Abstract

Cholangiocarcinoma (CCA) is an aggressive malignancy with limited treatment options. EGFR overexpression is associated with tumor recurrence and poor prognosis, yet current EGFR-targeted therapies show limited efficacy. To identify alternative therapeutic candidates, we performed subtractive bio-panning of a naïve human single-chain variable fragment (scFv) phage display library against CCA cell lysates. Three novel scFv antibodies-E1, G8, and H2-were selected based on preferential binding to CCA cells with minimal cross-reactivity to unrelated cancers and normal fibroblasts. Surface plasmon resonance and kinase inhibition assays demonstrated that G8 and H2 bound the EGFR tyrosine kinase (EGFR-TK) domain with nanomolar affinities and suppressed kinase activity, whereas E1 showed weak binding and no kinase inhibition. All three scFvs exhibited efficient internalization into EGFR-overexpressing HuCCA-1 cells. Functional analyses revealed distinct effects on cell growth: G8 and H2 reduced EGFR phosphorylation, decreased cell viability, and induced apoptosis accompanied by S/G₂-M phase accumulation, whereas E1 primarily inhibited proliferation through G₁-phase arrest without significantly affecting EGFR phosphorylation. Molecular docking predicted that G8 interacts near the ATP-binding pocket and H2 at the dimerization interface of EGFR-TK. These interactions may contribute to EGFR inhibition in HuCCA-1 cells. These findings suggest that G8 and H2 function as intracellular inhibitors of EGFR-TK activity and are promising candidates for antibody-based therapy in EGFR-driven CCA.

Abstract

This study aimed to develop hydrolyzed tuna blood powders (HTBP) with potential anti-apoptotic and muscle recovery properties. HTBP was produced alcalase, trypsin, and pepsin at concentrations of 0.5%, 1.0%, and 1.5%. The physical and chemical properties of HTBP were evaluated. The results showed no significant difference in yield (p > 0.05), with a range of 7.08% ± 0.78% to 9.26 % ± 1.74%. HTBP produced with alcalase at all concentrations yielded a higher amount of protein (76.86% ± 0.5%–79.1% ± 1.10%) compared to trypsin and pepsin. Glutamic acid, cysteine, and aspartic acid were identified as the main amino acids in HTBP. The water solubility index of all HTBP was high (>90%). Antioxidant values increased with higher protein content. Hydrolyzed tuna blood with 0.5% alcalase (HTBPA) and 0.5% trypsin were selected for further investigation of their biological activities, assessed using C2C12 muscle cells under oxidative stress conditions induced by hydrogen peroxide (H2O2). In H2O2-induced C2C12 cells, HTBPA and HTBPT reduced nitric oxide (NO) secretion and intracellular reactive oxygen species (ROS) levels. Furthermore, HTBPA and HTBPT significantly decreased the percentage of apoptotic cells in these H2O2-induced C2C12 cells. They also inhibited H2O2-induced apoptosis by modulating apoptosis-related proteins, including NF-κB, Bax, and caspase-3. Additionally, HTBPA and HTBPT suppressed the phosphorylation levels of key signaling proteins such as Akt, ERK1/2, and p38, thereby contributing to enhanced cell survival. These findings suggest that hydrolyzing tuna blood, waste of the canned tuna industry, can create a valuable ingredient for promoting muscle health.

Abstract

Glioblastoma (GBM) considered as aggressive brain cancer with high mortality rate in patients even after surgical resection. Resistant to chemotherapy is the major problem in GBM therapy. Discovery of novel bioactive compounds from algae is being investigated as alternative sources for potential treatment as well as prevention in glioblastoma. This study revealed the effects of marine red algae extract from hexane solvent fraction of Halymenia durvillei (HDHE) on proliferation and cell death in A172 human GBM cells. HDHE decreased proliferation and promoted cell cycle arrest at G2/M phase. HDHE induced apoptotic cell death in A172 cells through mitochondrial membrane dysfunction, the decrease of anti-apoptotic Bcl-2 protein expression, and activation of caspase 3/7. Moreover, HDHE increased intracellular reactive oxygen species (ROS) production and accumulation of LC3-II, an autophagic marker. The docked conformation of palmitic acid, a major component of HDHE, showed a high affinity binding to TP53 and Beclin-1 as cell death-related target molecules. This research conclusively demonstrated that HDHE might serve as a potent anticancer agent against glioblastoma by promoting apoptotic and autophagic cell death in A172 human GBM cells.

Abstract

Colorectal cancer is one of the most death-dealing cancers. However, conventional cancer treatments still have side effects. Therefore, novel chemotherapeutic agents with less side effects are still in search. A marine red seaweed, Halymenia durvillei, is recently interested in its anticancer effects. This study investigated the anticancer effect of ethyl acetate extract of H. durvillei (HDEA) on HT-29 colorectal cancer cells in association with the phosphoinositide 3-kinase (PI3K)/protein kinase B (AKT)/mammalian target of rapamycin (mTOR) signaling pathway. HDEA-treated HT-29 and OUMS-36 cells were used for cell viability tests by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyltetrazolium bromide assay. The effects of HDEA on apoptosis and cell cycle were evaluated. The nuclear morphology and mitochondrial membrane potential (ΔΨm) were observed by Hoechst 33342 and JC-1 staining, respectively. The gene expression of PI3K, AKT, and mTOR genes was evaluated using a real-time semiquantitative reverse transcription-polymerase chain reaction. The corresponding protein expressions were assessed by western blot analysis. The result revealed that the cell viability of treated HT-29 cells diminished while that of OUMS-36 cells was non-significant. By the down-regulation of cyclin-dependent ki-nase 4 and cyclin D1, HDEA-treated HT-29 cells were arrested in the G0/G1 phase. By the up-regulation of cleaved poly(adenosine diphosphate-ribose) polymerase, caspase-9, caspase-8, caspase-3, and Bax, HDEA-treated HT-29 cells underwent apoptosis, but suppressed Bcl-2, disrupted nuclear morphology and ΔΨm. Furthermore, treated HT-29 cells underwent autophagy by up-regulation of light chain 3-II and beclin-1. Lastly, HDEA suppressed the expression of PI3K, AKT, and mTOR. Therefore, HDEA exerts anticancer effects against HT-29 cells, confirmed by apoptosis, autophagy, and cell cycle arrest induction via regulation of the PI3K/AKT/mTOR signaling pathway.

สารสกัด Hexadecanoic acid จากสาหร่ายลิ้นมังกรสามารถกระตุ้นการตายของเซลล์มะเร็งเต้านมผ่านทาง ER stress

Abstract

Objective

To investigate the effect of the hexane solvent fraction of Halymenia durvillei (HDHE) on triple-negative breast cancer.

Methods

The phytochemical profile of HDHE was investigated by GC-MS. The cytotoxicity of HDHE against MDA-MB-231 cells was determined. The apoptotic and autophagic effects of HDHE were analyzed. The expression of molecular markers controlling apoptosis, autophagy, DNA damage, and endoplasmic reticulum (ER) stress was determined.

Results

HDHE contains a mixture of fatty acids, mainly hexadecanoic acid. HDHE at a cytotoxic concentration induced apoptotic death of MDA-MB-231 cells through mitochondrial membrane dysfunction, and induction of apoptosis markers, and increased the expression of proteins related to DNA damage response. HDHE also induced the expression of LC-3, a marker of autophagic cell death at a cytotoxic concentration. Moreover, HDHE modulated the expression of ER stress genes.

Conclusions

The hexadecanoic acid-enriched extract of Halymenia durvillei promotes apoptosis and autophagy of human triple-negative breast cancer cells. This extract may be further explored as an anticancer agent for triple-negative breast cancer.

Betulinic Acid Modulates the Expression of HSPA and Activates Apoptosis in Two Cell Lines of Human Colorectal Cancer https://anatomy.sc.mahidol.ac.th/betulinic-acid/ Thu, 09 Dec 2021 08:05:04 +0000 https://anatomy.sc.mahidol.ac.th/?p=12791 Highlight:

สาร Betulinic acid ชักนำให้เซลล์มะเร็งลำไส้ 2 ชนิด คือ HCT116 และ SW480 เกิดการตายแบบ apoptosis โดยลดการแสดงออกของ HSP70 เป็นต้น

Abstract

Betulinic acid (BA) is a pentacyclic triterpene usually isolated from botanical sources. Numerous studies have reported the inhibitory effect of BA against human colorectal cancer cells (CRC). However, its effect on the expression of the molecular chaperone HSPA is unclear. The aim of this research is to investigate the anti-cancer activities of BA purified from Piper retrofractum and study its effect on the expression of HSPA in colorectal cancer HCT116 and SW480 cells. The viability of both cancer cells was reduced after they were treated with an increasing dosage of BA. Flow cytometry assay revealed that levels of cell apoptosis significantly increased after incubation with BA in both cancer cells. Pro-apoptotic markers including Bax, cleaved-caspase-3 and cleaved-caspase-9 were increased while anti-apoptotic marker Bcl-2 was decreased after BA treatment. Western blot also showed that the expression of HSPA fluctuated upon BA treatment, whereby HSPA was increased at lower BA concentrations while at higher BA concentrations HSPA expression was decreased. Preliminary molecular docking assay showed that BA can bind to the nucleotide binding domain of the HSP70 at its ADP-bound state of the HSP70. Although further research is needed to comprehend the BA-HSPA interaction, our findings indicate that BA can be considered as potential candidate for the development of new treatment for colorectal cancer.

Moreover, previous studies demonstrated that the extract from H. scabra contains many bioactive compounds and has potent on inhibiting tumor cells survival and progression. However, the effect of the methanolic extract from the body wall of H. scabra (BWMT) on human prostate cancer cells has not yet been investigated. In this study, we aimed to investigate the effects and underlying mechanism of BWMT on prostate cancer cell viability and metastasis. BWMT was obtained by maceration with methanol. The effect of BWMT on cell viability was assessed by MTT and colony formation assays. The intracellular ROS accumulation was evaluated using a DCFH-DA fluorescence probe. Hoechst 33342 staining and Annexin V-FITC/PI staining were used to examine the apoptotic-inducing effect of the extract. A transwell migration assay was performed to determine the anti-metastasis effect. BWMT significantly reduced cell viability, triggered cellular apoptosis by accumulating intracellular ROS and resulting in the upregulation of JNK and p38 signaling pathways. In addition, BWMT also inhibited the invasion of PC3 cells by downregulating MMP-2/-9 expression via the ERK pathway. Consequently, our study provides BWMT from H. scabra as a putative therapeutic agent that could be applicable for prostate cancer progression.

Abstract

Introduction
Cancer cells utilize the modified glucose metabolism known as Warburg effect, with lactate production as the end product. In the search for alternative therapy, the body wall of sea cucumbers contains various substances with pharmacological activities. Herein, we investigate the effect of Holothuria scabra extract on the viability and Warburg effect of aggressive breast cancer cells.

Methods
Body wall of H. scabra was extracted using 95% ethanol. Triple-negative breast cancer cells, MDA-MB-231, were treated with the extract at various concentrations under normoglycemic and hyperglycemic conditions. Cytotoxicity test was performed using MTT assay. Apoptotic proteins were quantified using Western blot. Apoptotic cells were stained with Hoechst 33342. Lactate production was determined using L-lactate assay kit.

Results
By MTT assay, H. scabra extract suppressed the viability of breast cancer cells in a dose-dependent and time-dependent manner by enhancing apoptosis, indicated by a marked increase of proapoptotic Bax and pro-caspase three expressions, and decreased expression of anti-apoptotic Bcl-2. The extract could reduce hexokinase II expression, leading to reduced lactate production by blocking the Akt/mTOR/HIF-1 axis.

Discussion
Overall findings indicated that H. scabra extract could be a possible therapeutic against breast cancer progression in patients with hyperglycemia, for instance, diabetes mellitus.

Apoptotic effect of astaxanthin from white shrimp shells on lung cancer A549 cells https://anatomy.sc.mahidol.ac.th/msy-astaxanthin/ Wed, 18 Nov 2020 06:41:08 +0000 http://anatomy.sc.mahidol.ac.th/?p=10856 Fig 4 Caspase 3 - Morakot Sroyraya

ผลของ astaxanthin จากเปลือกกุ้งขาวต่อการตายของเซลล์มะเร็งปอด A549

ที่มาและความสำคัญ

Lung cancer is one of the major diseases worldwide, the third leading cause of cancer-related death after breast cancer and prostate cancer. Epidemiological studies have indicated non-small cell lung cancer (NSCLC) to be the predominant type of lung cancer. Current treatments for NSCLC patients are surgery and adjuvant therapy which may include radiation and chemotherapy. Major toxicities including neutropenia, peripheral neuropathy, nausea and vomiting, nephrotoxicity present as major side effects after chemotherapy. Thus, to develop a potent cancer therapeutic agent with low toxicities and fewer side effects would be desirable It has been reported that astaxanthin extracted from the shell of Arabian red shrimp Aristeus alcocki showed a more powerful antioxidant activity than the extract from the green microalgae Haematococcus pluvialis. In addition, free astaxanthin from Litopenaeus vannamei demonstrated a higher percentage of anti-oxidant activity than that from Haematococcus pluvialis. Nevertheless, the potential use of astaxanthin is often limited due to poor water solubility as well as low bioavailability after oral administration and susceptibility to thermal degradation. These obstacles continue to limit on its therapeutic application in aqueous-based system. Thus, the encapsulation of astaxanthin in nanoliposome, whose structure resembles the lipid membrane of living cells, as carrier system would preserve the native properties of astaxanthin as well as provide its stable aqueous dispersion thus improve its therapeutic potential. Therefore, the present study aimed to investigate the in vitro anti-cancer effects of astaxanthin extracted from white shrimp shells Litopenaeus vannamei and encapsulated in liposomes as well as its ability to induce apoptosis and cause cell cycle arrest in human NSCLC A549 cells.

Abstract
Purpose: To investigate the anti-cancer potential of astaxanthin from Litopenaeus vannamei encapsulated in liposomes (ASX) to treat lung cancer A549 cells.
Methods: Lung adenocarcinoma A549 cells were cultured and treated with ASX, following which cell viability and nuclear staining were performed. Generation of ROS was identified by the DCFH-DA assay while tetramethylrhodamine ethyl ester was used to determine the mitochondrial membrane potential. Flow cytometry was applied to investigate caspase-3/7 activity and cell cycle distribution.
Results: ASX inhibited growth of A549 in a concentration- and time- dependent manner. The IC50 values at 24, 48 and 72 h were 53.73, 22.85, 17.46 µg/mL, respectively (p < 0.05). After incubation with ASX, the morphological changes were observed in A549 cells following Hoechst 33342/PI fluorescent staining. ASX increased ROS generation and was associated with the collapse of mitochondrial membrane potential, which subsequently triggered the activation of caspase-3/7 activity leading to apoptosis (p < 0.05). In addition, A549 cells accumulated in the G0/G1 phase.
Conclusion: The results suggest that ASX is a valuable nutraceutical agent to target A549 lung cancer cells via ROS-dependent pathway as well as blockage of cell cycle progression.
