ที่มาและความสำคัญ

โรคพยาธิใบไม้ตับ (Fasciola gigantica) เป็นปัญหาสำคัญในสัตว์เคี้ยวเอื้อง โดยเฉพาะในแพะและโค ก่อให้เกิดการสูญเสียทางเศรษฐกิจจากการลดน้ำหนัก โตช้า และลดประสิทธิภาพการผลิต ปัจจุบันการควบคุมโรคยังพึ่งพายาถ่ายพยาธิเป็นหลัก ซึ่งมีความเสี่ยงต่อการดื้อยาเพิ่มขึ้นอย่างต่อเนื่อง จึงมีความจำเป็นเร่งด่วนในการพัฒนาวิธีป้องกันใหม่ เช่น วัคซีน การศึกษานี้จึงศึกษาศักยภาพของโปรตีน cathepsin L1H ในการลดการติดเชื้อพยาธิ และเป็นก้าวสำคัญสู่การพัฒนาวัคซีนที่มีประสิทธิภาพสำหรับควบคุมโรคพยาธิใบไม้ตับในแพะในอนาคต

Abstract

Cathepsin L1H has been identified as a potential vaccine candidate against fasciolosis. The present study was conducted to evaluate the immunoprophylactic efficacy of recombinant proFgCatL1H (rproFgCatL1H) antigen in goats subsequently challenged with the parasite. Immunization of goats with 200 µg of rproFgCatL1H formulated in Montanide™ ISA 61 VG conferred statistically significant reduction in worm recovery compared with both the infected control and adjuvant control groups, corresponding to reductions of 37.55 % and 34.07 %, respectively (p < 0.05). A negative correlation was observed between worm burden reduction and serum IgG levels (OD₄₅₀), measured both at the time of infection and at the study endpoint, although the association was not statistically significant. Scanning Electron Microscopy (SEM) analysis of adult F. gigantica recovered from vaccinated goats demonstrated pronounced tegumental damage, characterized by extensive swelling, surface erosion, and the presence of cell-like structures adhering to the tegumental surface. These findings suggest that the recombinant protein rproFgCatL1H holds promise as a potential vaccine candidate for the control of fasciolosis in goats.

การศึกษาวิจัยนี้มีวัตถุประสงค์เพื่อประเมินประสิทธิภาพของวัคซีนที่กระตุ้นด้วยโปรตีน cathepsin L1H และ B3 ในการป้องกันการติดเชื้อพยาธิใบไม้ตับ ทั้งในรูปแบบวัคซีนเดี่ยว (single vaccine) และวัคซีนกลุ่ม (combined vaccine) โดยการทดสอบในหนูทดลองร่วมกับสารเสริมฤทธิ์ (adjuvant) 2 ชนิดคือ Alum และ Freund ผลการทดลองพบว่าวัคซีนเดี่ยวและวัคซีนกลุ่มของ cathepsin L1H และ B3 มีผลป้องกันการติดเชื้อพยาธิได้ถึงร้อยละ 58.8 ถึง 75 โดยวัคซีนกลุ่มมีผลการป้องกันได้สูงกว่าวัคซีนเดี่ยว และวัคซีนทั้งหมดยังสามารถกระตุ้นภูมิคุ้มกัน โดยมีระดับแอนติบอดี IgG1 สูงกว่า IgG2a ซึ่งบ่งชี้ถึงกลไกการกระตุ้นผ่านทาง Th2 สูงกว่า Th1 ด้วย นอกจากนี้ผลการทดลองยังแสดงให้เห็นถึงประสิทธิภาพของการใช้สารเสริมฤทธิ์ Alum สูงเทียบเท่ากับ Freund ซึ่ง Alum จะมีความปลอดภัยเมื่อเทียบกับ Freund ที่มีความเป็นพิษและเกิดผลข้างเคียงในสิ่งมีชีวิตได้ ดังนั้นการศึกษานี้จึงสามารถใช้เป็นแนวทางการใช้วัคซีนเพื่อป้องกันการติดเชื้อพยาธิใบไม้ตับในสัตว์ใหญ่ต่อไป

Abstract
Protections against Fasciola gigantica infection in mice immunized with the individual and combined cathepsin L1H and cathepsin B3 vaccines were assessed. The vaccines comprised recombinant (r) pro-proteins of cathepsin L1H and B3 (rproFgCatL1H and rproFgCatB3) and combined proteins which were expressed in Pichia pastoris . The experimental trials were performed in ICR mice ( n = 10 per group) by subcutaneous injection with 50 μg of the recombinant proteins combined with Alum or Freund’s adjuvants. At two weeks after the third immunization, mice were infected with 15 F. gigantica metacercariae per mouse by oral route. The percents of protection of rproFgCatL1H, rproFgCatB3 and combined vaccines against F. gigantica were approximately 58.8 to 75.0% when compared with adjuvant-infected control. These protective effects were similar among groups receiving vaccines with Alum or Freund’s adjuvants. By determining the levels of IgG1 and IgG2a in the immune sera, which are indicative of Th1 and Th2 immune responses, it was found that both Th1 and Th2 humoral immune responses were significantly increased in vaccinated groups compared with the control groups, with higher levels of IgG1 (Th2) than IgG2a (Th1). Mice in vaccinated groups showed reduction in liver pathological lesions when compared with control groups. This study indicates that the combined rproFgCatB3 and rproFgCatL1H vaccine had a high protective potential than a single a vaccine, with Alum and Freund’s adjuvants showing similar level of protection. These results can serve as guidelines for the testing of this F. gigantica vaccine in larger economic animals.
