ที่มาและความสำคัญ

ในปัจจุบัน ความสนใจด้านการออกกำลังกายและสุขภาพร่างกายที่ดีเพิ่มสูงขึ้นอย่างต่อเนื่อง ส่งผลให้มีความต้องการผลิตภัณฑ์อาหารฟังก์ชันและอาหารเสริมที่ช่วยฟื้นฟูกล้ามเนื้อและเพิ่มสมรรถภาพทางกายมากขึ้น โดยภาวะเมื่อยล้าของกล้ามเนื้อและการสูญเสียมวลกล้ามเนื้อมักเป็นปัญหาที่เกิดขึ้นจากการออกกำลังกายอย่างหนัก ซึ่งมีสาเหตุหลักจากการสลายตัวของโปรตีนในกล้ามเนื้อร่วมกับการสะสมของอนุมูลอิสระ (ROS) ภายในเซลล์กล้ามเนื้อ การผลิต ROS ในปริมาณมากส่งผลให้เกิดความเครียดออกซิเดชัน ซึ่งทำให้การทำงานของกล้ามเนื้อแย่ลงและเพิ่มความเสี่ยงต่อการบาดเจ็บของเนื้อเยื่อกล้ามเนื้อ ดังนั้น การพัฒนากลยุทธ์เพื่อลดความเครียดออกซิเดชันและเร่งกระบวนการฟื้นฟูกล้ามเนื้อจึงเป็นสิ่งสำคัญ โดยเฉพาะสำหรับนักกีฬา ผู้ที่ออกกำลังกายเป็นประจำ และผู้ที่ต้องการรักษามวลกล้ามเนื้อ
ปัจจุบันเลือดปลาทูน่าเป็นของเสียจากอุตสาหกรรมที่ยังไม่ได้ใช้ประโยชน์ ทั้งที่อุดมด้วยกรดอะมิโนและแร่ธาตุสำคัญ อีกทั้งยังมีรายงานว่าเปปไทด์จากน้ำต้มปลาทูน่าสามารถยับยั้งเอนไซม์ ACE และกระตุ้นการเกิด apoptosis ในเซลล์มะเร็งเต้านม อย่างไรก็ตาม ยังไม่มีการศึกษาอย่างชัดเจนเกี่ยวกับศักยภาพของไฮโดรไลเซตจากเลือดปลาทูน่าในการเป็นวัตถุดิบเชิงหน้าที่เพื่อการฟื้นฟูกล้ามเนื้อ งานวิจัยนี้จึงมุ่งพัฒนาไฮโดรไลเซตจากเลือดปลาทูน่าเพื่อใช้เป็นส่วนผสมในผลิตภัณฑ์เพื่อสุขภาพกล้ามเนื้อ พร้อมส่งเสริมการใช้ทรัพยากรอย่างคุ้มค่าและยั่งยืน

ภาวะความล้าของกล้ามเนื้อจากการออกกำลังกายแบบทนทาน (exercise-induced muscle fatigue) เป็นภาวะที่ส่งผลให้สมรรถนะในการออกกำลังกายลดลง โดยมีปัจจัยสำคัญ ได้แก่ การลดลงของแหล่งพลังงาน การสะสมของสารเมแทบอไลต์ และภาวะ oxidative stress ทั้งนี้ AMPK/SIRT1/PGC-1α signaling pathway มีบทบาทสำคัญในการควบคุมสมดุลพลังงาน การสร้างไมโทคอนเดรีย และการผลิตพลังงาน ซึ่งมีส่วนช่วยในการคงสมรรถนะของกล้ามเนื้อและชะลอการเกิดความล้า

ปัจจุบันมีความสนใจในการนำสารออกฤทธิ์ทางชีวภาพจากแหล่งธรรมชาติมาพัฒนาเพื่อช่วยลดความล้าจากการออกกำลังกาย ขณะเดียวกัน ผลพลอยได้จากอุตสาหกรรมอาหารยังเป็นแหล่งของโปรตีนและสารออกฤทธิ์ทางชีวภาพที่มีศักยภาพแต่ยังไม่ได้รับการใช้ประโยชน์อย่างเต็มที่ โดยเฉพาะอุตสาหกรรมปลาทูน่าที่ก่อให้เกิดผลพลอยได้จำนวนมาก เช่น หัว กระดูก หนัง อวัยวะภายใน และเลือด การนำ เลือดปลาทูน่า ซึ่งเป็นผลพลอยได้ที่ยังมีการใช้ประโยชน์ค่อนข้างจำกัดมาเพิ่มมูลค่า จึงสามารถช่วยลดของเสียและผลกระทบต่อสิ่งแวดล้อมได้

การย่อยโปรตีนจากเลือดปลาทูน่าด้วยเอนไซม์ทำให้ได้ tuna blood hydrolysate (TBH) ซึ่งประกอบด้วยกรดอะมิโนและ bioactive peptides ที่มีศักยภาพทางชีวภาพ อย่างไรก็ตาม แม้จะมีรายงานว่า TBH มีฤทธิ์ต้านอนุมูลอิสระและต้านการอักเสบ รวมทั้งมีองค์ประกอบที่เกี่ยวข้องกับ energy metabolism แต่ข้อมูลเกี่ยวกับฤทธิ์ลดความล้าในสัตว์ทดลองและกลไกที่เกี่ยวข้องกับการควบคุมพลังงานและการทำงานของไมโทคอนเดรียยังมีจำกัด

ดังนั้น การศึกษานี้จึงมุ่งประเมินศักยภาพของ TBH ในการเพิ่มความทนทานและลดความล้าจากการออกกำลังกาย รวมถึงศึกษากลไกผ่าน AMPK/SIRT1/PGC-1α signaling pathway และวิเคราะห์ bioactive peptides ที่เกี่ยวข้อง เพื่อเป็นข้อมูลพื้นฐานสำหรับการพัฒนา TBH เป็น functional food หรือ nutritional supplement ตลอดจนส่งเสริมการเพิ่มมูลค่าและการใช้ประโยชน์จากผลพลอยได้ของอุตสาหกรรมปลาทูน่าอย่างยั่งยืน

Abstract

Exercise-induced muscle fatigue is associated with energy depletion, metabolite accumulation, and oxidative stress. These processes are closely linked to molecular mechanisms involving the AMPK/SIRT1/PGC-1α signaling pathway, which plays a crucial role in regulating energy homeostasis and mitochondrial biogenesis in response to exercise and nutritional interventions. Tuna blood hydrolysate (TBH) is a potential source of bioactive peptides that may regulate energy metabolism and alleviate exercise-induced muscle fatigue. Therefore, this study aimed to evaluate the effects of TBH as a nutritional supplement on muscle endurance and its anti-fatigue properties in vivo, as well as to identify its bioactive peptides using LC-MS/MS. The findings indicated that TBH significantly increased the time to exhaustion and also had no adverse effects. The liver and skeletal muscle glycogen were significantly increased after administration. TBH induced gene expression that associated with mitochondrial biogenesis. Additionally, the TBH significantly increased the expression of AMPK/SIRT1/PGC-1α signaling proteins. LC-MS/MS analysis identified over 14,000 peptides, with ATFSPYRDRIPLQIVR as the most abundant, and several peptides predicted to exhibit antioxidant and anti-inflammatory activities.

Abstract

Vibrio harveyi (V. harveyi) is a major bacterial pathogen that significantly impacts shrimp cultivation. This study aimed to evaluate the potential of bioflocs, harvested from shrimp pond systems, as a fishmeal substitute to combat V. harveyi infection in shrimp Penaeus vannamei (P. vannamei). Shrimp were fed with 6 diet formulations: Four experimental diets in which fishmeal was replaced with biofloc meal at 25, 50, 75 and 100 % (designated B25, B50, B75, and B100, respectively), a fishmeal-based control diet without biofloc (B0), and a positive control diet supplemented with β-glucan (BG). Feeding trials were conducted for 30 days, followed by a challenge with V. harveyi. Growth performance, survival rates, total hemocyte count (THC), phagocytic activity, and histopathological changes were evaluated. The results demonstrated that all biofloc-fed groups had growth performance comparable to the B0 and BG groups. Shrimp fed with the B25, B50, and B75 diets exhibited significantly enhanced immune responses, with elevated THC and phagocytic activity. In the V. harveyi challenge, the B25 group showed the highest survival rate among biofloc diets, comparable to that of the BG group. Histological analysis revealed that B25 and B50 diets reduced hepatopancreatic and muscle damage associated with infection. In conclusion, this study provides evidence that substituting fishmeal with biofloc (25 – 50 %) effectively enhances immune function and confers protection against V. harveyi infection in P. vannamei, supporting its potential as a sustainable functional feed ingredient in shrimp aquaculture.

Abstract

Red seaweeds (Rhodophyta) and their polysaccharides represent promising, sustainable solutions for boosting aquaculture productivity. Rich in bioactive compounds such as carrageenan, agar, and sulfated galactans, these seaweeds have demonstrated the ability to improve growth, health, and disease resistance in various aquaculture species. This review examines the applications, benefits, and limitations of red seaweeds in aquaculture, with particular attention to their roles as feed binders, partial fishmeal replacements, feed supplements, and immunostimulants. Species such as Gracilaria, Kappaphycus, and Porphyra have been shown to enhance immune responses, promote gut health, and mitigate nutrient pollution in aquaculture systems. Despite these advantages, challenges remain, including high cultivation costs, variability in bioactivity, and supply chain constraints. This review highlights the growing potential of red seaweeds for sustainable aquaculture while identifying research needs related to dosage optimization, delivery strategies, and elucidating the molecular mechanisms of their effects. Supplementation with red seaweeds aligns with the United Nations’ Sustainable Development Goals, offering an eco-friendly alternative to antibiotics with the potential to yield both environmental and economic benefits in aquaculture.

Abstract

Marine algal lectins are promising bioactive compounds with antimicrobial potential. This study produced and characterized a recombinant legume-type lectin from Gracilaria fisheri (rLGFL) and evaluated its antibacterial activity. The LGFL gene was cloned, sequence-verified, and expressed in Escherichia coli. Western blot analysis using a mouse polyclonal antiserum confirmed rLGFL expression. Functional assays demonstrated strong hemagglutination against rabbit erythrocytes, indicating carbohydrate recognition domain (CRD) functionality. rLGFL agglutinated Vibrio parahaemolyticus (3HP), Vibrio harveyi (1114), and Aeromonas veronii while exhibiting bactericidal activity against all tested pathogens, including non-agglutinated V. parahaemolyticus (A3212), Streptococcus agalactiaeAeromonas hydrophila, and Edwardsiella ictaluri. These findings suggest dual antibacterial mechanisms: agglutination-mediated pathogen immobilization and direct bactericidal effects. Seasonal analysis of G. fisheri from Songkhla, Thailand, revealed peak LGFL expression from January to March and July to September, identifying optimal harvesting periods. This study highlights rLGFL as a potential eco-friendly antibacterial agent with applications in aquaculture and disease management.

Highlight

This study developed hydrolyzed tuna blood powder (HTBP) from canned tuna industry by-products and demonstrated its antioxidant and anti-apoptotic effects on C2C12 muscle cells under oxidative stress. HTBPs, particularly those produced with alcalase and trypsin, reduced ROS and NO levels, suppressed apoptosis-related proteins, and improved cell survival. These findings suggest HTBP as a promising functional ingredient for muscle recovery and sustainable resource utilization.

ในปัจจุบัน ความสนใจด้านการออกกำลังกายและสุขภาพร่างกายที่ดีเพิ่มสูงขึ้นอย่างต่อเนื่อง ส่งผลให้มีความต้องการผลิตภัณฑ์อาหารฟังก์ชันและอาหารเสริมที่ช่วยฟื้นฟูกล้ามเนื้อและเพิ่มสมรรถภาพทางกายมากขึ้น โดยภาวะเมื่อยล้าของกล้ามเนื้อและการสูญเสียมวลกล้ามเนื้อมักเป็นปัญหาที่เกิดขึ้นจากการออกกำลังกายอย่างหนัก ซึ่งมีสาเหตุหลักจากการสลายตัวของโปรตีนในกล้ามเนื้อร่วมกับการสะสมของอนุมูลอิสระ (ROS) ภายในเซลล์กล้ามเนื้อ การผลิต ROS ในปริมาณมากส่งผลให้เกิดความเครียดออกซิเดชัน ซึ่งทำให้การทำงานของกล้ามเนื้อแย่ลงและเพิ่มความเสี่ยงต่อการบาดเจ็บของเนื้อเยื่อกล้ามเนื้อ ดังนั้น การพัฒนากลยุทธ์เพื่อลดความเครียดออกซิเดชันและเร่งกระบวนการฟื้นฟูกล้ามเนื้อจึงเป็นสิ่งสำคัญ โดยเฉพาะสำหรับนักกีฬา ผู้ที่ออกกำลังกายเป็นประจำ และผู้ที่ต้องการรักษามวลกล้ามเนื้อ

Abstract

This study aimed to develop hydrolyzed tuna blood powders (HTBP) with potential anti-apoptotic and muscle recovery properties. HTBP was produced alcalase, trypsin, and pepsin at concentrations of 0.5%, 1.0%, and 1.5%. The physical and chemical properties of HTBP were evaluated. The results showed no significant difference in yield (p > 0.05), with a range of 7.08% ± 0.78% to 9.26 % ± 1.74%. HTBP produced with alcalase at all concentrations yielded a higher amount of protein (76.86% ± 0.5%–79.1% ± 1.10%) compared to trypsin and pepsin. Glutamic acid, cysteine, and aspartic acid were identified as the main amino acids in HTBP. The water solubility index of all HTBP was high (>90%). Antioxidant values increased with higher protein content. Hydrolyzed tuna blood with 0.5% alcalase (HTBPA) and 0.5% trypsin were selected for further investigation of their biological activities, assessed using C2C12 muscle cells under oxidative stress conditions induced by hydrogen peroxide (H2O2). In H2O2-induced C2C12 cells, HTBPA and HTBPT reduced nitric oxide (NO) secretion and intracellular reactive oxygen species (ROS) levels. Furthermore, HTBPA and HTBPT significantly decreased the percentage of apoptotic cells in these H2O2-induced C2C12 cells. They also inhibited H2O2-induced apoptosis by modulating apoptosis-related proteins, including NF-κB, Bax, and caspase-3. Additionally, HTBPA and HTBPT suppressed the phosphorylation levels of key signaling proteins such as Akt, ERK1/2, and p38, thereby contributing to enhanced cell survival. These findings suggest that hydrolyzing tuna blood, waste of the canned tuna industry, can create a valuable ingredient for promoting muscle health.

แอมฟิพอด (amphipod) ในตะกอนชีวภาพ (bioflocs) เป็นอาหารทางธรรมชาติที่มีสารอาหารสูงเหมาะสำหรับการเพาะเลี้ยงสัตว์น้ำ งานวิจัยนี้จึงมีวัตถุประสงค์เพื่อศึกษาสปีชีส์ของแอมฟิพอดในบ่อเลี้ยงกุ้งที่ใช้ระบบตะกอนชีวภาพ (biofloc technology) ที่ฟาร์มกุ้ง อำเภอไชยา จังหวัดสุราษฎร์ธานี และศึกษาคุณค่าทางโภชนาการและการเพิ่มจำนวนของแอมฟิพอดในบ่อระบบตะกอนชีวภาพ โดยนำพ่อแม่พันธุ์ของแอมฟิพอดมาทำการทดลองเลี้ยงด้วยแหล่งคาร์บอน (carbon sources) ที่ต่างชนิดกัน คือกากน้ำตาล (molasses) รำข้าว(rice bran) และแป้งข้าวโพด (corn flour) ที่ระดับอัตราของคาร์บอนต่อไนโตรเจน (C:N) ratio เท่ากับ 16:1 เป็นเวลา 4 สัปดาห์ แล้วทำการตรวจนับจำนวน ตรวจสอบการวางไข่ และ คุณค่าทางโภชนาการของแอมฟิพอดที่เลี้ยงในตะกอนชีวภาพ ผลการทดลองพบ Bemlos quadrimanus sp เป็นสปีชีส์ของแอมฟิพอดในบ่อเลี้ยงกุ้งที่พบในระบบตะกอนชีวภาพ และระยะ juvenile เป็นระยะที่มีโปรตีนและลิปิดสูงกว่าระยะอื่น นอกจากนี้พบว่าปริมาณกรดอะมิโนและกรดไขมันที่จำเป็นในตะกอนชีวภาพมีค่าใกล้เคียงกับปลาป่น ตะกอนชีวภาพจากแป้งข้าวโพดส่งเสริมการเจริญเติบโตและเพิ่มจำนวนของแอมฟิพอดในระยะ juvenile ทำให้ได้แอมฟิพอดที่มีโปรตีและลิปิดสูงกว่าแหล่งคาร์บอนอื่น งานวิจัยนี้แสดงให้เห็นว่าแอมฟิพอด B. quadrimanus เป็นแหล่งโปรตีนที่สำคัญที่สามารถทดแทนปลาป่นในอาหารของกุ้งได้ และการเพาะเลี้ยงแอมฟิพอดในบ่อระบบตะกอนชีวภาพเป็นการเพิ่มมูลค่าตะกอนชีวภาพในอุตสาหกรรมการเพาะเลี้ยงสัตว์น้ำ

Abstract

Natural food items like amphipods in bioflocs have been shown to provide a nutrient-rich live feed for aquaculture species. In the present study, we aimed to identify the major species of amphipods in the biofloc technology shrimp ponds at Shrimp Village, Surat-Thani, Thailand, determine its nutritional values, and enrich the amphipods in the biofloc technology system (BFS). Breeder stage amphipods were cultured in BFS with different carbon sources; molasses, rice bran, and corn flour at a carbon: nitrogen (C:N) ratio of 16:1 for 4 weeks. The number and the growth of spawned juveniles, and the nutritional compositions of the amphipods enriched bioflocs were examined. We found that the major species of amphipods in bioflocs produced in the cultured shrimp ponds was Bemlos quadrimanus sp. Proximate analysis revealed that the juvenile stage B. quadrimanus had higher levels of protein and lipid contents than those in other stages. Moreover, their essential amino acid index (EAAI) and ratios of polyunsaturated fatty acid/saturated fatty acid (PUFA/SFA) and docosahexaenoic acid/eicosapentaenoic acid (DHA/EPA) were similar to those values in fishmeal. The bioflocs produced with corn flour enhanced the growth and increased the number of juvenile amphipods greater than in bioflocs produced with the other two carbon sources, and had the highest levels of crude protein and lipid. In summary, this study demonstrates that amphipod B. quadrimanus provide an important source of protein that shows potential to replace fishmeal in shrimp feed, and reveals the benefit of a culture of amphipods with BFS in the aquaculture industry.

ปลาป่น (fish meal) เป็นแหล่งโปรตีนที่สำคัญในอุตสาหกรรมผลิตอาหารกุ้ง ปลาป่นได้มาการลากอวนปลาเล็กปลาน้อยจากทะเลซึ่งเป็นการทำลายระบบนิเวศอย่างมาก ดังนั้นการหาแหล่งโปรตีนคุณภาพดีเพื่อทดแทนปลาป่นจึงมีความจำเป็นอย่างเร่งด่วน งานวิจัยนี้จึงมีวัตถุประสงค์เพื่อศึกษาคุณค่าทางโภชนาการในตะกอนชีวภาพ (bioflocs) การใช้ตะกอนชีวภาพทดแทนปลาป่นในอาหารกุ้ง และผลของตะกอนชีวภาพต่อการเจริญเติบโตและภูมิคุ้มกันของกุ้ง ตะกอนชีวภาพได้จากการเก็บเกี่ยวจากบ่อเลี้ยงกุ้ง (C:N ratio > 12:1) ที่ Shrimp Village อำเภอไชยา จังหวัดสุราษฎร์ธานี ผลการทดลองพบว่าโปรตีนและลิปิดในตะกอนชีวภาพมีค่าเท่ากับ 48% และ 5% ตามลำดับ ปริมาณกรดอะมิโนและกรดไขมันที่จำเป็นในตะกอนชีวภาพมีค่าใกล้เคียงกับปลาป่น เมื่อให้กุ้งกินอาหารที่ใช้ตะกอนชีวภาพแทนปลาป่นในปริมาณแตกต่างกัน (25, 50, 75 และ 100%) เป็นเวลา 42 วัน พบว่าผลของการเจริญเติบโตของกุ้ง (น้ำหนักที่เพิ่มขึ้น ความเฉพาะเจาะจงของการเจริญเติบโตและอัตราแลกเนื้อ) ในกลุ่มกินอาหารที่ใช้ตะกอนชีวภาพแทนปลาป่นไม่มีความแตกต่างจากอาหารปกติ นอกจากนี้ยังพบว่าอัตราการรอด ระดับของภูมิคุ้มกัน และการแสดงออกของยีน (proPO-I, PEN-4 and dicer) สูงขึ้นในกุ้งทุกกลุ่มที่กินอาหารตะกอนชีวภาพแทนปลาป่น และเมื่อให้กุ้งติดเชื้อVibrio parahaemolyticus กุ้งกลุ่มที่กินอาหารตะกอนชีวภาพแทนปลาป่น 25 และ 50% มีระดับของภูมิคุ้มกันและอัตราการรอดหลังจากการติดเชื้อV. parahaemolyticus) สูงกว่ากุ้งที่กินอาหารปกติอย่างมีนัยสำคัญทางสถิติ งานวิจัยนี้แสดงให้เห็นว่าตะกอนชีวภาพที่คุณค่าโปรตีนเทียบเท่าปลาป่น นำไปใช้ทดแทนปลาป่นในอาหารกุ้งได้ และยังเพิ่มภูมิคุ้มกันในกุ้ง

Abstract

Fishmeal is the main source of protein in the shrimp feed industry and is normally derived from trash fish. As such, the production of fishmeal has an adverse effect on the marine environment by taking away small and juvenile fish, leading to depletion of marine species. There is a need for alternative sources of protein which will substitute fishmeal in the aquaculture industry. This study evaluated the components and nutritional efficacy of bioflocs, which were used to substitute fishmeal protein. The effect of bioflocs diets on growth performance, survival rate, and immune response in shrimp compared to normal fishmeal feed were determined. Bioflocs were harvested from the shrimp ponds (C:N ratio >12:1) at Shrimp Village, Chaiya district, Surat Thani, Thailand. The total protein in bioflocs was about 48% and the total lipid was about 5% (dried weight) and the percentages of essential amino acids (EAA) and fatty acids (EFA) in bioflocs were similar to those of fishmeal feed. Shrimp fed with the different dietary bioflocs feed regimens [% to replace fishmeal; 0% (B0), 25% (B25), 50% (B50), 75% (B75), and 100% (B100)] for 42 days revealed that all growth parameters were almost similar to those of the control shrimp (shrimp fed with normal fishmeal, B0) including final body weight, weight gain, specific growth rate, and feed conversion ratio. Remarkably, the survival rates, the levels of immune parameters, and expression of immune genes (proPO-I, PEN-4 and dicer) were significantly higher in bioflocs fed shrimp, especially in B25 and B50 shrimp. Moreover, B25 and B50 bioflocs fed shrimp showed notably increased survival rates following Vibrio parahaemolyticus (V. parahaemolyticus) infection. In conclusion, the present study demonstrates that shrimp survival and immunity are enhanced by biofiocs substituted fishmeal. Significantly, the bioflocs diets activated the immune response to prevent V. parahaemolyticus infection.
