. Numerous studies have reported the inhibitory effect of BA against human colorectal cancer cells (CRC). However, its effect on the expression of the molecular chaperone HSPA is unclear. The aim of this research is to investigate the anti-cancer activities of BA purified from Piper retrofractum and study its effect on the expression of HSPA in colorectal cancer HCT116 and SW480 cells. The viability of both cancer cells was reduced after they were treated with an increasing dosage of BA. Flow cytometry assay revealed that levels of cell apoptosis significantly increased after incubation with BA in both cancer cells. Pro-apoptotic markers including Bax, cleaved-caspase-3 and cleaved-caspase-9 were increased while anti-apoptotic marker Bcl-2 was decreased after BA treatment. Western blot also showed that the expression of HSPA fluctuated upon BA treatment, whereby HSPA was increased at lower BA concentrations while at higher BA concentrations HSPA expression was decreased. Preliminary molecular docking assay showed that BA can bind to the nucleotide binding domain of the HSP70 at its ADP-bound state of the HSP70. Although further research is needed to comprehend the BA-HSPA interaction, our findings indicate that BA can be considered as potential candidate for the development of new treatment for colorectal cancer.

Betulinic acid (BA) เป็นสารกลุ่มไตรเตอร์ปีนที่ถูกสกัดจากพืช โดยมีคุณสมบัติเป็นสารที่ต้านมะเร็งในลำไส้ใหญ่ในระดับเซลล์ แต่ยังไม่ทราบเกี่ยวกับกลไกที่เกี่ยวข้องกับโปรตีน heat shock proteins (HSPs) ในการศึกษาวิจัยนี้จึงมุ่งเน้นศึกษาประโยชน์ของสารสกัดโดยงานวิจัยนี้พบว่า สาร BA สามารถยับยั้งการมีชีวิตของเซลล์มะเร็งลำไส้ใหญ่ทั้งสองชนิด HCT116 และ SW480 โดยขึ้นกับความเข้มข้นของสาร BA ผ่านกระบวนการชักนำให้เกิดการตายแบบ apoptosis โดยพบการแสดงออกของ Bax, cleaved-caspase-3 และ cleaved-caspase-9 ที่เพิ่มขึ้น และลดการแสดงออกของ Bcl-2 รวมทั้งพบการแสดงออกของ HSPA ที่จะลดลง ซึ่งสอดคล้องกับการศึกษาทางคอมพิวเตอร์ในเบื้องต้น พบว่า สาร BA สามารถจับกับตำแหน่งของ nucleotide binding domain ของ HSP70 แต่อย่างไรก็ตามคณะผู้วิจัยยังต้องศึกษารายละเอียดเพิ่มเติมของปฏิกิริยาที่เกิดขึ้นระหว่างสาร BA และ HSPA นี้ โดยคาดว่าการศึกษานี้อาจนำไปสู่การพัฒนาการรักษาโรคมะเร็งลำไส้ใหญ่ได้

Abstract

Possessing the quinone moiety, ilimaquinone (1), a sponge–derived sesquiterpene quinone, has been hypothesised to express its cytotoxicity through a redox cycling process, yielding active product(s) that can cause DNA damage. To determine the DNA damaging effects of 1 and examine whether a redox transformation may participate in its functions, the DNA damaging properties of 1, the corresponding hydroquinone (2) and hydroquinone triacetates (3) and their 5-epimeric counterparts (4–6) were tested and compared. When incubated directly with plasmid DNA, the hydroquinones were the only active species capable of cleaving the DNA. In cell-based assays, however, the quinones and hydroquinone triacetates were active in the same range as that of the corresponding hydroquinones, and all damaged the cellular DNA in a similar manner. The in situ reduction of 1 and 4 were supported by the decreases in the cytotoxicity when cells were pre-exposed to dicoumarol, an NAD(P)H:quinone oxidoreductase 1 (NQO1) inhibitor. The results confirmed the DNA damaging activities of the ilimaquinones 1 and 4, and indicated the necessity to undergo an in-situ transformation into the active hydroquinones, thereby exerting the DNA damaging properties as parts of the cytotoxic mechanisms.

ปัจจุบันการสแกนภาพแบบสามมิติ เป็นเทคโนโลยีที่มีการนำไปใช้อย่างแพร่หลาย โดยเฉพาะในด้านการศึกษา ที่ผ่านมาได้มีการนำเทคโนโลยีการสแกนภาพสามมิติมาใช้จัดทำเป็นสื่อประกอบการเรียนการสอนกายวิภาคศาสตร์ และพบว่าส่งผลให้ผู้เรียนสามารถจดจำเนื้อหาและระบุตำแหน่งของโครงสร้างต่าง ๆ ได้ดียิ่งขึ้น ซึ่งนอกจากเป็นการเพิ่มประสิทธิภาพให้กับการเรียนการสอนแล้วยังเป็นประโยชน์อย่างยิ่งสำหรับสถาบันการศึกษาที่กำลังประสบปัญหาขาดแคลนร่างอาจารย์ใหญ่หรือไม่สามารถดำเนินการจัดการเรียนการสอนแบบ on-site ได้ในช่วงสภาวะการระบาดของโรคโควิด-19 นี้ อย่างไรก็ตาม การนำเทคโนโลยีสแกนภาพสามมิติมาใช้ในเชิงวิจัยยังมีไม่มากนัก โดยเฉพาะอย่างยิ่งการนำภาพสแกนสามมมิติมาใช้เพื่อเป็นเครื่องมือในการนำเสนอผลงานวิจัยทางกายวิภาคศาสตร์ ยังไม่เคยพบว่ามีการนำมาใช้ ซึ่งปกติงานวิจัยทางด้านกายวิภาคศาสตร์จะใช้การนำเสนอ รายงานผล แบบรูปภาพสองมิติ (2D) หรือมีการวาดภาพประกอบร่วมด้วย ดังนั้น ในงานวิจัยฉบับนี้ ผู้วิจัยได้ทดลองการนำเสนอรายงานผลการวิจัย โดยใช้เครื่องสแกนภาพแบบสามมิติ (3D scanner) เพื่อสแกนแขนท่อนล่างของร่างอาจารย์ใหญ่เพื่อนำมารายงานโครงสร้างแปรผันทางกายวิภาค ซึ่งเป็นเส้นเอ็นที่เชื่อมระหว่างกล้ามเนื้อ flexor pollicis longus และกล้ามเนื้อ flexor digitorum profundus ของนิ้วชี้ (a reverse form of Linburg–Comstock variation) และนำไปสร้างเป็นไฟล์ 3D-PDF โดยผู้อ่านสามารถหมุน ขยาย และมีปฏิสัมพันธ์กับโครงสร้างนั้น ๆ ได้ ในลักษณะเดียวกับการปฏิบัติในห้องปฏิบัติการ ด้วยจุดเด่นเหล่านี้ผู้วิจัยเล็งเห็นว่าการรายงานผลงานวิจัยในรูปแบบสามมิติอาจกลายเป็นบรรทัดฐานใหม่ของการนำเสนอข้อมูลทางกายวิภาคศาสตร์ในอนาคตอันใกล้นี้ นอกจากนี้ ผู้วิจัยยังได้เสนอสมมติฐานซึ่งอธิบายกลไกลการเกิดขึ้นของโครงสร้างนี้ในเชิงวิวัฒนาการและวิเคราะห์ความสำคัญทางคลินิกของโครงสร้างนี้อีกด้วย

Abstract

Purpose

Two most common variations of flexor pollicis longus include its accessory head and its connection with the flexor digitorum profundus of the index (Linburg–Comstock variation). In addition, while three-dimensional (3D) screening has widely been used in anatomical education, its use as reporting tool in anatomical research is still limited. The objective of this study is to report a previously unrecognized form of the accessory head of flexor pollicis longus, discuss the potential etiology of Linburg–Comstock variation, and pilot the 3D scanning of a large-scale anatomical structure.

Methods

An unusual tendon slip was discovered during a routine dissection in the anterior compartment of the right forearm of a 54-year-old male cadaver. A 3D scanner was used to capture the surface topography of the specimen and an interactive portable document format (PDF) was created.

Results

An anomalous tendon was found originating from the lateral aspect of the flexor digitorum profundus muscle. This variant tendon then inserted onto the medial surface of the flexor pollicis longus tendon before entering the carpal tunnel. The variation resembles a reverse form of Linburg–Comstock variation, because pulling this variant tendon resulted in simultaneous flexion of the interphalangeal joint of thumb.

Conclusion

Surgeons should be aware of the reverse Linburg–Comstock variation, because it may not be detectable by the conventional provocative testing. Linburg–Comstock variation may be classified as an anatomical variant or a secondarily acquired condition depending on its type. Our demonstration of interactive 3D-PDF file highlights its potential use for delivering anatomical information in future cadaveric studies.

เพื่อให้ความรู้ทางด้านกายวิภาคศาสตร์พื้นฐานแก่นักเรียนจากโรงเรียนต่าง ๆ ทั่วประเทศเป็นการสร้างแรงบันดาลใจและประชาสัมพันธ์หลักสูตรการศึกษา คณะต่าง ๆ ของมหาวิทยาลัยมหิดล อีกทั้งยังส่งเสริมให้นักศึกษา และบุคลากรได้ทำกิจกรรมร่วมกัน ผลลัพธ์ที่นำไปใช้ประโยชน์จากกิจกรรม นักเรียนมีความรู้ความเข้าใจในร่างกายมนุษย์มากขึ้นกว่าเห็นรูปในหนังสือ มีส่วนช่วยให้อยากศึกษาต่อทางด้านการแพทย์และวิทยาศาสตร์สุขภาพ ซึ่งในงานนี้ได้รับความสนใจจากนักเรียน นักศึกษา เป็นอย่างมาก

ภาควิชากายวิภาคศาสตร์ได้จัดกิจกรรมแสดงความกตัญญูที่ศิษย์พึงปฏิบัติต่ออาจารย์ใหญ่ ส่งเสริมจริยธรรมและปฏิบัติบูชาเพื่ออุทิศบุญกุศลแด่ท่านอาจารย์ใหญ่ซึ่งเป็นเสมือนต้นแบบของความเสียสละร่างของท่านเพื่อการนำไปใช้ให้เกิดประโยชน์สูงสุด อันจะกระตุ้นให้นักศึกษาทุกคนได้ตระหนัก มีจรรยาบรรณ และทำประโยชน์เพื่อส่วนรวมต่อไป

ไนไตร์ท (NO2) เป็นตัวชี้วัดการทำงานของเซลล์บุผนังหลอดเลือด ไนไตรท์สามารถผลิตได้จากไนเตรต (NO-3) ที่หลั่งจากพลาสมาสู่น้ำลาย ไนเตรตรีดักเตสของแบคทีเรียในช่องปากจะเปลี่ยนไนเตรตในน้ำลายเป็นไนไตรต์ ซึ่งถูกกลืนกินและดูดซึมเข้าสู่กระแสเลือด การศึกษานี้จึงมีวัตถุประสงค์เพื่อศึกษาว่า ระดับของไนไตร์ทในน้ำลายมีความสอดคล้องกับระดับของไนไตร์ทในเลือด ไนไตร์ทในน้ำลายจึงสามารถใช้เป็นตัวชี้วัดการทำงานของเซลล์บุผนังหลอดเลือดได้เช่นกัน โดยระดับของไนไตร์ทในเลือด และในน้ำลายถูกวัดโดยเครื่องวัดการเรืองแสงทางเคมีสำหรับวัดระดับไนตริคออกไซด์ เพื่อศึกษาผลของการน้ำยาบ้วนปากที่มีผลในการกำจัดหรือลดปริมาณของแบคทีเรียในช่องปากที่สามารถเปลี่ยนไนเตรทในน้ำลายไปเป็นไนไตร์ทได้ ในการศึกษานี้ เรามีวัตถุประสงค์เพื่อตรวจสอบความเกี่ยวข้องระหว่างระดับน้ำลายและระดับเลือดของสายพันธุ์เหล่านี้ เราเก็บตัวอย่างน้ำลายทั้งหมด 3 ตัวอย่าง (น้ำลายกระตุ้น กระตุ้นพาราฟิน และน้ำยาบ้วนปากหลังการใช้คลอเฮกซิดีน) และเลือดจากอาสาสมัครที่มีสุขภาพดี 75 คน เราวัดไนไตรต์และไนเตรตด้วยวิธีเคมีลูมิเนสเซนซ์ ระดับไนไตรท์ในน้ำลายที่ถูกกระตุ้นและน้ำลายที่ถูกกระตุ้นหลังการบ้วนปากมีความสัมพันธ์ที่อ่อนแอกับไนไตรท์ในเลือด ไม่มีความสัมพันธ์ระหว่างไนไตรท์ในน้ำลายที่ไม่ถูกกระตุ้นกับไนไตรท์ในเลือด กิจกรรมของเกล็ดเลือดที่เส้นฐาน ซึ่งพิจารณาจากการแสดงออกของ P-selectin มีความสัมพันธ์เชิงลบกับไนไตรต์ในพลาสมาและน้ำลายที่ถูกกระตุ้นหลังการบ้วนปาก น้ำลายไนเตรตในตัวอย่างน้ำลายทั้งหมดมีความสัมพันธ์กับระดับพลาสมา โดยสรุป ไนไตรท์ในน้ำลายที่ถูกกระตุ้นสัมพันธ์กับระดับในพลาสมาและเลือดครบส่วน หลังการบ้วนปาก ไนไตรท์ในน้ำลายที่ถูกกระตุ้นจะมีความสัมพันธ์ผกผันกับการทำงานของเกล็ดเลือด ไนเตรตในพลาสมาสัมพันธ์กับตัวอย่างน้ำลาย

Abstract

Nitrite anion (NO2−) is a circulating nitric oxide (NO) metabolite considered an endothelial function marker. Nitrite can be produced from nitrate (NO3−) secreted from plasma into saliva. The nitrate reductase of oral bacteria converts salivary nitrate to nitrite, which is swallowed and absorbed into circulation. In this study, we aimed to examine the relevance between these species’ salivary and blood levels. We collected three whole saliva samples (unstimulated, paraffin-stimulated, and post-chlorhexidine mouthwash stimulated saliva) and blood from 75 healthy volunteers. We measured the nitrite and nitrate by the chemiluminescence method. The nitrite levels in stimulated saliva and post-mouthwash stimulated saliva exhibited weak correlations with blood nitrite. There was no correlation between nitrite in unstimulated saliva with blood nitrite. The baseline platelet activity, determined as P-selectin expression, negatively correlated with nitrite in plasma and post-mouthwash stimulated saliva. The salivary nitrate in all saliva samples showed correlations with its plasma levels. We conclude that nitrite in stimulated saliva correlates with blood nitrite.

Abstract

Background

Mature lysostaphin (~28-kDa Lss) from Staphylococcus simulans proves effective in killing methicillin-resistant Staphylococcus aureus (MRSA) which is endemic in hospitals worldwide. Lss is Zn2+-dependent endopeptidase, but its bacteriolytic activity could be affected by exogenously added Zn2+.

Objective

To gain greater insights into structural and functional impacts of Zn2+and Ni2+on Lss-induced bioactivity.

Methods

Lss purified via immobilized metal ion-affinity chromatography was assessed for bioactivity using turbidity reduction assays. Conformational change of metal ion-treated Lss was examined by circular dichroism and intrinsic fluorescence spectroscopy. Co-sedimentation assay was performed to study interactions between Zn2+-treated Lss and S. aureus peptidoglycans. Metal ionbinding prediction and intermolecular docking were used to locate an extraneous Zn2+-binding site.

Results

A drastic decrease in Lss bioactivity against S. aureus and MRSA was revealed only when treated with Zn2+, but not Ni2+, albeit no negative effect of diethyldithiocarbamate—Zn2+-chelator on Lss-induced bioactivity. No severe conformational change was observed for Lss incubated with exogenous Zn2+ or Ni2+. Lss pre-treated with Zn2+ efficiently bound to S. aureus cell-wall peptidoglycans, suggesting non-interfering effect of exogenous metal ions on cell-wall targeting (CWT) activity. In silico analysis revealed that exogenous Zn2+, but not Ni2+, preferably interacted with a potential extraneous Zn2+-binding site (His253, Glu318 and His323) placed near the Zn2+-coordinating Lssactive site within the catalytic (CAT) domain.

Conclusions

Our present data signify the adverse influence of exogenous Zn2+ ions on Lss-induced staphylolytic activity through the exclusive presence within the CAT domain of an extraneous inhibitory Zn2+-binding site, without affecting the CWT activity.

ยาเม็ดคุมกำเนิดแบบผสม (COCs) สามารถลดการแทรกซึมของเซลล์ macrophage ในเยื่อบุโพรงมดลูก รวมทั้งช่วยควบคุมภาวะสมดุลของเซลล์ระบบภูมิคุ้มกันโดยการเพิ่มจำนวน natural killer cells และ regulatory T cells ในบริเวณเยื่อบุโพรงมดลูกที่เจริญผิดที่ด้วย

ที่มาและความสำคัญ

ความผิดปกติของระบบภูมิคุ้มกันสามารถทำให้มีการเกิดและพัฒนาของเยื่อบุโพรงมดลูกเจริญผิดที่ได้ ในกรณีที่มีการไหลย้อนของประจำเดือนบางส่วนเข้าไปในท่อนำไข่ เมื่อระบบภูมิคุ้มกันไม่สามารถกำจัดเลือดและเนื้อเยื่อที่อยู่ในประจำเดือนเหล่านี้ได้หมด จะทำให้เยื่อบุโพรงมดลูกที่เหลืออยู่ยึดเกาะกับผนังในอุ้งเชิงกรานและเจริญต่อไปได้ ซึ่งเยื่อบุโพรงมดลูกที่เจริญผิดที่นี้จะไปกระตุ้นระบบภูมิคุ้มกันทำให้เกิดการกระตุ้นการตอบสนองการอักเสบที่นำไปสู่การหลั่งไซโตไคน์ต่างๆ ในเนื้อเยื่อ และในเลือด รวมทั้งการแทรกซึมของเซลล์เม็ดเลือดขาว เช่น macrophage และ natural killer cells การให้ Progestin และยาเม็ดคุมกำเนิดแบบผสม (combined oral contraceptives, COCs) เป็นวิธีรักษาเริ่มแรกเมื่อมีอาการปวด โดยยาเม็ดคุมกำเนิดแบบผสมเป็นที่นิยมเนื่องจากมีประสิทธิภาพสูง ผลข้างเคียงต่ำ และราคาไม่แพง ยาชนิดนี้จะไปมีผลยับยั้งการหลั่งฮอร์โมน gonadotropins ทำให้เซลล์ในเยื่อบุโพรงมดลูกอยู่ในระยะ decidualization นอกจากนี้ยาเม็ดคุมกำเนิดแบบผสมยังช่วยลดการเพิ่มจำนวนของเซลล์เยื่อบุโพรงมดลูกที่เจริญผิดที่อีกด้วย อย่างไรก็ดียังไม่มีผู้ใดศึกษาผลของยานี้ต่อเซลล์ในระบบภูมิคุ้มกันในเนื้อเยื่อบุโพรงมดลูก ดังนั้นการศึกษาครั้งนี้มีวัตถุประสงค์เพื่อเพื่อประเมินผลของยาเม็ดคุมกำเนิดแบบผสม (COCs) ที่ประกอบด้วย ethinyl estradiol และ desogestrel ในการแทรกซึมของเซลล์ macrophage และ natural killer cells ในเยื่อบุโพรงมดลูก รวมทั้งศึกษาความหนาแน่นของ regulatory T cells ซึ่งเป็นเซลล์ที่ช่วยควบคุมภาวะสมดุลของเซลล์ระบบภูมิคุ้มกัน

Abstract

Background

Dysregulation of immune response is associated with development of endometriosis. The study aim was to evaluate effect of combined oral contraceptive pills (COCs) consisting of ethinyl estradiol (EE) and desogestrel on the expression of macrophage, natural killer cells, and regulatory T cells of ovarian endometriotic cysts.

Methods

Endometriotic cyst wall tissues were collected from women with endometriosis who were treated (n = 22) with COCs (one table per day of EE 0.03 mg and desogestrel 0.15 mg administered for 28 to 35 days before surgery) or untreated (n = 22). The tissues were collected from endometriotic cyst wall during laparoscopic or laparotomy ovarian cystectomy. Immunohistochemistry for anti-CD68, anti-CD56, and anti-forkhead–winged helix transcription factor (FoxP3), a marker for macrophages, natural killer cells, and regulatory T cells, respectively, were investigated.

Results

The median (interquartile range [IQR]) number of anti-CD68 positive cells in the COC group was significantly lower than in the untreated group (12.7; 4.9–19.3) versus 45.7 (26.0–70.7), p < 0.001). Tissue infiltration of anti-CD56 positive cells in endometriotic cyst was significantly higher after the treatment when compared with tissue from untreated group (42.9, 27.4–68.9 versus 25.3 (14.1–37.3; p = 0.009). The number of regulatory T cells was also significantly increased in the COC group (6.3, 2.8–15.5) versus 0 (0–1.8; p < 0.001).

Conclusions

The effects of COC, containing EE 0.30 mg with desogestrel 0.15 mg, on the immune system was demonstrated by a significant decrease in the number of macrophages and an increase in natural killer and regulatory T cells.

ฤทธิ์ของ Frondoside A และ Ginsenoside Rg3 ในการฟื้นฟูเซลล์ประสาทจากโรคพาร์กินสันในสัตว์ทดลองต้นแบบ Caenorhabditis elegans

ที่มาและความสำคัญ

โรคพาร์กินสันเกิดจากการสูญเสียเซลล์ประสาทสร้างโดปามีนและการสะสมของโปรตีนแอลฟ่าซิลนิวคลิอินในเซลล์ โดยในปัจจุบันยังไม่มีการรักษาโรคพาร์กินสันอย่างมีประสิทธิภาพ และการรักษาที่มีอยู่เป็นเพียงการประคับประคองและรักษาตามอาการที่เกิดขึ้นเท่านั้น ซึ่งมีผลการศึกษาที่ผ่านมาพบว่าสารสกัดซาโปนินมีผลในการป้องกันเซลล์ประสาทในโรคความเสื่อมของระบบประสาทได้ ซึ่งสารสกัดซาโปนินนี้สามารถพบได้ในโสมและปลิงทะเล แต่ยังไม่มีการศึกษาที่ชัดเจนถึงกลไกดังกล่าว ในการศึกษาวิจัยนี้จึงได้ทำการศึกษาการออกฤทธิ์ป้องกันเซลล์ประสาทในการต้านโรคพาร์กินสันของสารสกัด frondoside A จากปลิงทะเล และ ginsenoside Rg3 จากโสม ในสัตว์ทดลองต้นแบบ C. elegans โดยผลการทดลองพบว่าสารสกัด frondoside A และ ginsenoside Rg3 สามารถลดการทำลายของเซลล์ประสาทสร้างโดปามีน และฟื้นฟูพฤติกรรมการหาอาหารซึ่งเป็นหน้าที่หลักของสารสื่อประสาทโดปามีนใน C. elegans รวมถึงการยืดอายุขัยเฉลี่ยของ C. elegans ได้ ซึ่งกลไกนี้เกิดขึ้นจากการลดกระบวนการ apoptosis และการเพิ่มขึ้นของยีนต้านอนุมูลอิสระ นอกจากนี้สารสกัด frondoside A ยังสามารถลดการเกาะกลุ่มกันของโปรตีนแอลฟ่าซิลนิวคลิอิน โดยกระตุ้นผ่านทางกลไกการย่อยโปรตีนที่ผิดปกติภายในเซลล์ ดังนั้นงานวิจัยนี้จึงแสดงให้เห็นถึงประสิทธิภาพของสารสกัด frondoside A และ ginsenoside Rg3 ในการฟื้นฟูเซลล์ประสาทจากการถูกทำลาย และลดการเกาะกลุ่มของโปรตีนแอลฟ่าซิลนิวคลิอิน ซึ่งจะนำไปสู่การใช้ประโยชน์ของสารสกัดในการป้องกันรักษาโรคพาร์กินสันต่อไป

Abstract

Parkinson’s disease (PD) is a currently incurable neurodegenerative disorder characterized by the loss of dopaminergic (DAergic) neurons in the substantia nigra pars compacta and α-synuclein aggregation. Accumulated evidence indicates that the saponins, especially from ginseng, have neuroprotective effects against neurodegenerative disorders. Interestingly, saponin can also be found in marine organisms such as the sea cucumber, but little is known about its effect in neurodegenerative disease, including PD. In this study, we investigated the anti-Parkinson effects of frondoside A (FA) from Cucumaria frondosa and ginsenoside Rg3 (Rg3) from Panax notoginseng in C. elegans PD model. Both saponins were tested for toxicity and optimal concentration by food clearance assay and used to treat 6-OHDA-induced BZ555 and transgenic α-synuclein NL5901 strains in C. elegans. Treatment with FA and Rg3 significantly attenuated DAergic neurodegeneration induced by 6-OHDA in BZ555 strain, improved basal slowing rate, and prolonged lifespan in the 6-OHDA-induced wild-type strain with downregulation of the apoptosis mediators, egl-1 and ced-3, and upregulation of sod-3 and cat-2. Interestingly, only FA reduced α-synuclein aggregation, rescued lifespan in NL5901, and upregulated the protein degradation regulators, including ubh-4hsf-1hsp-16.1 and hsp-16.2. This study indicates that both FA and Rg3 possess beneficial effects in rescuing DAergic neurodegeneration in the 6-OHDA-induced C. elegans model through suppressing apoptosis mediators and stimulating antioxidant enzymes. In addition, FA could attenuate α-synuclein aggregation through the protein degradation process.

ปัจจุบัน ได้มีหลักฐานของการใช้บรรจุภัณฑ์นาโนที่บรรจุสารพันธุกรรมออกฤทธิ์ (dsRNA) เพื่อลดการติดเชื้อไวรัสในกุ้งที่สามารถหวังผลสัมฤทธิ์ได้สูง ในการศึกษาครั้งนี้ เราจึงทดสอบการใช้บรรจุภัณฑ์นาโนเพื่อบรรจุสารพันธุกรรมออกฤทธิ์ 2 ชนิดพร้อมกัน (Co-encapsulation) คือ สารพันธุกรรมออกฤทธิ์ต่อโปรตีน VP28 และ VP37 ของไวรัสก่อโรคตัวแดงดวงขาวในกุ้ง (WSSV) จำนวนอย่างละ 5 ไมโครกรัม เพื่อทดสอบความสามารถในการลดปริมาณเชื้อไวรัสในกุ้ง ผลการทดลองพบว่า สารพันธุกรรมออกฤทธิ์ต่อโปรตีน VP37 สามารถบรรจุเข้าไปในช่องว่างของบรรจุภัณฑ์นาโนได้มากกว่าสารพันธุกรรมออกฤทธิ์ต่อโปรตีน VP28 และให้ผลในการป้องกันเชื้อไวรัส WSSV ได้ดีกว่าการบรรจุสารพันธุกรรมออกฤทธิ์เพียงชนิดเดียว โดยทดสอบจากปริมาณของยีนและจำนวนตัวไวรัส WSSV ที่หลงเหลืออยู่ในเนื้อเยื่อเหงือกของกุ้ง นอกจากนี้ยังช่วยเพิ่มปริมาณของเม็ดเลือดขาว และสารต่อต้านเชื้อโรคชนิดโปรอ็อกซิเดส (Pro-oxidase, PO) และกระตุ้นการแสดงออกของยีน PrePO ในเนื้อเยื่อของกุ้งให้สูงขึ้น โดยมีผลในการต่อสู้กับเชื้อไวรัส ทำให้ลดอัตราการตายของกุ้งได้อย่างมีประสิทธิภาพมากกว่าการใช้บรรจุภัณฑ์นาโนที่บรรจุสารพันธุกรรมออกฤทธิ์เพียงชนิดเดียว ผลการทดลองนี้น่าจะนำไปสู่กรรมวิธีในการออกแบบผลิตภัณฑ์ที่ใช้ในการป้องกันการติดเชื้อไวรัสในเชิงเพาะเลี้ยงต่อไป

Abstract

Accumulative evidence of using double stranded (ds) RNA encapsulated into virus like particle (VLP) nanocarrier has open feasibility to fight against shrimp viral infection in aquaculture field. In this study, we co-encapsulated VP37 and VP28 dsRNA into hypodermal and hematopoietic necrosis virus (IHHNV) like particle and investigated its protection against white spot syndrome virus (WSSV). Five micrograms of each dsRNA were used as starting materials to load into VLP, while the loading efficiency was slightly different, i.e, VP37 dsRNA had somewhat a better load into VLP’s cavity. It was apparent that co-encapsulation of dual dsRNA showed a superior WSSV silencing ability than the single dsRNA counterpart as evidence by the lower WSSV gene expression and its copy number in the gill tissues. Besides, we also demonstrated that co-encapsulated dual dsRNA into IHHNV-VLP stimulated the increased number of hemocytes and the corresponding PO activity as well as up-regulated proPO gene expression in hemocytes to resist viral invasion after an acute stage of WSSV infection. This synergistic action of dual dsRNA encapsulated into IHHNV-VLPs could thus act to delay time of shrimp death and reduced shrimp cumulative mortality greater than the single, naked dsRNA treatment and positive control groups. The obtaining results would encourage the feasibility to use it as a new weapon to fight WSSV infection in shrimp aquaculture.
