ที่มาและความสำคัญ

งานวิจัยนี้มีวัตถุประสงค์เพื่อประเมินผลกระทบของสูตรอาหารกุ้งสามชนิด ซึ่งมีปริมาณปลาป่น (Fish Meal – FM) และกากถั่วเหลือง (Soybean Meal – SBM) ที่แตกต่างกัน (F1: FM สูง, F2: ผสม, F3: SBM สูง) ต่อการเจริญเติบโตและการทำงานของระบบทางเดินอาหารของกุ้ง โดยทำการทดลองเป็นระยะเวลา 28 วัน

Abstract

This study evaluated the growth and gut performance of shrimp fed three isonitrogenous diets (37% crude protein) with varying inclusions of fish meal (FM) and soybean meal (SBM): F1 (27.5% FM), F2 (10% FM + 23.5% SBM), and F3 (38% SBM). Over a 28-day period, feed intake, feed conversion ratio (FCR), and survival rates showed no significant differences among the groups. However, shrimp fed F2 and F3 exhibited significantly higher weight gain and average daily growth (ADG) compared to those fed F1 (P < 0.05). Gut performance analysis revealed that F3 consistently had the highest gut passage time (GPT), while F1 had the lowest. By day 28, shrimp fed F2 displayed elevated gut retention time (GRT). F1-fed shrimp showed a high gut passage rate (GPR), whereas F3-fed shrimp had a low GPR until day 21, with differences becoming negligible by day 28. Histological examination of the hepatopancreas revealed an increased R-cell population in shrimp fed F3. These findings highlight the adaptability of shrimp to different dietary compositions and underscore the importance of considering multiple factors when assessing the impacts of feed on growth and physiology.

ที่มาและความสำคัญ

จากการศึกษาเบื้องต้นของเรา พบว่าสารดึงดูดหรืออาจเป็นฟีโรโมนที่หลั่งออกมาจากกุ้งก้ามกรามเพศเมียที่อยู่ในระยะลอกคราบและพร้อมผสมพันธุ์ (Macrobrachium rosenbergii) สามารถกระตุ้นการแสดงออกของฮอร์โมนอินซูลินคล้ายแอนโดรจีนิกจากต่อมสร้างฮอร์โมนเพศผู้ในระบบการเพาะเลี้ยงร่วมกันได้

สารดึงดูดนี้จะถูกรับรู้โดยตัวรับกลิ่นที่มีขนรับความรู้สึก (setae) ซึ่งอยู่บนหนวดคู่หน้าด้านข้างที่สั้น (short lateral antennules หรือ slAn) ของกุ้งก้ามกรามเพศผู้ สัญญาณประสาทที่เกิดขึ้นนี้จะถูกส่งต่อไปยังศูนย์กลางระบบประสาท (central nervous system หรือ CNS) โดยมี นิวโรเปปไทด์ อย่างน้อย 4 ชนิดเป็นสื่อกลาง ได้แก่ neuropeptide F (NPF), short NPF (sNPF), tachykinin (TK), และ allatostatin-A (ATS-A) ซึ่งพบการแสดงออกของยีนที่สร้างนิวโรเปปไทด์เหล่านี้ในการศึกษาปัจจุบัน เพื่อทำความเข้าใจถึงบทบาทหน้าที่ของนิวโรเปปไทด์ในกลุ่มนี้ การศึกษาลักษณะการแสดงออกของยีนที่สัมพันธ์กับรูปแบบการเลี้ยงที่กล่าวข้างต้น

Abstract

Our previous studies revealed a mating attractant or possibly a pheromone released from molting reproductive mature female prawns, Macrobrachium rosenbergii, stimulates the expression of insulin-like androgenic gland hormones in a co-culture system. The released attractant is perceived by olfactory receptors with setae located on the short lateral antennules (slAn), which connect to the olfactory neuropil in the central nervous system (CNS) of male prawns. This neural signaling propagating through the CNS is mediated by at least four neuropeptides, namely neuropeptide F (NPF), short NPF (sNPF), tachykinin (TK), and allatostatin-A (ATS-A) whose transcripts have been detected in the present study. These deduced sequences, along with their conserved domains, serve as signatures of the identified neuropeptides, which were then compared with those found in other crustaceans and insects, whose nucleotide sequences were obtained from the nucleotide database. RT-PCR identified the expressions of the transcripts encoding these neuropeptides in the CNS. In situ hybridization specifically localized these transcripts in olfactory-associated neurons of cluster 9/11 of the deutocerebrum. Quantitative real-time PCR was used to quantify the expressions of the transcripts in response to the female attractants under different co-culture conditions: males with molting females (G1), males with intermolt females (G2), and slAn ablated males with molting females (G3). The transcripts were significantly increased on days 4–8 in the brain (Br) of males in G1 but not in G2 and G3. This suggests that expressions of the transcripts encoding the neuropeptides are associated with the perception of female mating pheromones through the slAn. This study is the first to show that female mating chemicals regulate the expressions and abundance of the olfactory neuropeptides, thus providing valuable insights for manipulation of mating of this species in aquaculture production.

Abstract

The hematopoietic tissue (HPT) and anterior proliferation center (APC) are the main hemocyte-producing organs of the freshwater crayfish, Pacifastacus leniusculus. To deepen our understanding of immune responses to various pathogens, it is essential to identify distinct hemocyte subpopulations with specific functions and to further explore how these cells are generated. Here we provide an in-depth histological study of the HPT and APC in order to localize cell types in different developmental stages, and to provide some information regarding the hemocyte differentiation in the crayfish. We localized mRNA expression of previously identified genes in the HPT/APC and hemocytes by RNA-FISH. The expression of hemolectin and transglutaminase 1 was shown to be co-localized in a high number of the HPT cells, while transglutaminase 2 was expressed in different cell types mainly associated with epithelium or endothelium. Furthermore, by double RNA-FISH for hemolectin and a previously unidentified PDGF-like factor, combined with immunostaining for prophenoloxidase, we could identify several different subtypes of hemocytes, indicating that the immune function of hemocytes in crayfish is more diversified and complex than previously appreciated.

Abstract

The crayfish plague is caused by the oomycete Aphanomyces astaci with North American crayfish (for example Pacifastacus leniusculus and Procambarus clarkii) serving as carriers and vectors for this pathogen. This poses a constant threat to native crayfish in Europe, Asia, South America and Australia, which all are highly susceptible to this pathogen. In this study we now show how the symbiotic balance between the pathogen and its host are maintained at the molecular level. The host factors involved in this balance between the pathogen, A. astaci and the host, P. leniusculus, are one glycine-rich antimicrobial peptide (GRP) that is specifically active against A. astaci, but not to other microorganisms and two Kazal proteinase inhibitors (KPI2 and KPILA) inhibit secreted A. astaci proteases by binding to subtilisin enzymes from the pathogen.Accordingly, the expression of GRP, KPI2, KPILA, as well as proPO mRNAs increases following A. astaci infection. Silencing GRP, or KPI2 + KPILA mRNAs results in death of the crayfish from infection. Over time, this host-pathogen relationship has evolved to allow resistant crayfish to coexist with A. astaci in their cuticle for life, provided critical components remain unaltered by environmental changes or other pathogens. It is unclear whether a similar relationship could develop between currently susceptible crayfish and A. astaci.

Abstract

Tilapia tilapinevirus or tilapia lake virus (TiLV) is an emerging virus that inflicts significant mortality on farmed tilapia globally. Previous studies reported detection of the virus in multiple organs of the infected fish; however, little is known about the in-depth localization of the virus in the central nervous system. Herein, we determined the distribution of TiLV in the entire brain of experimentally infected Nile tilapia. In situ hybridization (ISH) using TiLV-specific probes revealed that the virus was broadly distributed throughout the brain. The strongest positive signals were dominantly detected in the forebrain (responsible for learning, appetitive behaviour and attention) and the hindbrain (involved in controlling locomotion and basal physiology). The permissive cell zones for viral infection were observed mostly to be along the blood vessels and the ventricles. This indicates that the virus may productively enter into the brain through the circulatory system and widen broad regions, possibly through the cerebrospinal fluid along the ventricles, and subsequently induce the brain dysfunction. Understanding the pattern of viral localization in the brain may help elucidate the neurological disorders of the diseased fish. This study revealed the distribution of TiLV in the whole infected brain, providing new insights into fish–virus interactions and neuropathogenesis.

Tilapia tilapinevirus หรือ tilapia lake virus (TiLV) เป็นไวรัสอุบัติใหม่ที่สร้างความเสียหายให้กับปลานิลที่เลี้ยงในฟาร์มทั่วโลก การศึกษาก่อนหน้านี้รายงานการตรวจพบไวรัสในหลายอวัยวะของปลาที่ติดเชื้อ อย่างไรก็ตาม ยังไม่ค่อยมีความรู้เชิงลึกถึงการกระจายตัวของไวรัสชนิดนี้ในระบบประสาทส่วนกลางของปลา งานวิจัยนี้ได้ศึกษาการกระจายตัวของไวรัส TiLV ในสมองทุกส่วนของปลานิลที่ถูกทำให้ติดเชื้อในห้องปฏิบัติการ ผลการศึกษาด้วยเทคนิค in situ hybridization โดยใช้โพรบ TiLV จำเพาะ พบไวรัสอยู่ในทุกส่วนทั่วสมอง ตำแหน่งที่ให้ผลบวกที่เด่นชัดที่สุดอยู่ในสมองส่วนหน้า (รับผิดชอบการเรียนรู้ การอยากอาหาร และความสนใจ) และสมองส่วนหลัง (เกี่ยวข้องกับการควบคุมการเคลื่อนไหวและสรีรวิทยาพื้นฐาน) บริเวณเซลล์ติดเชื้อโดยส่วนใหญ่อยู่ตามหลอดเลือดและโพรงสมอง ข้อมูลเหล่านี้บ่งชี้ว่าไวรัสอาจเข้าสู่สมองผ่านระบบไหลเวียนเลือดและขยายขอบเขตกว้างออกไป โดยอาจผ่านทางน้ำหล่อเลี้ยงสมองไปตามโพรงสมอง จนทำให้เกิดความผิดปกติของสมองในที่สุด การทำความเข้าใจรูปแบบของการกระจายตัวของไวรัสในสมองอาจช่วยอธิบายได้ถึงความผิดปกติของระบบประสาทของปลาป่วย การศึกษาครั้งนี้เผยให้เห็นการกระจายของ TiLV ในสมองปลา ซึ่งเป็นข้อมูลใหม่เชิงลึกเกี่ยวกับปฏิสัมพันธ์ระหว่างปลากับไวรัส และการเกิดโรคทางระบบประสาทของปลา

ปูม้า (Portunus pelagicus) เป็นสัตว์เศรษฐกิจที่มีการบริโภคกันในหลายประเทศรวมถึงประเทศไทย ดังนั้นการศึกษาวิจัยที่เกี่ยวข้องกับระบบสืบพันธุ์ของปูม้าจึงมีความสำคัญต่อเกษตรกรผู้เลี้ยงปูม้าเพื่อสามารถนำไปประยุกต์ใช้เพิ่มผลผลิตในการเลี้ยงปูม้าต่อไป ซึ่งมีการศึกษาก่อนหน้าพบว่ากรดอะมิโนสายสั้นๆ ที่เรียกว่า oxytocin/vasopressin-like peptide (OT/VP-like peptide) มีผลต่อการยับยั้งการหลั่งฮอร์โมนสเตอรอยด์จากรังไข่ งานวิจัยครั้งนี้จึงมุ่งเน้นเพื่อศึกษาหาลำดับสารพันธุกรรมของตัวรับ OT/VP-like peptide (OT/VP-like peptide receptor) และทำให้ค้นพบลำดับสารพันธุกรรม mRNA ที่มีการแปลรหัสเป็นตัวรับของ OT/VP-like peptide ในปูม้าได้สำเร็จ จึงสามารถทำนายลักษณะโครงสร้างของโปรตีนตัวรับจากลำดับโปรตีนที่ถอดรหัสออกมาได้ และพบว่ามีการแสดงออกของตัวรับนี้สูงสุดในสมองของปูม้าแต่ไม่มีความแตกต่างกันในการแสดงออกในไข่อ่อนและไข่แก่ ในขณะที่ OT/VP-like peptide มีการแสดงออกมากที่ปมประสาทส่วนท้องของปูม้าไข่อ่อนและไข่แก่ ซึ่งคาดว่าโมเลกุลที่อาจส่งผลต่อการยับยั้งการหลั่งฮอร์โมนสเตอรอยด์จากรังไข่น่าจะเป็นผลมาจาก OT/VP-like peptide เท่านั้น ดังนั้นหากทราบกลไกลที่ระเอียดขึ้นของตัวรับและ OT/VP-like peptide ในปูม้าอาจจะสามารถนำไปประยุกต์ใช้กระตุ้นระบบสืบพันธุ์ของปูม้าเพศเมียเพื่อเพิ่มผลผลิตต่อไปได้

Abstract 
The authors recently reported the presence and distribution of oxytocin/vasopressin-like peptide in Portunus pelagicus as well as demonstrated its function to inhibit ovarian steroid release (Saetan et al., 2018). Here, the full-length receptor of this peptide, namely oxytocin/vasopressin-like peptide receptor (PpelOT/VP-like peptide receptor) is reported. The coding region of the PpelOT/VP-like peptide receptor contained 1497 bp which translationally corresponded to 499 amino acids. Sequence analysis revealed its seven transmembrane characteristics, with -two N-linked glycosylation residues located before the first transmembrane domain (TM I). The phylogenetic tree revealed that the PpelOT/VP-like peptide receptor was placed in the group of invertebrate OT/VP-like receptors, and was clearly distinguishable from the V1R, V2R and OTR of vertebrates. Also, this receptor gene transcript was detected in several organs of the blue swimming crab with highest abundance found in brain tissue. In situ hybridization exhibited its distribution in all neuronal clusters of the eyestalk, brain, ventral nerve cord (VNC), as well as in the ovary. Comparative gene expressions between this receptor and its corresponding peptide in immature and mature female crabs revealed no significant difference of the PpelOT/VP-like peptide receptor gene expression in the central nervous system (CNS) and ovary. In contrast, the PpelOT/VP-like peptide gene was shown to significantly express higher in the VNC of immature crabs and in the ovary of mature crabs. Changes in expression of this peptide gene, but not its receptor, might result in ovarian steroid release inhibition. However, the detailed mechanism of this peptide in reproduction regulation will be included in our further studies.
