คณะผู้วิจัยได้ทำการศึกษาผลของกลูตาเมท (Glutamate, Glu) ต่อการกระตุ้นการกินอาหาร การเปลี่ยนแปลงของเนื้อเยื่อวิทยาของลำไส้ การเจริญเติบโต และระบบแอนตี้ออกซิแดนซ์ภูมิคุ้มกัน ในลูกกุ้งก้ามกรามระยะจูวิไนล์ โดยพบว่า การให้อาหารที่ผสมด้วย Glu ที่ระดับความเข้มข้น 55 และ 550 mg/kg feed สามารถกระตุ้นการกินอาหาร โดยการเพิ่ม food consumption, digestive enzyme activities, และกระตุ้น feeding behaviors นอกจากนี้ยังพบการเปลี่ยนแปลงของเนื้อเยื่อวิทยาของลำไส้ เมื่อเปรียบเทียบกับกลุ่มควบคุม (P < 0.05) สารสื่อประสาท Glu สามารถกระตุ้นการเจริญเติบโต โดยการเพิ่มขนาด, weight gain (WG), growth rate (GR), specific growth rate (SGR), และaverage daily gain (ADG) ในช่วงสัปดาห์ที่ 8 ถึง 12 เมื่อเปรียบเทียบกับกลุ่มควบคุม อย่างมีนัยสำคัญทางสถิติ (P < 0.05). นอกจากนี้ อาหารที่ผสม Glu สามารถเพิ่มการแสดงออกของ appetite/feeding-related gene อาทิ neuropeptide F (NPF) และ muscle growth-, immune-related genes ตลอดจน immune ต่างๆ รวมทั้ง พบอัตราการอยู่รอด (Survival rate) ที่สูงกว่ากลุ่มควบคุม (P < 0.05) จากผลการวิจัยนี้ คณะผู้วิจัยได้ค้นพบองค์ความรู้พื้นฐานใหม่ที่สำคัญมาก ที่แสดงให้เห็นว่า Glu สามารถกระตุ้นการกินอาหาร การเจริญเติบโต และภูมิคุ้มกัน ในลูกกุ้งก้ามกรามระยะจูวิไนล์ นอกจากนี้ ยังค้นพบกลไกการควบคุม Glu ที่สำคัญต่อการกินอาหาร และ/หรือ การเจริญเติบโต ที่อาจจะทำงานร่วมกัน (interplay) กับ NPF ในการควบคุมสรีระวิทยาเหล่านี้ โดยองค์ความรู้ใหม่ที่ได้รับนี้ อาจจะสามารถนำ Glu ซึ่งมีราคาไม่แพง นำมาประยุกต์ใช้โดยการผสมอาหาร (functional feed) เพื่อเพิ่มอัตราการกินอาหาร การเจริญเติบโต ตลอดจนเพิ่มภูมิคุ้มกัน ในการเพาะเลี้ยงลูกกุ้งก้ามกรามได้ในอนาคต

ที่มาและความสำคัญ

The giant freshwater prawn (Macrobrachium rosenbergii) culturing in Thailand currently faces significant challenges, notably the high-density captive breeding system, which increases stress and retards growth rate of juvenile prawns. Stress leads to diminished appetite, decreased food intake, stunted growth, heightened vulnerability to diseases, and elevated mortality rates. Feeding is an important process for acquiring nutrients essential to energy production and growth in juvenile prawns. Numerous factors influence feeding, including food type, temperature, light, hormones, and neurotransmitters (Volkoff, 2016; Volkoff & Rønnestad, 2020; Tierney, 2020). These factors interact through complex regulatory mechanisms that control digestive function for nutrient absorption, energy balance, and growth. Increasing feeding rates can also lead to higher growth, improved health, as well as enhanced production and economic value of cultured prawns. Glutamate (Glu) is a neurotransmitter and a versatile non-essential amino acid. Glu in intestinal cells stimulates neural networks, consequently influencing brain regions through nutrient sensing. This indicates that Glu is a significant regulator of food consumption, digestion, and nutrient absorption. However, the effects and underlying regulatory mechanisms of Glu on feed consumption, digestibility, growth, appetite-related peptides, and antioxidant–immune gene expression in juvenile freshwater prawn, M. rosenbergii, remain unclear. Given its physiological importance, Glu may serve as a key regulator of feeding, growth, and immune responses in juvenile prawns.

Abstract

The aim of the study is to explore the effects of glutamate (Glu) supplementation on feed consumption, digestibility, intestinal histology, growth, gut barrier, and the antioxidant–immune-related genes during the juvenile stage of freshwater prawn (Macrobrachium rosenbergii). The supplementation of Glu at doses of 55 and 550 mg/kg feed significantly enhanced feed consumption, digestion, digestive enzyme activities, and gut histology (P < 0.05). Feeds supplemented with these two doses of Glu improved growth, demonstrated by increased body size, weight gain (WG), growth rate (GR), specific growth rate (SGR), and average daily gain (ADG) during weeks 8–12 (P < 0.05). Additionally, juvenile prawns fed diets supplemented with both doses of Glu exhibited significant upregulation of appetite-related gene, as well as muscle growth-related and immune-related genes (P < 0.05). Survival rate (SR) was significantly higher for juvenile prawns receiving diets with Glu supplementation at weeks 8 and 12. Collectively, these results indicate that Glu function as an appetite stimulant, growth enhancer, and immune modulator; therefore, this neurotransmitter may be a potential food additive for improving the production of juvenile prawns raised in an aquaculture system.

ที่มาและความสำคัญ

Cultivation of freshwater prawn (Macrobrachium rosenbergii) has expanded considerably, thus emerging as a substantial contributor to Thailand’s aquaculture sector (New and Nair 2012; Tinikul et al., 2023; FAO, 2024). The global production of M. rosenbergii has increased significantly, reaching more than 300,000 tons. Meanwhile, demand has also grown significantly in both local and international markets. However, freshwater prawn farming also faces considerable obstacles, particularly disease outbreaks and hormonal imbalances in prawns caused by stressful situations, such as high-density culture, global warming, and overcrowded agricultural habitats (Hooper et al., 2023; Bhassu et al., 2024). These environmental stressors can lead to decreased feeding, nutritional deficiency, high mortality rate, and retarded growth, ultimately resulting in reduced productivity and significant economic losses (Pillai and Bonami, 2012; Kamaruding and Abdullah, 2021; Calado et al., 2022; Paul et al., 2025). Hence, feeding is a crucial behavior that ensures the intake of sufficient nutrients to maintain energy balance for these physiological processes (Celi et al., 2017). Feed constitutes the main expense in the aquaculture and ensures the quality, quantity, and good health of the prawns being raised (Yu et al., 2021). Thus, it is important to develop a suitable feed that optimizes feeding rates, promotes growth, and enhances immunity during the key developmental stages of this prawn. Neuropeptide F (NPF) is an important neuropeptide that potentially regulates feeding, foraging, metabolism, development, and reproduction, in insects and crustaceans (Fadda et al., 2019; Zhao et al., 2023). Considering the critical roles of NPF in regulating feeding, digestive enzyme activity, development and immunity in insects and crustaceans, we question whether M. rosenbergii NPF (MrNPF) could promote feeding, digestive functions, growth and immunity in juvenile prawns. This important knowledge of MrNPF supplementation may be developed as a viable feed additive in prawn aquaculture to improve feeding, growth performance, muscle mass, and health status of juvenile prawns, M. rosenbergii.

Abstract

Neuropeptide F (NPF) regulates feeding, growth, development, and reproduction in insects and crustaceans. In this study, we examined the effects of Macrobrachium rosenbergii NPF (MrNPF) on food intake, digestive functions, growth performance, and immunity in juvenile freshwater prawns (M. rosenbergii). Supplementing MrNPF at 5 and 0.5 μg/g of feed significantly stimulated food intake and digestive functions by enhancing food consumption, hepatosomatic index, and feeding behaviors; increasing digestive amylase, protease, and lipase activities; and improving gut histomorphology (P < 0.05). In addition, both doses of MrNPF-supplemented feed promoted growth performance by increasing mean weight gain (WG), specific growth rate, and daily WG from weeks 6 to 12 compared with the control groups (P < 0.05). Survival rate was significantly higher in the high-dose MrNPF group than in controls only at week 9 (P < 0.05). Furthermore, MrNPF supplementation significantly upregulated the expression of muscle growth-related genes, including MrTORelF4B, and LIM mRNA. Finally, MrNPF significantly enhanced the expression of immune-related genes. Collectively, our findings suggest that MrNPF is an essential neuropeptide that may serve as a potential regulator of feeding, growth, and immunity in juvenile M. rosenbergii. This knowledge could be used to enhance the aquaculture productivity of this crustacean species.

Abstract

Staphylococcus epidermidis is a leading opportunistic pathogen in medical device-associated infections due to its ability to adhere to abiotic materials and develop biofilms that are difficult to eradicate. This study investigated the antibiofilm potential of an ethanolic extract of the red seaweed Gracilaria fisheri and its purified constituent, N-benzylcinnamamide, against S. epidermidis. Antibacterial activity was determined, and antibiofilm effects were assessed using the crystal violet assay and confocal laser scanning microscopy (CLSM). Early bacterial adhesion on glass and polyurethane (PU) surfaces was measured. The effect on catheter-associated biofilms was evaluated by scanning electron microscopy (SEM). Transcripts of biofilm- and quorum-sensing-associated genes (icaA and luxS) were assessed by semi-quantitative RT-PCR. Cytotoxicity was evaluated by MTT assay. At 200 µg/mL, biofilm biomass decreased to 48.21 ± 5.52% with the extract and to 36.65 ± 6.82% with N-benzylcinnamamide. CLSM time-course imaging showed delayed biofilm maturation and less consolidated, discontinuous structures. Surface exposure to the extract markedly reduced early attachment on both materials. On PU catheter segments, SEM demonstrated that N-benzylcinnamamide markedly reduced surface coverage and disrupted three-dimensional biofilm architecture. At the molecular level, transcription of icaA and luxS was reduced. Both the extract and N-benzylcinnamamide showed minimal cytotoxicity in HeLa cells. These findings support further evaluation of these marine-derived agents as candidates for antibiofilm surface treatments to reduce early medical device colonization.

การเพาะเลี้ยงกุ้งขาวได้รับผลกระทบมากจากการติดเชื้อแบคทีเรีย Vibrio harveyi ทำให้เกิดการสูญเสียและลดผลผลิต ขณะเดียวกันอาหารกุ้งที่พึ่งพา fishmeal มีต้นทุนสูงและมีข้อจำกัดด้านความยั่งยืน จึงมีความจำเป็นต้องหาแหล่งโปรตีนทดแทนที่หากช่วยเสริมภูมิคุ้มกันด้วย จะมีประโยชน์ลดต้นทุน และเพิ่มปริมาณผลผลิตในอุตสาหกรรมการเพาะเลี้ยงกุ้ง งานวิจัยนี้จึงศึกษาการใช้ biofloc meal จากระบบบ่อกุ้งเป็นวัตถุดิบทดแทน fishmeal และประเมินศักยภาพในการเพิ่มภูมิคุ้มกันและเพิ่มการรอดของกุ้งเมื่อติดเชื้อ V. harveyi

Abstract

Vibrio harveyi (V. harveyi) is a major bacterial pathogen that significantly impacts shrimp cultivation. This study aimed to evaluate the potential of bioflocs, harvested from shrimp pond systems, as a fishmeal substitute to combat V. harveyi infection in shrimp Penaeus vannamei (P. vannamei). Shrimp were fed with 6 diet formulations: Four experimental diets in which fishmeal was replaced with biofloc meal at 25, 50, 75 and 100 % (designated B25, B50, B75, and B100, respectively), a fishmeal-based control diet without biofloc (B0), and a positive control diet supplemented with β-glucan (BG). Feeding trials were conducted for 30 days, followed by a challenge with V. harveyi. Growth performance, survival rates, total hemocyte count (THC), phagocytic activity, and histopathological changes were evaluated. The results demonstrated that all biofloc-fed groups had growth performance comparable to the B0 and BG groups. Shrimp fed with the B25, B50, and B75 diets exhibited significantly enhanced immune responses, with elevated THC and phagocytic activity. In the V. harveyi challenge, the B25 group showed the highest survival rate among biofloc diets, comparable to that of the BG group. Histological analysis revealed that B25 and B50 diets reduced hepatopancreatic and muscle damage associated with infection. In conclusion, this study provides evidence that substituting fishmeal with biofloc (25 – 50 %) effectively enhances immune function and confers protection against V. harveyi infection in P. vannamei, supporting its potential as a sustainable functional feed ingredient in shrimp aquaculture.

Abstract

Red seaweeds (Rhodophyta) and their polysaccharides represent promising, sustainable solutions for boosting aquaculture productivity. Rich in bioactive compounds such as carrageenan, agar, and sulfated galactans, these seaweeds have demonstrated the ability to improve growth, health, and disease resistance in various aquaculture species. This review examines the applications, benefits, and limitations of red seaweeds in aquaculture, with particular attention to their roles as feed binders, partial fishmeal replacements, feed supplements, and immunostimulants. Species such as Gracilaria, Kappaphycus, and Porphyra have been shown to enhance immune responses, promote gut health, and mitigate nutrient pollution in aquaculture systems. Despite these advantages, challenges remain, including high cultivation costs, variability in bioactivity, and supply chain constraints. This review highlights the growing potential of red seaweeds for sustainable aquaculture while identifying research needs related to dosage optimization, delivery strategies, and elucidating the molecular mechanisms of their effects. Supplementation with red seaweeds aligns with the United Nations’ Sustainable Development Goals, offering an eco-friendly alternative to antibiotics with the potential to yield both environmental and economic benefits in aquaculture.

ในการศึกษาครั้งนี้ คณะผู้วิจัยได้ทำการศึกษาการปรากฎของกลูตาเมท (glutamate, Glu) และซีโรโทนิน (serotonin, 5-HT) ในอวัยวะย่อยอาหาร และทำการศึกษาผลของการออกฤทธิ์ของกลูตาเมท และซีโรโทนิน ต่อการกินอาหาร แอคติวิตี้ของเอนไซม์การย่อยอาหาร และระบบภูมิคุ้มกัน ในกุ้งก้ามกรามเพศเมีย (Macrobrachium rosenbergii) คณะผู้วิจัยพบว่า Glu และ 5-HT มีการปรากฎใน foregut, midgut, hindgut, และ hepatopancreas และมีระดับความเข้มข้นสูงในระยะเต็มวัยของรังไข่ ในขณะที่สารสื่อประสาททั้งสองตัวนี้ พบระดับความเข้มข้นลดลง ในช่วงของรังไข่ระยะอ่อน โดยใช้วิธี high-performance liquid chromatography (HPLC) นอกจากนี้ ยังพบการปรากฏของ Glu-immunoreactivity (-ir) และ 5-HT-ir ในอวัยวะทางเดินอาหาร โดยใช้วิธี immunohistochemistry นอกจากนี้ กุ้งก้ามกรามที่ถูกฉีดด้วย Glu และ 5-HT ที่ระดับความเข้มข้น 2.5 × 10−6 และ 2.5 × 10−7 mol/prawn ส่งผลต่อ food consumption, gastric emptying midgut index, และเพิ่ม digestive enzymes อาทิ amylase, protease, และ lipase activities เมื่อเปรียบเทียบกับกลุ่มควบคุม อย่างมีนัยสำคัญทางสถิติ (P < 0.05). Glu และ 5-HT ยังสามารถกระตุ้น epithelial cell height, villi height, crypt depth และสามารถเพิ่มปฏิกิริยาของ PAS/AB ในอวัยวะทางเดินอาหาร นอกจากนี้ Glu และ 5-HT สามารถเพิ่มระดับของ M. rosenbergii NPF (MrNPF) และ immune-related transcripts อย่างมีนัยสำคัญทางสถิติ (P < 0.05) จากผลการวิจัยนี้ คณะผู้วิจัยได้ค้นพบองค์ความรู้พื้นฐานใหม่ที่สำคัญอย่างมาก ที่อาจจะสามารถนำสารสื่อประสาท Glu และ 5-HT มาประยุกต์ใช้เพื่อที่จะเพิ่มอัตราการกินอาหาร และการเพิ่มภูมิคุ้มกัน ในส่วนของการเพาะเลี้ยงกุ้งก้ามกรามเพศเมียในอนาคตต่อไปได้

ที่มาและความสำคัญ

Currently, prawn aquaculture with high-density populations faces several challenges, including diseases and infections, which cause stress and lead to diminished appetite and reduced feeding (Hooper et al., 2023). These factors directly impact gonadal development, particularly in the ovaries, leading to a decrease in the quantity and quality of eggs and, consequently, reduced offspring production. Feeding is affected by external factors such as water temperature, light, and food composition and taste properties (Kestemont and Baras, 2001; Volkoff and Rønnestad, 2020). In addition, feeding is regulated by internal factors, including various neurotransmitters, e.g., glutamate (Glu), serotonin (5-HT), and catecholamines (Scannell et al., 2008; Tierney, 2020). These factors are crucial in controlling feeding by activating the digestive and nervous systems, which is essential for assuring enough nutrient ingestion to maintain energy balance in the body (Celi et al., 2017). In female crustacean broodstocks, feeding is a crucial physiological process to acquire sufficient nutrients that are utilized to produce energy for ovarian development, oocyte production, and vitellogenesis. The quantity and quality of oocytes influence the successful development of fertilized eggs, which subsequently impacts crustacean production (Fernández-Palacios et al., 2011; Feng et al., 2023). Considering the importance of Glu and 5-HT in physiological functions, such as feeding, digestive enzyme activity, and immunity in aquatic animals like fish and decapod crustaceans, we question whether these neurotransmitters display alterations and regulatory functions in the digestive organs as essential modulators of feeding and immunity in female M. rosenbergii during the ovarian cycle.

Abstract

This study investigated the presence of glutamate (Glu) and serotonin (5-HT) in the digestive organs and their effects on feeding performance, digestive enzyme activity, and immunity throughout the ovarian cycle in female giant freshwater prawns (Macrobrachium rosenbergii). By high-performance liquid chromatography (HPLC), Glu and 5-HT were detected in the foregut, midgut, hindgut, and hepatopancreas, at the highest concentrations in the late ovarian stage and the lowest concentrations in the early ovarian stage. Through immunohistochemistry, the presence of Glu-immunoreactivity (-ir) and 5-HT-ir were observed in all digestive organs, and their intensities were shown to correlate with the levels detected by those observed via HPLC. Prawns injected with Glu and 5-HT at the doses of 2.5 × 10−6 and 2.5 × 10−7 mol/prawn, markedly increased food consumption, gastric emptying, and midgut index as well as enhanced amylase, protease, and lipase activities, compared with the control group (P < 0.05). Glu and 5-HT also enhanced epithelial cell height, villi height, and crypt depth and promoted the reactivity of PAS/AB in the digestive organs. In addition, treatment with Glu or 5-HT significantly upregulated M. rosenbergii NPF (MrNPF) and immune-related transcripts (P < 0.05). Collectively, these findings demonstrated the promising effects of Glu and 5-HT at a dose of 2.5 × 10−6 mol/prawn in stimulating feeding, digestive activities, and immunity, which could potentially be used to enhance feeding, growth, and maintain the health status of female prawn in aquaculture.

Abstract

Sperm activation occurring in both male and female reproductive tract involves a highly complex series of biomolecular dynamics, particularly on membrane lipids and proteins. In mammals, the universal anticipation in cholesterol (CHO) sequestration plays a role in mammalian sperm maturation/capacitation, subsequently enhancing sperm fertilizing ability. In shrimp, we have previously shown that the level of cholesterol (CHO) is significantly reduced in vas deferens sperm when compared with sperm in the testes, presumably due to the sequestering action of the lipid-binding protein, one of which is Niemann-Pick Type C-2 (NPC2). Here, we used recombinant MrNPC2 protein to treat testicular sperm (Tsp) and quantitatively compared the amount of CHO with non-treated or 2-OH-β-CD treated sperm. HPTLC of the extracted lipids from Macrobrachium rosenbergii sperm revealed the presence of major phospholipids and CHO. Tsp treated with 2-OH-β-CD or MrNPC2 showed lower CHO levels, which was comparable to that of the vas deferens sperm as verified by the Amplex Red assay. Finally, the enhanced levels of protein tyrosine phosphorylation and ionophore-induced AR levels in the MrNPC2 treated Tsp significantly increased in a concentration-dependent manner, similar to that of the Vsp sperm. Altogether, our results indicated the importance of MrNPC2 as CHO moderator in sperm membrane, leading to an enhancement in sperm fertilizing ability.

Abstract

One of the most successful applications to protect aquatic animals from viral infection is the use of RNA interference (RNAi) in the form of double-stranded RNA (dsRNA). However, its administration into animals is usually via injection route (laboratory scale) or mixing feed pellets with bacterial extracts (containing therapeutic compounds); the outcome of which remains to be improved. In this study, we encapsulated purified dsRNA into a virus-like particle (VLP) and delivered it into the prawn tissues by a submersion route using interiorly modified Macrobrachium rosenbergii nodavirus derived VLP with deca-arginine peptide (10R-MrN-VLP) to encapsulate a large scale (gram-level) of dsRNA against RNA2 gene of MrNV capsid. For tracing purposes of submersion administration, GFP plasmid-loaded VLP successfully delivered and expressed green fluorescent protein in prawn-targeted tissues. Delivery efficiency at 12-h submersion was about 50% of the prawn population and became 100% within 72 h, and they remained GFP-positive for up to 7 days during submersion. The protective effect of dsRNA-VLP submersion was tested with a large group (10,000) of post-larvae, which were naturally challenged with MrNV-positive dead prawn. Transcriptome analysis revealed up- and down-regulations of gene clusters involved in metabolism and signal transduction related to an antiviral immune system. The survival rate of prawn treated with dsRNA-VLP reached 80% at Day 7 post-MrNV challenge, significantly different from that of the MrNV-challenged group (control). PCR screening revealed an apparent decrease of MrNV genes and the copy number of MrNV in prawn sampling tissues. Together, we believe that the integration between the chimeric nanocontainer, RNAi technology and submersion application should be foreseen as one of the practical, powerful means to fight against prawn viral infection in field application.

ที่มาและความสำคัญ

งานวิจัยนี้มีวัตถุประสงค์เพื่อประเมินผลกระทบของสูตรอาหารกุ้งสามชนิด ซึ่งมีปริมาณปลาป่น (Fish Meal – FM) และกากถั่วเหลือง (Soybean Meal – SBM) ที่แตกต่างกัน (F1: FM สูง, F2: ผสม, F3: SBM สูง) ต่อการเจริญเติบโตและการทำงานของระบบทางเดินอาหารของกุ้ง โดยทำการทดลองเป็นระยะเวลา 28 วัน

Abstract

This study evaluated the growth and gut performance of shrimp fed three isonitrogenous diets (37% crude protein) with varying inclusions of fish meal (FM) and soybean meal (SBM): F1 (27.5% FM), F2 (10% FM + 23.5% SBM), and F3 (38% SBM). Over a 28-day period, feed intake, feed conversion ratio (FCR), and survival rates showed no significant differences among the groups. However, shrimp fed F2 and F3 exhibited significantly higher weight gain and average daily growth (ADG) compared to those fed F1 (P < 0.05). Gut performance analysis revealed that F3 consistently had the highest gut passage time (GPT), while F1 had the lowest. By day 28, shrimp fed F2 displayed elevated gut retention time (GRT). F1-fed shrimp showed a high gut passage rate (GPR), whereas F3-fed shrimp had a low GPR until day 21, with differences becoming negligible by day 28. Histological examination of the hepatopancreas revealed an increased R-cell population in shrimp fed F3. These findings highlight the adaptability of shrimp to different dietary compositions and underscore the importance of considering multiple factors when assessing the impacts of feed on growth and physiology.

จากการศึกษาเบื้องต้นของเรา พบว่าสารดึงดูดหรืออาจเป็นฟีโรโมนที่หลั่งออกมาจากกุ้งก้ามกรามเพศเมียที่อยู่ในระยะลอกคราบและพร้อมผสมพันธุ์ (Macrobrachium rosenbergii) สามารถกระตุ้นการแสดงออกของฮอร์โมนอินซูลินคล้ายแอนโดรจีนิกจากต่อมสร้างฮอร์โมนเพศผู้ในระบบการเพาะเลี้ยงร่วมกันได้

สารดึงดูดนี้จะถูกรับรู้โดยตัวรับกลิ่นที่มีขนรับความรู้สึก (setae) ซึ่งอยู่บนหนวดคู่หน้าด้านข้างที่สั้น (short lateral antennules หรือ slAn) ของกุ้งก้ามกรามเพศผู้ สัญญาณประสาทที่เกิดขึ้นนี้จะถูกส่งต่อไปยังศูนย์กลางระบบประสาท (central nervous system หรือ CNS) โดยมี นิวโรเปปไทด์ อย่างน้อย 4 ชนิดเป็นสื่อกลาง ได้แก่ neuropeptide F (NPF), short NPF (sNPF), tachykinin (TK), และ allatostatin-A (ATS-A) ซึ่งพบการแสดงออกของยีนที่สร้างนิวโรเปปไทด์เหล่านี้ในการศึกษาปัจจุบัน เพื่อทำความเข้าใจถึงบทบาทหน้าที่ของนิวโรเปปไทด์ในกลุ่มนี้ การศึกษาลักษณะการแสดงออกของยีนที่สัมพันธ์กับรูปแบบการเลี้ยงที่กล่าวข้างต้น

Abstract

Our previous studies revealed a mating attractant or possibly a pheromone released from molting reproductive mature female prawns, Macrobrachium rosenbergii, stimulates the expression of insulin-like androgenic gland hormones in a co-culture system. The released attractant is perceived by olfactory receptors with setae located on the short lateral antennules (slAn), which connect to the olfactory neuropil in the central nervous system (CNS) of male prawns. This neural signaling propagating through the CNS is mediated by at least four neuropeptides, namely neuropeptide F (NPF), short NPF (sNPF), tachykinin (TK), and allatostatin-A (ATS-A) whose transcripts have been detected in the present study. These deduced sequences, along with their conserved domains, serve as signatures of the identified neuropeptides, which were then compared with those found in other crustaceans and insects, whose nucleotide sequences were obtained from the nucleotide database. RT-PCR identified the expressions of the transcripts encoding these neuropeptides in the CNS. In situ hybridization specifically localized these transcripts in olfactory-associated neurons of cluster 9/11 of the deutocerebrum. Quantitative real-time PCR was used to quantify the expressions of the transcripts in response to the female attractants under different co-culture conditions: males with molting females (G1), males with intermolt females (G2), and slAn ablated males with molting females (G3). The transcripts were significantly increased on days 4–8 in the brain (Br) of males in G1 but not in G2 and G3. This suggests that expressions of the transcripts encoding the neuropeptides are associated with the perception of female mating pheromones through the slAn. This study is the first to show that female mating chemicals regulate the expressions and abundance of the olfactory neuropeptides, thus providing valuable insights for manipulation of mating of this species in aquaculture production.
