Abstract

Sulfated polysaccharides from marine macroalgae are increasingly recognized as high-value bioproducts with diverse biofunctional properties. This study investigated sulfated galactan (SG) extracted from the red alga Gracilaria fisheri (G. fisheri) as a gut-modulating algal biopolymer and evaluated its structural stability, microbial selectivity, and effects on intestinal health. SG exhibited a high molecular weight (259 kDa) that remained largely unchanged during simulated oral, gastric, and intestinal digestion, indicating strong resistance to enzymatic and acidic degradation. In vitro fermentation showed that SG selectively stimulated the growth of the probiotics Lactobacillus casei and Lactobacillus plantarum and enhanced β-galactosidase activity. Molecular docking further demonstrated promising interactions between SG subunits and β-galactosidase catalytic residues. An intestinal permeability assay confirmed that SG did not traverse the Caco-2 monolayer, suggesting that it remains unabsorbed and available for probiotic fermentation upon reaching the colon. Consistent with these in vitro results, SG supplementation in mice revealed improved colonic epithelial proliferation and enhanced mucosal architecture, supporting its benefits to gut barrier integrity. Taken together, these findings identify SG as a promising high-value algal bioproduct derived from G. fisheri with potential applications in gut health.

Abstract

Staphylococcus epidermidis is a leading opportunistic pathogen in medical device-associated infections due to its ability to adhere to abiotic materials and develop biofilms that are difficult to eradicate. This study investigated the antibiofilm potential of an ethanolic extract of the red seaweed Gracilaria fisheri and its purified constituent, N-benzylcinnamamide, against S. epidermidis. Antibacterial activity was determined, and antibiofilm effects were assessed using the crystal violet assay and confocal laser scanning microscopy (CLSM). Early bacterial adhesion on glass and polyurethane (PU) surfaces was measured. The effect on catheter-associated biofilms was evaluated by scanning electron microscopy (SEM). Transcripts of biofilm- and quorum-sensing-associated genes (icaA and luxS) were assessed by semi-quantitative RT-PCR. Cytotoxicity was evaluated by MTT assay. At 200 µg/mL, biofilm biomass decreased to 48.21 ± 5.52% with the extract and to 36.65 ± 6.82% with N-benzylcinnamamide. CLSM time-course imaging showed delayed biofilm maturation and less consolidated, discontinuous structures. Surface exposure to the extract markedly reduced early attachment on both materials. On PU catheter segments, SEM demonstrated that N-benzylcinnamamide markedly reduced surface coverage and disrupted three-dimensional biofilm architecture. At the molecular level, transcription of icaA and luxS was reduced. Both the extract and N-benzylcinnamamide showed minimal cytotoxicity in HeLa cells. These findings support further evaluation of these marine-derived agents as candidates for antibiofilm surface treatments to reduce early medical device colonization.

Abstract

Psoriasis is a chronic immune‑mediated inflammatory skin disorder characterized by keratinocyte hyperproliferation and immune cell infiltration. Low molecular weight sulfated galactan (LSG), derived from the red seaweed Gracilaria fisheri, has immunomodulatory potential relevant to psoriasis pathogenesis. The present study investigated the therapeutic potential of LSG using an imiquimod (IMQ)‑induced murine model of psoriasis. BALB/c mice received intraperitoneal administration of LSG once daily, 2 h prior to IMQ application, for 7 consecutive days. Clinical severity was assessed using the Psoriasis Area and Severity Index (PASI). Histopathological and immunohistochemical analyses were performed to evaluate epidermal architecture, vascular change and immune cell infiltration. Expression of keratinocyte proliferation and differentiation markers, proinflammatory cytokines and JAK/STAT pathway components was analyzed using reverse transcription‑quantitative PCR, ELISA and western blotting. Systemic inflammation was assessed by spleen and lymph node size. LSG significantly decreased PASI scores, neovascularization, epidermal thickness and dermal inflammation. LSG downregulated keratinocyte proliferation and differentiation markers (Ki67, keratin 6, 16 and 17, involucrin) and decreased proinflammatory cytokine expression (TNF‑α, IL‑1β, IL‑17A, IL‑23, IL‑6), accompanied by decreased CD4+ T and mast cell infiltration. LSG downregulated JAK2/STAT2/STAT3 signaling and downstream genes (BCL2, CCND1). Furthermore, LSG alleviated systemic inflammation without inducing hepatic or renal toxicity. These findings indicated that LSG effectively ameliorates IMQ‑induced psoriatic inflammation via coordinated reduction of keratinocyte hyperproliferation, cytokine production and immune cell activity. LSG represents a promising marine‑derived therapeutic candidate for psoriasis management.

Abstract

Breast cancer is one of the most prevalent diseases affecting the female population, with its incidence increasing globally. Previous studies have identified cyclins, CDKs and upstream signaling molecules as key therapeutic targets, as their overexpression can drive the transformation of normal cells into cancerous ones. Sulfated galactan (SG), a polysaccharide derived from Gracilaria fisheri, has demonstrated potential in modulating cellular functions. Recent research suggests that low molecular weight SG (LSG), when supplemented with an octanoyl ester (LSGO), exhibits an enhanced biological activity. However, the anticancer effects of SG and its derivatives in breast cancer remain underexplored. The present study thus aimed to examine the effects of SG, LSG and LSGO on MCF‑7 breast cancer cells. Cytotoxicity was initially assessed in L929 normal fibroblast cells and MCF‑7 cells. While all three forms were non‑toxic to L929 cells, LSGO exhibited slight cytotoxicity and significantly induced cell cycle arrest at the G2/M phase. Mechanistically, LSGO suppressed the PI3K/AKT/mTOR and ERK pathways, downregulated cyclins and CDKs, and led to cell cycle arrest and reduced cell proliferation. These results suggest that the structural modification of SG enhances its anti‑proliferative capacity, highlighting LSGO as a promising candidate for the treatment of MCF‑7 cells. Overall, these findings provide insight into the molecular mechanisms by which SG derivatives affect breast cancer cell proliferation and underscore their potential as anti‑proliferative agents targeting cell cycle regulatory proteins.

เลคติน (Lectins) เป็นโปรตีนที่สามารถจับกับหมู่คาร์โบไฮเดรตบนผิวเซลล์ของจุลชีพได้อย่างจำเพาะ และมีบทบาทสำคัญในระบบภูมิคุ้มกันโดยธรรมชาติ เช่น การกระตุ้นการรวมตัวของเชื้อ การจับและกำจัดเชื้อโรคผ่านกลไกของภูมิคุ้มกัน ตลอดจนการยับยั้งการเจริญเติบโตของจุลชีพหลายชนิด เลคตินที่พบในสาหร่ายทะเล โดยเฉพาะสาหร่ายแดง มีความหลากหลายทั้งในด้านโครงสร้างและหน้าที่ และมีรายงานถึงฤทธิ์ทางชีวภาพที่น่าสนใจหลายประการ อย่างไรก็ตาม เลคตินจาก Gracilaria fisheri ยังไม่ได้รับการศึกษาทางหน้าที่อย่างเป็นระบบ งานวิจัยนี้จึงมีวัตถุประสงค์เพื่อผลิตและศึกษาลักษณะการทำงานของเลคตินชนิด L (L-type lectin) จาก Gracilaria fisheri โดยใช้เทคนิครีคอมบิแนนต์โปรตีนใน Escherichia coli และประเมินฤทธิ์ต้านเชื้อแบคทีเรียที่เกี่ยวข้องกับโรคในอุตสาหกรรมเพาะเลี้ยงสัตว์น้ำ สำหรับการพัฒนาเลคตินเป็นสารชีวภาพทางเลือกสำหรับการควบคุมโรคในระบบการเลี้ยงสัตว์น้ำอย่างยั่งยืนในอนาคต

Abstract

Marine algal lectins are promising bioactive compounds with antimicrobial potential. This study produced and characterized a recombinant legume-type lectin from Gracilaria fisheri (rLGFL) and evaluated its antibacterial activity. The LGFL gene was cloned, sequence-verified, and expressed in Escherichia coli. Western blot analysis using a mouse polyclonal antiserum confirmed rLGFL expression. Functional assays demonstrated strong hemagglutination against rabbit erythrocytes, indicating carbohydrate recognition domain (CRD) functionality. rLGFL agglutinated Vibrio parahaemolyticus (3HP), Vibrio harveyi (1114), and Aeromonas veronii while exhibiting bactericidal activity against all tested pathogens, including non-agglutinated V. parahaemolyticus (A3212), Streptococcus agalactiaeAeromonas hydrophila, and Edwardsiella ictaluri. These findings suggest dual antibacterial mechanisms: agglutination-mediated pathogen immobilization and direct bactericidal effects. Seasonal analysis of G. fisheri from Songkhla, Thailand, revealed peak LGFL expression from January to March and July to September, identifying optimal harvesting periods. This study highlights rLGFL as a potential eco-friendly antibacterial agent with applications in aquaculture and disease management.

อุตสาหกรรมการเพาะเลี้ยงกุ้งได้รับผลกระทบผลผลิตต่ำจากการติดเชื้อแบคทีเรียก่อโรคVibrio parahaemolyticus และ Vibrio harveyi  งานวิจัยก่อนหน้าพบสารซัลเฟตโพลิแซคคาไรด์ธรรมชาติที่สกัดจากสาหร่ายผมนางมีฤทธิ์ยับยั้งการเจริญของแบคทีเรียทั้ง 2 ชนิดนี้ได้ ฤทธิ์ทางชีวภาพของสารกลุ่มโพลิแซคคาไรด์มีความสัมพันธ์กับน้ำหนักโมเลกุลและโครงสร้างทางเคมี การศึกษานี้มีวัตถุประสงค์เพื่อปรับโครงสร้างสารซัลเฟตกาแลคแตนสกัดธรรมชาติจากสาหร่ายผมนางโดยการย่อยให้มีขนาดเล็กลง ศึกษาโครงสร้างและทดสอบการเพิ่มศักยภาพในการยับยั้งการเจริญและฆ่าแบคทีเรียของสารที่ได้จากการปรับโครงสร้างเปรียบเทียบกับสารสกัดธรรมชาติ เพื่อการใช้ประโยชน์สูงสุดจากการใช้สารสกัดจากสาหร่ายผมนางผสมเป็นสูตรอาหารกุ้งต่อไป

Abstract

Various seaweed sulfated polysaccharides have been explored for antimicrobial application. This study aimed to evaluate the antibacterial activity of the native Gracilaria fisheri sulfated galactans (NSG) and depolymerized fractions against the marine pathogenic bacteria Vibrioparahaemolyticus and Vibrio harveyi. NSG was hydrolyzed in different concentrations of H2O2 to generate sulfated galactans degraded fractions (SGF). The molecular weight, structural characteristics, and physicochemical parameters of both NSG and SGF were determined. The results revealed that the high molecular weight NSG (228.33 kDa) was significantly degraded to SGFs of 115.76, 3.79, and 3.19 kDa by hydrolysis with 0.4, 2, and 10% H2O2, respectively. The Fourier transformed spectroscopy (FTIR) and 1H- and 13C-Nuclear magnetic resonance (NMR) analyses demonstrated that the polysaccharide chain structure of SGFs was not affected by H2O2 degradation, but alterations were detected at the peak positions of some functional groups. In vitro study showed that SGFs significantly exerted a stronger antibacterial activity against V. parahaemolyticus and V.harveyi than NSG, which might be due to the low molecular weight and higher sulfation properties of SGF. SGF disrupted the bacterial cell membrane, resulting in leakage of intracellular biological components, and subsequently, cell death. Taken together, this study provides a basis for the exploitation and utilization of low-molecular-weight sulfated galactans from G. fisheri to prevent and control the shrimp pathogens.
