ช่องกระดูกขากรรไกร (mandibular canal) เป็นช่องที่สำคัญในการส่งผ่านเส้นประสาทและหลอดเลือดในขากรรไกรล่าง แต่รูปแบบความแปรผันของช่องกระดูกขากรรไกรยังมีรายงานไม่มากนัก การศึกษานี้รายงานความแปรผันทางกายวิภาคที่ไม่เคยอธิบายมาก่อนของช่องกระดูกขากรรไกรแบบไม่สมบูรณ์ทั้งสองข้างในกระดูกขากรรไกรจากซากศพชาย ด้วยการตรวจทางกายวิภาคและภาพเอกซเรย์คอมพิวเตอร์ จากนั้นใช้ชิ้นเนื้อจากตัวอ่อนมนุษย์เพื่อสร้างแบบจำลองสามมิติ วิเคราะห์ความสัมพันธ์ระหว่างกระดูกขากรรไกรที่กำลังพัฒนา เส้นประสาทขากรรไกรล่าง และต่อมน้ำลาย ผลการศึกษาสนับสนุนสมมติฐานว่า การก่อตัวของช่องที่ไม่สมบูรณ์อาจเกิดจากตำแหน่งของเส้นประสาทหรือการถูกกักขังของเนื้อเยื่อต่อมน้ำลายในระหว่างการพัฒนา ความแปรผันนี้มีความสำคัญต่อการตีความภาพทางรังสีและการวางแผนผ่าตัดขากรรไกรล่างในทางคลินิก

Abstract

Purpose

This study aims to report a previously undocumented anatomical variation of the mandible and investigate its developmental origin using embryological analysis.

Methods

Anatomical inspection and computed tomography were performed on a dried human mandible from a male cadaver of unknown age. Additionally, serial histological sections from a Carnegie Stage 23 human embryo were reconstructed in 3D model to examine the spatial relationships between the developing mandible and surrounding structures.

Results

The mandibular canal exhibited breaching of the medial wall into the lingual cortex on both sides. Computed tomography confirmed that the canals originated from the mandibular foramen and gave rise to the mental foramina at the medial third of the mandible. These defects were found along with a left-sided Stafne bone cavity. Embryological analysis revealed a close spatial proximity between the developing mandible, the inferior alveolar nerve, and the salivary glands, supporting the hypothesis that incomplete ossification could result from variant nerve positioning or glandular entrapment.

Conclusion

This study presents rare anatomical variations of the mandibular canal and their possible developmental mechanism. These findings have implications for radiological interpretation and surgical planning involving the mandible.

Abstract

Osteoporosis is characterized by an imbalance between osteoblastic bone formation and osteoclastic bone resorption, leading to an increased risk of fractures. The water-soluble matrix (WSM) of nacre exhibits osteoinductive properties in osteoblastic cells, both in vitro and in vivo. However, its release from natural nacre remains challenging due to its solid and compact surface. This study aimed to prepare nano-nacre particles with smaller diameters than intact aragonite crystals to enhance WSM release and to investigate its effects on osteoblast differentiation. Size analysis and SEM imaging showed that the nano-nacre particles had an average size of about 600 nm. Furthermore, their effects on osteoblast differentiation and mineralization were evaluated through qPCR and ARS assay. The results showed that WSM significantly upregulated key osteogenic genes, including RUNX2, ALP, and OCN, in a dose- and time-dependent manner over 14 days, with fold-changes ranging from 1.6 to 3.6. Additionally, the mineralization effects showed calcium deposition levels comparable to those of the positive group. These findings suggest that WSM may be a promising soluble factor for osteoblast differentiation and mineralization. Therefore, understanding the effects of the WSM from H. diversicolor nano-nacre particles on osteoblasts in vitro may provide evidence suggesting that it could be a promising anti-osteoporosis agent.

ในการศึกษานี้ ได้พัฒนาอนุภาคลูกผสม โดยนำโปรตีนเปลือกไวรัส (virus-like particles; VLPs) ของไวรัสกุ้งก้ามกราม (MrNV) มาแทนที่ส่วนยื่น (protruding domains) ด้วยโดเมน(tsCRD) ที่สามารถจับกับกรดไซลิอาลิก ที่พัฒนามาจากเลคติน SNA-I โดยให้ชื่ออนุภาคลูกผสมนี้ว่า (tsCRD-MrNV-VLPs) ส่วนยื่นที่นำมาแทนที่นี้ มีบทบาทในการจับกับโปรตีนหนามของไวรัส SARS-CoV-2 ซึ่งเมื่อผลิตออกมาได้แล้ว พบว่าอนุภาคลูกผสมมีความ เสถียรและสามารถผลิตได้ในระบบแบคทีเรีย มีความสามารถในการจับและเข้าสู่เซลล์ HEK293T ที่แสดงโปรตีน ACE2 สูง (HEK293T-hACE2) ซึ่งเซลล์นี้เป็นเซลล์เป้าหมายของ SARS-CoV-2 จากการทดลองพบว่าอนุภาคลูกผสมสามารถลดการติดเชื้อของไวรัส SARS-CoV-2 สายพันธุ์ Wuhan, Delta และ Omicron ในเซลล์ได้อย่างมีนัยสำคัญ. ผลการศึกษานี้ชี้ให้เห็นว่า tsCRD-MrNV-VLPs อาจพัฒนานำมาใช้ในการป้องกันหรือรักษาการติดเชื้อไวรัสที่ใช้กลไกการจับกับกรดไซลิอาลิก เช่น SARS-CoV-2 ได้

ในงานวิจัยนี้ เราได้สร้างอนุภาคเลียนแบบไวรัส (เรียกว่า VLP) ซึ่งมีรูปร่างคล้ายไวรัสจริงแต่ไม่มีสารพันธุกรรม จึงไม่สามารถก่อโรคได้ โดยใช้ไวรัสจากกุ้งก้ามกรามที่ไม่ติดต่อสู่มนุษย์ แล้วเปลี่ยนส่วนหนึ่งของเปลือกไวรัสให้เป็นส่วนที่สามารถจับกับกรดไซลิอาลิกได้ดี ซึ่งได้มาจากเลคตินชนิดหนึ่งชื่อ SNA-I จากการสร้างอนุภาคลูกผสมนี้ขึ้นมาพบว่า อนุภาคใหม่นี้สามารถจับกับเซลล์มนุษย์ที่มี ACE2 ได้ดี และยังช่วยลดการติดเชื้อของไวรัสโควิดแบบจำลองได้อีกด้วย ผลลัพธ์นี้แสดงให้เห็นว่าอนุภาคเหล่านี้อาจช่วยป้องกันไวรัสโควิดไม่ให้เข้ามาในเซลล์ และอาจนำไปพัฒนาต่อยอดเป็นการรักษาหรือป้องกันในอนาคตได้ เพราะผลิตง่าย ราคาถูก และมีความเสถียรสูง

Abstract

SARS-CoV-2, the cause of COVID-19, primarily targets lung tissue, leading to pneumonia and lung injury. The spike protein of this virus binds to the common receptor on susceptible tissues and cells called the angiotensin-converting enzyme-2 (ACE2) of the angiotensin (ANG) system. In this study, we produced chimeric Macrobrachium rosenbergii nodavirus virus-like particles, presenting a short peptide ligand (ACE2tp), based on angiotensin-II (ANG II), on their outer surfaces to allow them to specifically bind to ACE2-overexpressing cells called ACE2tp-MrNV-VLPs. Replacing the ACE2tp at the protruding domains (P-domain) of the MrNV capsid proteins did not affect their normal assembly into icosahedral VLPs. The presentation of the ACE2tp on the P-domains significantly improved the binding and internalization of ACE2tp-MrNV-VLPs to hACE2-overexpressing HEK293T cells in a concentration-dependent manner. Furthermore, ACE2tp-MrNV-VLPs exhibited the ability to block the binding and infection of SARS-CoV-2 pseudovirus variants, including Wuhan, BA.2 Omicron, and Delta subtypes. Our results suggest that chimeric ACE2tp-MrNV-VLPs can serve as a blocking agent against various SARS-CoV-2 mutated variants and could also potentially serve as target-specific nano-containers to carry therapeutic agents to combat SARS-CoV-2 infections in the future.

Abstract

Sperm activation occurring in both male and female reproductive tract involves a highly complex series of biomolecular dynamics, particularly on membrane lipids and proteins. In mammals, the universal anticipation in cholesterol (CHO) sequestration plays a role in mammalian sperm maturation/capacitation, subsequently enhancing sperm fertilizing ability. In shrimp, we have previously shown that the level of cholesterol (CHO) is significantly reduced in vas deferens sperm when compared with sperm in the testes, presumably due to the sequestering action of the lipid-binding protein, one of which is Niemann-Pick Type C-2 (NPC2). Here, we used recombinant MrNPC2 protein to treat testicular sperm (Tsp) and quantitatively compared the amount of CHO with non-treated or 2-OH-β-CD treated sperm. HPTLC of the extracted lipids from Macrobrachium rosenbergii sperm revealed the presence of major phospholipids and CHO. Tsp treated with 2-OH-β-CD or MrNPC2 showed lower CHO levels, which was comparable to that of the vas deferens sperm as verified by the Amplex Red assay. Finally, the enhanced levels of protein tyrosine phosphorylation and ionophore-induced AR levels in the MrNPC2 treated Tsp significantly increased in a concentration-dependent manner, similar to that of the Vsp sperm. Altogether, our results indicated the importance of MrNPC2 as CHO moderator in sperm membrane, leading to an enhancement in sperm fertilizing ability.

ในปัจจุบันได้มีพัฒนาเทคนิคใหม่ในการป้องกันกุ้งจากไวรัส โดยใช้วิธี RNA interference (RNAi) โดยการฉีดหรือผสมในอาหารที่ค่อยข้างยุ่งยาก ทีมวิจัยจึงได้พัฒนาการบรรจุสารพันธุกรรม dsRNA ลงใน อนุภาคเลียนแบบไวรัส (VLPs) แล้วให้กุ้งดูดซึมผ่านการแช่น้ำ แทนการฉีดเข้าไปในตัวกุ้งซึ่งทำได้ง่ายขึ้นต่อเกษตรกรผู้เพาะเลี่บงกุ้ง

Abstract

One of the most successful applications to protect aquatic animals from viral infection is the use of RNA interference (RNAi) in the form of double-stranded RNA (dsRNA). However, its administration into animals is usually via injection route (laboratory scale) or mixing feed pellets with bacterial extracts (containing therapeutic compounds); the outcome of which remains to be improved. In this study, we encapsulated purified dsRNA into a virus-like particle (VLP) and delivered it into the prawn tissues by a submersion route using interiorly modified Macrobrachium rosenbergii nodavirus derived VLP with deca-arginine peptide (10R-MrN-VLP) to encapsulate a large scale (gram-level) of dsRNA against RNA2 gene of MrNV capsid. For tracing purposes of submersion administration, GFP plasmid-loaded VLP successfully delivered and expressed green fluorescent protein in prawn-targeted tissues. Delivery efficiency at 12-h submersion was about 50% of the prawn population and became 100% within 72 h, and they remained GFP-positive for up to 7 days during submersion. The protective effect of dsRNA-VLP submersion was tested with a large group (10,000) of post-larvae, which were naturally challenged with MrNV-positive dead prawn. Transcriptome analysis revealed up- and down-regulations of gene clusters involved in metabolism and signal transduction related to an antiviral immune system. The survival rate of prawn treated with dsRNA-VLP reached 80% at Day 7 post-MrNV challenge, significantly different from that of the MrNV-challenged group (control). PCR screening revealed an apparent decrease of MrNV genes and the copy number of MrNV in prawn sampling tissues. Together, we believe that the integration between the chimeric nanocontainer, RNAi technology and submersion application should be foreseen as one of the practical, powerful means to fight against prawn viral infection in field application.

Abstract

Purpose

While Thiel embalming effectively preserves cadavers in a lifelike and flexible state, its implementation is constrained by high costs and the use of ammonium nitrate, which may be restricted in certain countries due to regulatory controls and safety concerns. This study aims to develop a more economical and safer approach to Thiel embalming by using technical grade chemicals and synthesizing ammonium nitrate on-site and, both overcoming economic barriers to Thiel embalming and addressing regulatory constraints associated with ammonium nitrate.

Methods

In the present study, technical grades were used instead of reagent grades. Ammonium nitrate was synthesized through a controlled reaction of nitric acid and ammonium hydroxide under strict safety protocols. The solution was incorporated into Thiel embalming fluids and used for arterial and venous embalming of 28 cadavers at two institutions.

Results

The cadavers embalmed using the methods presented in this study were comparable to conventional Thiel-embalmed cadavers, preserving tissue flexibility, color, and structural integrity. The only notable difference was the reduced submersion duration, which was limited to three to four months.and the technique effectively mitigated the hazards of ammonium nitrate storage.

Conclusion

The use of technical grade chemicals, combined with on-site production of ammonium nitrate, presents a more cost-effective alternative for Thiel embalming, particularly in regions with stringent regulations, making Thiel embalming more accessible for anatomy departments worldwide.

Abstract

SARS-CoV-2, the cause of COVID-19, primarily targets lung tissue, leading to pneumonia and lung injury. The spike protein of this virus binds to the common receptor on susceptible tissues and cells called the angiotensin-converting enzyme-2 (ACE2) of the angiotensin (ANG) system. In this study, we produced chimeric Macrobrachium rosenbergii nodavirus virus-like particles, presenting a short peptide ligand (ACE2tp), based on angiotensin-II (ANG II), on their outer surfaces to allow them to specifically bind to ACE2-overexpressing cells called ACE2tp-MrNV-VLPs. Replacing the ACE2tp at the protruding domains (P-domain) of the MrNV capsid proteins did not affect their normal assembly into icosahedral VLPs. The presentation of the ACE2tp on the P-domains significantly improved the binding and internalization of ACE2tp-MrNV-VLPs to hACE2-overexpressing HEK293T cells in a concentration-dependent manner. Furthermore, ACE2tp-MrNV-VLPs exhibited the ability to block the binding and infection of SARS-CoV-2 pseudovirus variants, including Wuhan, BA.2 Omicron, and Delta subtypes. Our results suggest that chimeric ACE2tp-MrNV-VLPs can serve as a blocking agent against various SARS-CoV-2 mutated variants and could also potentially serve as target-specific nano-containers to carry therapeutic agents to combat SARS-CoV-2 infections in the future.

Abstract

Background

Application of a virus-like particle (VLP) as a nanocontainer to encapsulate double stranded (ds)RNA to control viral infection in shrimp aquaculture has been extensively reported. In this study, we aimed at improving VLP’s encapsulation efficiency which should lead to a superior fighting weapon with disastrous viruses.

Results

We constructed 2 variants of chimeric Macrobrachium rosenbergii nodavirus (MrNV)-like particles (V1- and V2-MrN-VLPs) and tested their efficiency to encapsulate VP37 double stranded RNA as well as WSSV protection in P. vannamei. Two types of short peptides, RNA-binding domain (RBD) and deca-arginine (10R) were successfully engineered into the interior surface of VLP, the site where the contact with VP37-dsRNA occurs. TEM and dynamic light scattering (DLS) analyses revealed that the chimeric VLPs remained their assembling property to be an icosahedral symmetric particle with a diameter of about 30 nm, similar to the original MrN-VLP particle. The superior encapsulation efficiency of VP37-dsRNA into V2-MrN-VLP was achieved, which was slightly better than that of V1-MrN-VLP but far better (1.4-fold) than its parental V0-MrN-VLP which the mole ratio of 7.5–10.5 for all VLP variants. The protection effect against challenging WSSV (as gauged from the level of VP37 gene and the remaining viral copy number in shrimp) was significantly improved in both V1- and V2-MrN-VLP compared with an original V0-MrN-VLP template.

Conclusion

MrN-VLP (V0-) were re-engineered interiorly with RBD (V1-) and 10R (V2-) peptides which had an improved VP37-dsRNA encapsulation capability. The protection effect against WSSV infection through shrimp administration with dsRNA + V1-/V2-MrN VLPs was experimentally evident.

Abstract

Mannose receptor (MR) is a transmembrane protein and a type of pattern-recognition receptor (PRR) that plays a critical role in the immunity of mammals and fish. In this study, we examined the role of MR in binding with infectious hypodermal and haematopoietic necrosis virus-like particle (IHHN-VLP) and the downstream immune pathway that it triggers in the shrimp Peneaus vannamei. Upon IHHN-VLP challenge, transcripts of MR in P. vannamei (PvMR) increased significantly in all examined tissues, particularly those related to shrimp immunity, including hemocyte, hepatopancreas and gill tissues. Specifically, IHHN-VLP bound to the 34-kDa PvMR protein in shrimp-tissue extracts. Immunohistochemistry results of hemocytes showed that PvMR was initially localised on the plasma membrane but later internalised and dispersed throughout the cytoplasm after IHHN-VLP administration. Binding between IHHN-VLP and PvMR also induced significant upregulation of genes for the antimicrobial peptides (AMPs) penaeidin 3 and crustin, presumably to protect the shrimp against the viral infection. However, knocking down PvMR resulted in down-regulation of all immune-related genes examined. Overall, as an immune-related PRR, PvMR serves as a receptor for invading viruses, which then trigger the expression of AMPs. Strategic designs using PvMR could be developed to either block the interaction of native virus with the host cells or provoke its up-regulation to enhance shrimp immunity, which could open up opportunities to fight against IHHNV infection in shrimp.

Abstract

Although betulinic acid (BA) has been shown to attenuate breast cancer cell lines, owing to its interaction with several signaling molecules; its potential interaction with estrogen receptors (ERs) and p53 is not fully understood. Hence, we aimed to investigate the anti-cancer effect of BA on breast cancer cells, focusing on its molecular mechanisms involving the ER and p53 signaling pathways. The cell cytotoxicity of ER-positive (MCF-7) and ER-positive (MDA-MB231) breast cancer cells was studied using MTT assay. Apoptosis was investigated by flow cytometry and Western blot analysis. The expression levels of ER?/ER? and wt-p53/mu-p53 were studied using Western blotting. Finally, a possible interaction between BA and its molecular targets was predicted using molecular docking. Upon BA treatment, both breast cancer cell lines underwent significant cell death and inhibition of cell proliferation. Flow cytometry and Western blot analysis showed that the MCF-7 cells underwent early and late apoptosis, while MDA-MB-231 underwent both apoptosis and necrosis within 48 h. The expression levels of ER?/ER? and wt-p53/mu-p53 were significantly altered. This could be partly attributed to the activation of apoptosis and inhibition of proliferation through the p53 signaling pathway, as induced by the interaction of BA with its coupling molecules.
