ที่มาและความสำคัญ

การสร้างและดัดแปลงอนุภาคโนด้าไวรัส MrNV-VLPs ให้มีความจำเพาะต่อกลุ่มน้ำตาลฟูโคสบนเปลือกหุ้ม คาดว่าจะสามารถยับยั้งการติดเชื้อไวรัสได้อย่างมีประสิทธิภาพ เนื่องโนด้าไวรัส ใช้กลุ่มน้ำตาลฟูโคสในการจับและเข้าสู่เซลล์ นอกจากนี้อนุภาคโนด้าไวรัสยังมีคุณสมบัติเด่นด้านความเสถียร และความแรงของการจับ (avidity) ที่สูง นอกจากนี้ยังสามารถนำมาใช้ประโยชน์ได้ง่าย เนื่องจากสามารถผลิต ปริมาณมาก เก็บรักษา และใช้งานได้ง่ายกว่าเลคทินหรือไกลแคนแบบดั้งเดิม

Abstract

Our previous studies demonstrated that Macrobrachium rosenbergii nodavirus (MrNV) infects and replicates in Sf9 insect cells, where fucosylated N- and O-glycans (HexNAc (Fuc)HexNAc-R or Fuc-LacdiNAc) serve as essential binding molecules for viral attachment. Additionally, we showed that the virus-like particles of the virus itself (MrNV-VLPs) were able to bind to the white-tail disease (WTD)-associated tissues and infect Sf9 cells; and these processes were significantly reduced by removing 27 amino acids from the C-terminus of the protruding (P) domain. In this study, we demonstrated that icosahedral particles could still form following the truncation of the P-domain at sites adjacent to known calcium-binding domains (CBDs) present in the S-shell domain in the MrNV capsid protein. We further showed that icosahedral particles could still form after truncation of the P-domain near calcium-binding sites in the S-domain. This generated smaller but intact particles lacking protrusions, designated V250-MrNV-VLPs. The particles with their intact S-shells enabled our complete replacement of the original MrNV capsid protein P-domains with the fucose-binding carbohydrate-recognition domain (CRD) of CLEC17A lectin (Prolectin), successfully creating CLEC17A/CRD-MrNV-VLPs. The chimeric CLEC17A/CRD-MrNV-VLPs were stable icosahedral particles with P-domains structurally distinct from both the smooth V250-MrNV-VLPs and the blade-like domains of wild-type MrNV-VLPs. Furthermore, the chimeric VLPs presented specific binding activities towards immobilized fucosylated glycoconjugates and Sf9 cell protein lysates. Notably, CLEC17A/CRD-MrNV-VLPs were able to reduce MrNV binding and infection in Sf9 cells. Therefore, this study demonstrated the potential for CLEC17A/CRD-MrNV-VLPs as particles that can effectively bind to fucosylated glycans, and their potential development as nanoparticles that can significantly reduce the level of infection by MrNV in susceptible cells.

ที่มาและความสำคัญ

โรคพยาธิใบไม้ตับ (Fasciola gigantica) เป็นปัญหาสำคัญในสัตว์เคี้ยวเอื้อง โดยเฉพาะในแพะและโค ก่อให้เกิดการสูญเสียทางเศรษฐกิจจากการลดน้ำหนัก โตช้า และลดประสิทธิภาพการผลิต ปัจจุบันการควบคุมโรคยังพึ่งพายาถ่ายพยาธิเป็นหลัก ซึ่งมีความเสี่ยงต่อการดื้อยาเพิ่มขึ้นอย่างต่อเนื่อง จึงมีความจำเป็นเร่งด่วนในการพัฒนาวิธีป้องกันใหม่ เช่น วัคซีน การศึกษานี้จึงศึกษาศักยภาพของโปรตีน cathepsin L1H ในการลดการติดเชื้อพยาธิ และเป็นก้าวสำคัญสู่การพัฒนาวัคซีนที่มีประสิทธิภาพสำหรับควบคุมโรคพยาธิใบไม้ตับในแพะในอนาคต

Abstract

Cathepsin L1H has been identified as a potential vaccine candidate against fasciolosis. The present study was conducted to evaluate the immunoprophylactic efficacy of recombinant proFgCatL1H (rproFgCatL1H) antigen in goats subsequently challenged with the parasite. Immunization of goats with 200 µg of rproFgCatL1H formulated in Montanide™ ISA 61 VG conferred statistically significant reduction in worm recovery compared with both the infected control and adjuvant control groups, corresponding to reductions of 37.55 % and 34.07 %, respectively (p < 0.05). A negative correlation was observed between worm burden reduction and serum IgG levels (OD₄₅₀), measured both at the time of infection and at the study endpoint, although the association was not statistically significant. Scanning Electron Microscopy (SEM) analysis of adult F. gigantica recovered from vaccinated goats demonstrated pronounced tegumental damage, characterized by extensive swelling, surface erosion, and the presence of cell-like structures adhering to the tegumental surface. These findings suggest that the recombinant protein rproFgCatL1H holds promise as a potential vaccine candidate for the control of fasciolosis in goats.

การปฏิสนธิของสัตว์นำ้เศรษฐกิจยังพบว่าอัตราการปฏิสนธิตามธรรมชาติยังมีอัตราการสูญเสียของไข่ที่ไม่ได้รับการผสม ทีมวิจัยจึงศึกษากระบวนการปฏิสนธิในสัตว์น้ำเศรษฐกิจเพื่อเป็นพื้นฐานของการเหนี่ยวนำการปฏิสนธิ โดยศึกษาโปรตีนที่เกี่ยวข้องกับการทำให้เซลล์อสุจิเกิด maturation เพื่อนำไปสู่ fertilization ที่สมบูรณ์

Abstract

Sperm activation occurring in both male and female reproductive tract involves a highly complex series of biomolecular dynamics, particularly on membrane lipids and proteins. In mammals, the universal anticipation in cholesterol (CHO) sequestration plays a role in mammalian sperm maturation/capacitation, subsequently enhancing sperm fertilizing ability. In shrimp, we have previously shown that the level of cholesterol (CHO) is significantly reduced in vas deferens sperm when compared with sperm in the testes, presumably due to the sequestering action of the lipid-binding protein, one of which is Niemann-Pick Type C-2 (NPC2). Here, we used recombinant MrNPC2 protein to treat testicular sperm (Tsp) and quantitatively compared the amount of CHO with non-treated or 2-OH-β-CD treated sperm. HPTLC of the extracted lipids from Macrobrachium rosenbergii sperm revealed the presence of major phospholipids and CHO. Tsp treated with 2-OH-β-CD or MrNPC2 showed lower CHO levels, which was comparable to that of the vas deferens sperm as verified by the Amplex Red assay. Finally, the enhanced levels of protein tyrosine phosphorylation and ionophore-induced AR levels in the MrNPC2 treated Tsp significantly increased in a concentration-dependent manner, similar to that of the Vsp sperm. Altogether, our results indicated the importance of MrNPC2 as CHO moderator in sperm membrane, leading to an enhancement in sperm fertilizing ability.

Abstract

Marine algal lectins are promising bioactive compounds with antimicrobial potential. This study produced and characterized a recombinant legume-type lectin from Gracilaria fisheri (rLGFL) and evaluated its antibacterial activity. The LGFL gene was cloned, sequence-verified, and expressed in Escherichia coli. Western blot analysis using a mouse polyclonal antiserum confirmed rLGFL expression. Functional assays demonstrated strong hemagglutination against rabbit erythrocytes, indicating carbohydrate recognition domain (CRD) functionality. rLGFL agglutinated Vibrio parahaemolyticus (3HP), Vibrio harveyi (1114), and Aeromonas veronii while exhibiting bactericidal activity against all tested pathogens, including non-agglutinated V. parahaemolyticus (A3212), Streptococcus agalactiaeAeromonas hydrophila, and Edwardsiella ictaluri. These findings suggest dual antibacterial mechanisms: agglutination-mediated pathogen immobilization and direct bactericidal effects. Seasonal analysis of G. fisheri from Songkhla, Thailand, revealed peak LGFL expression from January to March and July to September, identifying optimal harvesting periods. This study highlights rLGFL as a potential eco-friendly antibacterial agent with applications in aquaculture and disease management.

Abstract

Antibiotic resistant bacteria are present in various environments and are concerning for public health. Antibiotic resistance genes in soil may proliferate throughout different ecosystems and be acquired by pathogens that pose health risks to humans and animals. This study investigated the antibiotic resistance of cultivable bacteria isolated from livestock-exposed and unexposed areas in a former perlite mining area in the Fa-La-Mee Mountain range, Lopburi, Thailand. Soil samples were collected in five different locations. Soil bacteria were isolated and tested for susceptibility against seven antibiotics, followed by 16S rRNA gene-based identification. A total of 56 bacterial isolates were isolated from the perlite-rich soil samples, including 34 isolates from the livestock-exposed areas and 22 isolates from the unexposed areas. Most of the isolates were resistant to cefepime (26.5% in exposed areas and 22.7% in unexposed areas) and ampicillin (23.5% in exposed areas and 22.7% in unexposed areas). 16S rRNA gene sequences revealed that most of the resistant bacteria isolated from perlite-rich soil were Bacillus spp. A multidrug-resistant isolate of Bacillus cereus resistant to ampicillin, trimethoprim/ sulfamethoxazole, and cefepime was found in a livestock-unexposed area. The majority of the culturable antibiotic-resistant bacteria isolated from perlite-rich soil were cefepime-resistant Bacillus. Interestingly, the pattern of antibiotic resistance in exposed and unexposed areas was not different, which indicated that antibiotic resistance in perlite-rich soil is likely caused by other factors, such as physicochemical characteristics and/or the microbial population of the soil rather than by antibiotics used in livestock husbandry.

การตรวจพบไวรัสชนิดใหม่ Macrobrachium hepatopancreatic bidnavirus (MHBV) ในกุ้งก้ามกราม (Macrobrachium rosenbergii) ค้นพบในปี 2023 การระบาดของไวรัสนี้อาจส่งผลกระทบต่อการเพาะเลี้ยงกุ้งและก่อให้เกิดความสูญเสียอย่างรุนแรง ดังนั้นการพัฒนาวิธีตรวจที่รวดเร็ว แม่นยำ และมีความไวสูงจึงมีความจำเป็นอย่างยิ่ง เพื่อให้สามารถตรวจคัดกรองและควบคุมการแพร่ระบาดของ MHBV ได้อย่างมีประสิทธิภาพในฟาร์มกุ้ง การใช้เทคนิคการตรวจวิเคราะห์ที่แท่นยำเป็นสิ่งที่จำเป็น งานวิจัยพัฒนาเทคนิค qPCR ที่แม่นยำไม่เพียงแต่ช่วยในการป้องกันและจัดการโรคเท่านั้น แต่ยังทำให้เห็นการมีอยู่ของไวรัสนี้ในประเทศไทยมาตั้งแต่ปี 2005 ซึ่งช่วยให้เกิดความเข้าใจในประวัติการแพร่ระบาดและวางแผนการป้องกันโรคในอนาคตได้ดียิ่งขึ้น

Abstract

Macrobrachium hepatopancreatic bidnavirus (MHBV) was first identified in 2023 from the hepatopancreas of moribund river prawns, Macrobrachium rosenbergii (Mr) post-larvae (PL). Developing a rapid and precise quantitative PCR (qPCR) method would enable accurate screening of broodstock river prawns and PL for MHBV. This study developed a specific and sensitive probe-based qPCR assay for the detection of MHBV. Primers and probes were designed from the MHBV genome sequence MN714903 available at GenBank. Two MHBV genes, ORF3 (non-capsid protein) and ORF4 (DNA polymerase domain) were evaluated as target genes. ORF3 exhibited approximately 4-cycles higher sensitivity, thus it was selected for further development. The method developed in this study involved a one tube, duplex qPCR designed to simultaneously target ORF3 from MHBV and an internal housekeeping gene (elongation factor) from prawns. Subsequently, the protocol was validated using templates comprising DNA extracted from Mr, both infected and uninfected with MHBV, and standard DNA plasmids containing the qPCR target sequences. The duplex qPCR method demonstrated high efficiency (92.14 %) with an analytical sensitivity as low as 10 plasmid copies per reaction and a Cq value cut off at 36.40 (± 0.63). Analytical specificity testing was performed using templates extracted from shrimp infected with various viruses (DHPV, XSV, MrNV, WSSV, YHV, PmNV, IHHNV, and IMNV) and all gave negative results. Furthermore, the application of this qPCR method to archived DNA samples extracted from asymptomatic Mr in 2005, 2006, 2010, and 2011 revealed the long presence of MHBV in Thailand before its discovery in 2023 in moribund Mr PL dually infected with MHBV and DHPV. This qPCR method not only provides a valuable tool for the specific detection of MHBV in shrimp but also sheds light on the historical prevalence of MHBV in river prawns.

Abstract

Background: Coffee leaves are a major source of bioactive components and are used as ethnomedicine. However, despite their traditional medicinal use, information about their effects on antihyperlipidemia remains limited. Methods: The aims of this study were to evaluate the main components of leaf extracts from Arabica and Robusta coffees and to examine the potential of these coffee leaves in reducing lipid digestion and absorption in vitro. Results: Coffee leaf extracts from Arabica coffee contain a high amount of caffeine, whereas extracts from Robusta coffee contain high amounts of chlorogenic acid (CGA) and caffeine. Additionally, leaf extracts from Arabica and Robusta coffee demonstrated the inhibition of pancreatic lipase, decreased micellar cholesterol solubility, and reduced bile acid binding. Furthermore, these extracts resulted in a reduction in cholesterol uptake in Caco-2 cells. Molecular docking experiments supported this discovery, showing CGA and caffeine binding to Niemann–Pick C1-like 1 (NPC1L1), a key protein in cholesterol absorption. The results indicated that CGA and caffeine can competitively bind to NPC1L1 at the cholesterol binding pocket, reducing its cholesterol binding rate. These findings suggest that coffee leaves might help suppress lipid absorption and digestion, highlighting their potential use in preventing and treating hyperlipidemia.

Abstract

Bimetallic (Au/Ag) nanoparticles (BNPs) have shown enhanced antibacterial activity compared to their monometallic counterparts. Sulfated galactans (SG) are a naturally occurring polymer commonly found in red seaweed Gracilaria fisheri. They are biocompatible and biodegradable and environmentally friendly. In this study, we utilized SG in combination with BNPs to develop composite materials that potentially enhance antibacterial activity against shrimp pathogens Vibrio parahaemolyticus and Vibrio harveyi, compared to BNPs or SG alone. BNPs were coated with sulfated galactan (SGBNPs) and characterized using UV–vis spectroscopy, Fourier transform infrared (FTIR) spectroscopy, zeta potential, and transmission electron microscopy (TEM). UV–vis spectroscopy analysis revealed that the surface plasmon peaks of BNPs and SGBNPs appeared at 530 nm and 532 nm, respectively. Zeta potential measurements showed that SGBNPs had a negative charge of −32.4 mV, while the BNPs solution had a positive charge of 38.7 mV. TEM images demonstrated the spherical morphology of both BNPs and SGBNPs with narrow size distributions (3–10 nm). Analysis of the FTIR spectra indicated that SG maintained its backbone structure in SGBNPs, but some functional groups were altered. Notably, SGBNPs showed superior antimicrobial and antibiofilm activities against V. parahaemolyticus and V. harveyi compared to SG and BNPs. Furthermore, treatment with SGBNPs significantly down-regulated the expression of virulence-related genes (toxRcpsQ, and mfpA) for V. parahaemolyticus 3HP compared to the respective control, bacteria treated with BNPs or SG. Diets supplemented with SGBNPs, BNPs, or SG showed no detrimental impact on the growth of shrimp Penaeus vannamei. Shrimp fed with SGBNPs-supplemented feed showed significantly higher survival rates than those fed with BNPs-supplemented feed when infected with 3HP after being on the supplemented feed for seven days and a subsequent number of fifteen days. These findings collectively demonstrate the benefit of using SG capped Au–Ag BNPs as an antibacterial agent for the prevention and control of Vibrio sp. Infection in shrimp while reducing the risk of environmental contamination.

We have discovered Macrobrachium hepatopancreatic bidnavirus (MHBV) as the cause of eosinophilic to magenta, intranuclear inclusions in the hepatopancreas of Mr that are sometimes associated with mortality and sometimes not. Decapod hepanhamaparvovirus (DHPV) has shown no recognizable lesions in our samples, unlike its relatives in penaeid shrimp, although grossly normal nuclei were shown to be infected with it by ISH. PCR testing revealed that the two viruses can occur independently in shrimp samples that show no signs of disease. On the other hand, dual infections of MHBV and DHPV exhibited mortality, raising the possibility that shrimp mortality occurs only with dual infections. Based on the similarity of MHBV’s ssDNA genome structure and a DNA polymerase B domain homologous to that of Bombyx mori bidensovirus V1, we believe that MHBV should currently be classified as a new member of the family Bidnaviridae with a genome consisting of a singlessDNA fragment.

Recently, universal PCR detection method with an independent set of Macrobrachium rosenbergii (Mr) samples indicated the presence of a type of decapod hepanhamaparvovirus (DHPV) in the Mr samples. However, the in-situ hybridization (ISH) test results revealed that the hepatopancreatic (HP) cell nuclei of normal morphology gave positive ISH results for DHPV but that the intranuclear inclusions in the same samples did not. The negative ISH results raised the possibility that the intranuclear inclusions arose from something other than a type of DHPV, and possibly a novel virus. Here, we describe the discovery of a novel virus in Mr that is distinct from DHPV and is linked by positive ISH results to the mystery intranuclear inclusions. We call it Macrobrachium hepatopancreatic bidnavirus (MHBV) and we show that it sometimes occurs together with DHPV in dual infections.

Abstract

A purported parvovirus producing circular, eosinophilic inclusions in the nuclei of tubule epithelial cells of the hepatopancreas (HP) of the giant freshwater prawn Macrobrachium rosenbergii (Mr) was first reported from Malaysia in 1990. In 2009, similar inclusions in Mr from Thailand were confirmed to contain single-stranded DNA (ssDNA). In 2021, Mr samples showing similar HP inclusions were reported to give positive PCR test results for decapod hepanhamaparvovirus (DHPV) using universal DHPV primers designed from many viral isolates of 3 penaeid shrimp species. However, the matching DNA probe revealed positive in situ hybridization (ISH) results with host HP nuclei only, and not with the intranuclear inclusions. At the time, this anomaly could not be explained. Here we describe metagenomic analysis and de novo assembly from 2016 samples of Mr with similar intranuclear inclusions that showed negative PCR test results using the DHPV universal primers. Genomes identified through metagenomics revealed a novel ssDNA genome sequence of 6723 bases that was confirmed by a single PCR amplicon. ISH using specific probes derived from this genome sequence revealed that the ssDNA was present in both grossly normal nuclei and in the intranuclear inclusions. Tests with archived DNA from the samples used in the 2021 publication yielded positive PCR results for both DHPV and the novel sequence, suggesting that the shrimp had dual infections. Phylogenetic analysis of the novel sequence revealed no relationship to any known DHPV type. Instead, the sequence closely matched the polymerase B gene in the V1 fragment from Bombyx mori bidensovirus 2 (BmBDV2) that is characterized by 2 ssDNA genome fragments and has been classified in the family Bidnaviridae. The 2 BmBDV2 fragments V1 and V2 each produce their own independent capsid protein genes and virions. In contrast, the new Mr virus is independent with a single 6723 base ssDNA genome, and we propose the name Macrobrachium hepatopancreatic bidnavirus (MHBV).

โรคพยาธิใบไม้ตับเป็นโรคที่เกิดจากการติดเชื้อพยาธิ Fasciola ซึ่งการรักษาโดยยาในปัจจุบันทำให้เกิดผลข้างเคียงและนำไปสู่การดื้อยาที่เพิ่มมากขึ้น การรักษาโดยใช้วัคซีนจึงเป็นแนวทางใหม่ในการป้องกันการติดเชื้อพยาธิใบไม้ตับได้ โดยเลือกศึกษาโดยใช้โปรตีน saposin-2 (SAP) และ leucine aminopeptidase (LAP) เป็นโปรตีนที่พบมากในทางเดินอาหารของพยาธิและมีความสำคัญในการดำรงชีพของพยาธิ

Abstract

Saposin-like protein-2 (SAP-2) and leucine aminopeptidase (LAP) are major proteins involved in the digestive process of Fasciola gigantica (Fg). Both SAP-2 and LAP are highly expressed in F. gigantica; therefore, they could be vaccine candidates for fasciolosis. The aims of this study are (1) to observe the tissue expression of F. gigantica SAP-2 (FgSAP-2) and F. gigantica LAP (FgLAP) in F. gigantica by indirect immunofluorescence technique under confocal microscopy and (2) to test the vaccine potentials of individual and combined recombinant (r) FgSAP-2 and rFgLAP against F. gigantica in Imprinting Control Region (ICR) mice (n = 10 per group). By indirect immunofluorescence-confocal microscopy, FgSAP-2 and FgLAP were localized in the caecal epithelium but at different sites: FgSAP-2 appeared in small granules that are distributed in the middle and lower parts of the cytoplasm of epithelial cells, while FgLAP appeared as a line or zone in the apical cytoplasm of caecal epithelial cells. For vaccine testing, the percent protection of combined rFgSAP-2 and rFgLAP vaccines against F. gigantica was at 80.7 to 81.4% when compared with aluminum hydroxide (alum) adjuvant and unimmunized controls, respectively. The levels of IgG1 and IgG2a in the sera were significantly increased in single and combine vaccinated groups compared with the control groups. Vaccinated mice showed reduced liver damage when compared with control groups. This study indicates that the combined rFgSAP-2 and rFgLAP vaccine had a higher vaccine potential than a single vaccine. These results support the further testing and application of this combined vaccine against F. gigantica infection in farmed livestock animals.
